Expansion in apple fruit (Malus domestica Borkh. cv. Braeburn) during early development was induc... more Expansion in apple fruit (Malus domestica Borkh. cv. Braeburn) during early development was induced by injecting 2,4-dichlorophenoxyacetic acid (2,4-D) through the calyx of the fruit and by dipping the apples in N-(2-chloro-4-pyridyl)-N -phenylurea (CPPU). Cell wall composition was analysed, focusing on the hemicellulose fraction containing xyloglucan, a polysaccharide believed to play an important role in cell expansion. Changes were observed in the yields of the cell wall fractions of the fruit treated with either 2,4-D or CPPU, although the monosaccharide composition of the fractions exhibited few differences. There was no decrease in the molecular weight of the xyloglucan from treated fruit. These results are discussed in terms of current cell wall expansion mechanisms.

The new apple (Malus ×domestica Borkh.) cultivar Sciros , resulting from a cross of 'Gala with 'S... more The new apple (Malus ×domestica Borkh.) cultivar Sciros , resulting from a cross of 'Gala with 'Splendour , is marketed internationally from New Zealand. A characteristic of this cultivar is the presence of dense, dark green or black nodules in the fruit cortex, located in close proximity to the five sepal vascular bundles. Nodules are visible as green spots beginning about 40 days after bloom and they continue to develop, reaching a length of up to 8 mm at fruit maturity, although there is considerable variation in their size. Large vascular nodules often develop dark brown centers and reduce the visual quality of the fruit flesh. The frequencies of vascular nodules in 61 'Gala x 'Splendour hybrids from New Zealand and British Columbia, Canada, were examined. These ranged from a mean of 0 to 12.1, depending on the hybrid. Thirteen hybrids were in the high frequency class (2.76-12.1), 28 in the low frequency (0.04-1.86), and 20 were without nodules. The mean nodule frequency in Sciros TM was 12.1 nodules per fruit, the highest of all hybrids examined. Our survey of 44 other cultivars confirmed the occurrence of vascular nodules in 'Gala and 'Splendour , with mean vascular nodule frequency of 1.9 and 0.5 nodules per fruit, respectively. Nodules were also found in 'Newtown Pippin (frequency 0.8), and in a 'Newtown Pippin x 'Granny Smith hybrid (frequency 0.1).

The influence of plant hormones on the replication of white clover mosaic potexvirus (WClMV) in P... more The influence of plant hormones on the replication of white clover mosaic potexvirus (WClMV) in Phaseolus ulgaris L. cv. Top Crop was examined. Plants were wick-fed hormones and the influence on virus replication was assessed using ELISA to measure virus titre. Abscisic acid, indole-3-acetic acid and gibberellic acid did not affect virus replication, while dihydrozeatin, 1aminocyclopropane-1-carboxylic acid (ACC), salicylic acid and jasmonic acid all inhibited virus replication. Hormone treatment did not prevent systemic spread of the virus but virus titre in the systemic leaves was reduced by dihydrozeatin, ACC, salicylic acid and jasmonic acid. Pathogenesisrelated (PR) proteins from groups 2 and 3 were detected in hormone treated plants but only following virus challenge. PR-proteins are unlikely to act directly upon viruses, and the expression of PR-proteins may be the result of viral-induced hormone imbalance. The absence of both induced PR-1 proteins and a hypersensitive response indicated that resistance to WClMV was independent of systemic acquired resistance.

Identifying transformed plant lines carrying the antibiotic resistance marker gene, neomycin phos... more Identifying transformed plant lines carrying the antibiotic resistance marker gene, neomycin phosphotransferase II, requires a more definitive test than the ability of the plant to grow on kanamycin. Although a number of alternative assays have been described, most are cumbersome, time consuming and/ or require the use of radioisotopes. This report describes an ELISA for the detection of the neomycin phosphotransferase II enzyme in transformed plant tissue. The ELISA utilises commercially available antibodies and provides a number of advantages, including an extremely low background, a reduction in the amount of tissue required for testing, and semi-quantitative data on neomycin phosphotransferase II gene expression. This method has been applied successfully to a number of independently transformed lines in nine plant species.

In this work the native promoter of a cytokinin synthase gene (ipt) was removed and replaced with a Cu-controllable promoter. Tobacco (Nicotiana tabacum L. cv tabacum) transformed with this Cu-inducible ipt gene (Cu-ipt) was morphologically identical to controls under noninductive conditions in almost all lines produced. However, three lines grew in an altered state, which is indicative of cytokinin overproduction and was confirmed by a full cytokinin analysis of one of these lines. The in vitro treatment of morphologically normal Cu-ipt transformants with Cu2+ resulted in delayed leaf senescence and an increase in cytokinin concentration in the one line analyzed. The abaxial and adaxial surfaces of Anthurium andraeanum Hort. (anthurium) spathe have marked cut... more The abaxial and adaxial surfaces of Anthurium andraeanum Hort. (anthurium) spathe have marked cuticular thickening. In addition, the spathe tissue has high levels of polyphenolics, evidenced by rapid browning upon injury. Both of these features hinder the extraction of high-quality RNA. To overcome these difficulties, an effective reproducible extraction method was developed using polyvinylpolypyrrolidone (PVPP) and 2-mercaptoethanol. PVPP binds phenolics while 2-mercaptoethanol prevents oxidation of phenolics. Both of these chemicals acting in concert possibly accounted for the marked improvement in the yield and quality of isolated RNA from anthurium spathe. Using this approach we obtained high quality RNA suitable for cDNA library construction, RT-PCR and northern analysis. The method was also effective with other members of the Araceae family.

The influence of seed set on fruit shape was examined in lopsided ´Granny Smith` apple (Malus dom... more The influence of seed set on fruit shape was examined in lopsided ´Granny Smith` apple (Malus domestica Borkh.). Variability in seed weight reflected the dynamics of seed development while asymmetrically distributed seed resulted in asymmetrical fruit. A three-order model was developed which predicted the influence of seed weight on fruit sector weight. The growth of a particular sector was principally influenced by the seeds within its locule (first order), less by the seeds in the two flanking locules (second order) and not by the seeds in the two most distant locules of the fruit (third order). The growth behaviour appears to be modular where each carpel exerts a somewhat autonomous effect on fruit growth and, at least partially, develops as a unit. A weakening seed influence with increasing distance from the sector indicates that seeds principally govern fruit development.

The plant hormone group the cytokinins has been implicated in the delay of senescence, and so the aim of this work was to investigate the link between cytokinin levels in ripening fruit and chlorophyll de-greening. The expression of genes related to cytokinin metabolism and signal transduction and the concentration of cytokinin metabolites were measured. Productivity of T. repens is sometimes threatened by insect pests, and it has been suggested that phenylpropanoid-derived isoflavonoids such as formononetin can protect white clover from insect damage. The aim of this study was to isolate and functionally characterize an isoflavone synthase (IFS2_12) from T. repens by expressing it in Nicotiana tabacum (tobacco), a plant which does not naturally produce isoflavonoids. To induce anthocyanin production and increase isoflavonoid precursors in tobacco, the tomato R2R3 MYB transcription factor ANT1 was expressed in tobacco (Nt-ANT1 plants).
