Western Blot - Tissue or Cell Lysate

Western blot using CPTC-CD40-4 as primary antibody against A549 (lane 2), HeLa (lane 3), Jurkat (lane 4), MCF7 (lane 5), H226 (lane 6), and PBMC (lane 7) whole cell lysates. Expected molecular weight - 30.6 kDa and 22.3 kDa. Molecular weight standards are also included (lane 1).

Automated western blot using CPTC-CD40-4 as primary antibody against A549 (lane 2), HeLa (lane 3), Jurkat (lane 4), MCF7 (lane 5), H226 (lane 6), and PBMC (lane 7) whole cell lysates. Expected molecular weight - 30.6 kDa and 22.3 kDa. Molecular weight standards are also included (lane 1). HeLa is positive. A549, MCF7, and H226 are presumed positive. All other cell lines are negative. Target protein is subject to glycosylation which can affect the migration in electrophoresis. This can make the target appear as a higher molecular weight protein.

Function:
This gene is a member of the TNF-receptor superfamily. The encoded protein is a receptor on antigen-presenting cells of the immune system and is essential for mediating a broad variety of immune and inflammatory responses including T cell-dependent immunoglobulin class switching, memory B cell development, and germinal center formation. AT-hook transcription factor AKNA is reported to coordinately regulate the expression of this receptor and its ligand, which may be important for homotypic cell interactions. Adaptor protein TNFR2 interacts with this receptor and serves as a mediator of the signal transduction. The interaction of this receptor and its ligand is found to be necessary for amyloid-beta-induced microglial activation, and thus is thought to be an early event in Alzheimer disease pathogenesis. Mutations affecting this gene are the cause of autosomal recessive hyper-IgM immunodeficiency type 3 (HIGM3). Multiple alternatively spliced transcript variants of this gene encoding distinct isoforms have been reported. CD40 (CD40 Molecule) is a Protein Coding gene. Diseases associated with CD40 include Immunodeficiency With Hyper-Igm, Type 3 and Immunodeficiency With Hyper-Igm, Type 1.
