Ancienne élève de l'École normale supérieure de jeunes filles, élève de Pierre Vilar, agrégée d'histoire, docteur ès lettres, professeur émérite d'histoire contemporaine à l'université Paris VII - Denis Diderot, elle est spécialiste des relations internationales dans la première moitié du XXe siècle. Elle a notamment écrit sur l'histoire politique, économique et sociale de la Troisième République et du régime de Vichy, sur la collaboration pendant la Seconde Guerre mondiale, sur les relations entre le Vatican et l'Allemagne nazie, ainsi que sur la stratégie des élites politiques et économiques françaises avant et après le conflit mondial. Ses publications, tant sur la période 1939-45 que sur le régime stalinien en URSS, ont entraîné diverses polémiques. Plusieurs autres historiens contemporanéistes, ainsi que le site Conspiracy Watch, ont remis en cause la méthodologie historique d'Annie Lacroix-Riz, lui reprochant d'appliquer dans ses ouvrages une grille de lecture qui subordonne la déduction à ses convictions idéologiques, notamment sa croyance en la théorie du complot de la « Synarchie ».
