En este contexto, se han evaluado algunos compuestos antimetanogénicos tales como la lovastatina (LOV), un inhibidor de la hidroximetilglutaril-CoA reductasa, enzima clave para la síntesis de membrana en arqueas metanogénicas. Esta podría ser una opción para mitigar las emisiones de metano ruminal. Previous studies on phytoremediation of soils of those sites reported that this species can remove large amounts of petroleum hydrocarbons (> 300,000 ppm), including polyaromatic (HPA), with a simultaneous support of its rhizosphere. However, the mechanism used by this species for removal of such compounds, especially in the absence of microorganisms, and the effect of oil on the germination and its plant physiology is unknown. Lovastatin (Lv) addition to feedstocks could be a strategy to mitigate rumen methane emissions via decreasing the population of methanogenic archaea (MA). Thus, this paper provides the first overview of the effects of Lv supplementation, focusing on the inhibition of methane production, rumen microbiota, and ruminal fermentation. Results indicated that Lv treatment had a strong anti-methanogenic effect on pure strains of MA. However, there are uncertainties from in vitro rumen fermentation trials with complex substrates and rumen inoculum.Solid-state fermentation (SSF) has emerged as a cost-effective option to produce Lv. Several nitrogen fixing consortia (NFC) were isolated from kerosene contaminated soil, where Phas... more Several nitrogen fixing consortia (NFC) were isolated from kerosene contaminated soil, where Phaseolus vulgaris plants were being cultivated. The capability of these consortia for kerosene hydrocarbons removal was investigated and demonstrated. The NFC cultivated under aerobic conditions, and kerosene as sole carbon source, effected a maximum of 75% of reduction of the total kerosene hydrocarbons. Also, from experiments conducted to evaluate their atmospheric nitrogen fixing capability, all consortia showed nitrogenase activity: from 4 to 183 nmol N2/3E09 bacteria.day. To our knowledge, this is the first report that shows a group of bacteria with the dual characteristic of fixing atmospheric nitrogen and capability to use kerosene hydrocarbons as a sole carbon source. Application of these microorganisms to soil bioremediation processes is discussed.

In this work, hairy root cultures of Brassica oleracea var. italica (broccoli) were established in an airlift with mesh bioreactor to produce isoform 1 of the human growth hormone (hGH1) as a model therapeutic protein. The hGH1 cDNA was cloned into the pCAMBIA1105.1 binary vector to induce hairy roots in hypocotyls of broccoli plantlets via Agrobacterium rhizogenes. Most of the infected plantlets (90%) developed hairy roots when inoculated before the appearance of true leaves, and keeping the emerging roots attached to hypocotyl explants during transfer to solid Schenk and Hildebrandt medium. The incorporation of the cDNA into the hairy root genome was confirmed by PCR amplification from genomic DNA. The expression and structure of the transgenic hGH1 was assessed by ELISA, western blot, and MALDITOF-MS analysis of the purified protein extracted from the biomass of hairy roots cultivated in bioreactor for 24 days. Production of hGH1 was 5.1 6 0.42 mg/g dry weight (DW) for flask cultures, and 7.8 6 0.3 mg/g DW for bioreactor, with productivity of 0.68 6 0.05 and 1.5 6 0.06 mg/g DW*days, respectively, indicating that the production of hGH1 was not affected by the growth rate, but might be affected by the culture system. These results demonstrate that hairy root cultures of broccoli have potential as an alternative expression system for production of hGH1, and might also be useful for production of other therapeutic proteins. Encystment of Azotobacter nigricans was induced by its diazotrophic cultivation on kerosene. Its ... more Encystment of Azotobacter nigricans was induced by its diazotrophic cultivation on kerosene. Its growth and nitrogenase activity were affected by kerosene in comparison to cultures grown on sucrose. Electron microscopy of vegetative cells showed that when nitrogenase activity was higher and the poly-beta-hydroxybutyrate granules were not present to a significant extent, peripheral bodies were abundant. After 8 days of culture on kerosene, the presence of cysts with intracellular bunches of poly-beta-hydroxybutyrate granules was observed. Germination of cysts bears germinating multicelled yet unbroken capsule cysts with up to three cells inside. This is the first report of encystment induction of Azotobacter species grown on kerosene.

In this work, hairy root cultures of Brassica oleracea var. italica (broccoli) were established in an airlift with mesh bioreactor to produce isoform 1 of the human growth hormone (hGH1) as a model therapeutic protein. The hGH1 cDNA was cloned into the pCAMBIA1105.1 binary vector to induce hairy roots in hypocotyls of broccoli plantlets via Agrobacterium rhizogenes. Most of the infected plantlets (90%) developed hairy roots when inoculated before the appearance of true leaves, and keeping the emerging roots attached to hypocotyl explants during transfer to solid Schenk and Hildebrandt medium. The incorporation of the cDNA into the hairy root genome was confirmed by PCR amplification from genomic DNA. The expression and structure of the transgenic hGH1 was assessed by ELISA, western blot, and MALDITOF-MS analysis of the purified protein extracted from the biomass of hairy roots cultivated in bioreactor for 24 days. Production of hGH1 was 5.1 6 0.42 mg/g dry weight (DW) for flask cultures, and 7.8 6 0.3 mg/g DW for bioreactor, with productivity of 0.68 6 0.05 and 1.5 6 0.06 mg/g DW*days, respectively, indicating that the production of hGH1 was not affected by the growth rate, but might be affected by the culture system. These results demonstrate that hairy root cultures of broccoli have potential as an alternative expression system for production of hGH1, and might also be useful for production of other therapeutic proteins. Its growth and nitrogenase activity were affected by kerosene in comparison to cultures grown on sucrose. Electron microscopy of vegetative cells showed that when nitrogenase activity was higher and the poly-beta-hydroxybutyrate granules were not present to a significant extent, peripheral bodies were abundant. After 8 days of culture on kerosene, the presence of cysts with intracellular bunches of poly-beta-hydroxybutyrate granules was observed. Germination of cysts bears germinating multicelled yet unbroken capsule cysts with up to three cells inside. This is the first report of encystment induction of Azotobacter species grown on kerosene.
