Characterization Data [Compare Characterization Data]
Result: Positive
Protein Array in which CPTC-IFIT1-1 is screened against the NCI60 cell line panel for expression. Data is normalized to a mean signal of 1.0 and standard deviation of 0.5. Color conveys over-expression level (green), basal level (blue), under-expression level (red).
Western Blot - Tissue or Cell Lysate
Result: Presumed Positive (with additional bands)
Western blot using CPTC-IFIT1-1 as primary antibody against PBMC (lane 2), HeLa (lane 3), Jurkat (lane 4), A549 (lane 5), MCF7 (lane 6), and NCI-H226 (lane 7) whole cell lysates. Expected molecular weight - 55.4 kDa and 51.7 kDa. Molecular weight standards are also included (lane 1). HeLa is presumed positive. All other cell lines are negative.

Function:
This gene encodes a protein containing tetratricopeptide repeats that was originally identified as induced upon treatment with interferon. The encoded protein may inhibit viral replication and translational initiation. This gene is located in a cluster on chromosome 10 with five other closely related genes. There is a pseudogene for this gene on chromosome 13. Alternatively spliced transcript variants encoding multiple isoforms have been observed.IFIT1 (Interferon Induced Protein With Tetratricopeptide Repeats 1) is a Protein Coding gene. Diseases associated with IFIT1 include Hepatitis C and Hepatitis. Among its related pathways are Interferon gamma signaling and Innate Immune System. Gene Ontology (GO) annotations related to this gene include RNA binding. An important paralog of this gene is IFIT1B.Interferon-induced antiviral RNA-binding protein that specifically binds single-stranded RNA bearing a 5'-triphosphate group (PPP-RNA), thereby acting as a sensor of viral single-stranded RNAs and inhibiting expression of viral messenger RNAs. Single-stranded PPP-RNAs, which lack 2'-O-methylation of the 5' cap and bear a 5'-triphosphate group instead, are specific from viruses, providing a molecular signature to distinguish between self and non-self mRNAs by the host during viral infection. Directly binds PPP-RNA in a non-sequence-specific manner. Viruses evolved several ways to evade this restriction system such as encoding their own 2'-O-methylase for their mRNAs or by stealing host cap containing the 2'-O-methylation (cap snatching mechanism). Exhibits antiviral activity against several viruses including human papilloma and hepatitis C viruses.
