A series of esters derived from 3,7-dimethyl-3,7-diazabicyclo[3.3.1]nonan-9-ol has been synthesiz... more A series of esters derived from 3,7-dimethyl-3,7-diazabicyclo[3.3.1]nonan-9-ol has been synthesized and studied using 1H and 13C NMR spectroscopy and the crystal structure of 9-(3-indolycarbonyloxy)-3,7-dimethyl-3,7-diazabicyclo[3.3.1]nonane hydrochloride (V·HCl) has been determined by X-ray diffraction. ABSTRACT The di-μ-hydroxo-complex [NBu4]2[Ni(C6F5)2(μ-OH)2] reacts with protic electrophiles H(LL... more ABSTRACT The di-μ-hydroxo-complex [NBu4]2[Ni(C6F5)2(μ-OH)2] reacts with protic electrophiles H(LL) in the 1/2 molar ratio to give (NBu4) [Ni(C6F5)2(LL)][(LL) = acetylacetonate (acac) (I), benzoylacetonate (bzac) (II), 8-hydroxyquinolinate (oxin) (III), pyridine-2-thiolate (pyt) (IV), pyrimidine-2-thiolate (pymt) (V), 2,6-dimethyl-5-oxo-1,2,4-triazine-3-thiolate (tazt) (VI), N-methylimidazole-2-thiolate (mimt) (VII), thiazolidine-2-thiolate (tzt) (VIII), benzoimidazole-2-thiolate (bimt) (IX) or benzothiazole-2-thiolate (btzt) (X)]. The new compounds have been characterized by analysis, conductivity measurements, and spectroscopic (IR, UV-VIS, 1H, and 19F) methods. The crystal structure of compound IV has been determined by X-ray diffraction. It crystallizes in the monoclinic space group P21/n with a 11.7076(4), b 22.183(1), c 14.0280(6) Å, β 105.012(3)°. Final R = 0.075 and Rw = 0.067 based on 3815 reflections.

We have investigated activation of two isoenzymes (lip1 and lip3) from Candida rugosa in polyethy... more We have investigated activation of two isoenzymes (lip1 and lip3) from Candida rugosa in polyethylene glycol (PEG) media. Aqueous solutions of PEG 8000 and 20,000 activate lip3 but not lip1 from C. rugosa. Maximum activation (260%) of lip3 requires 6 h of pre-incubation with PEG 8000 (4%, w/v). PEG seems to shift the equilibrium between the open and the closed forms of lip3 towards the active conformation. Inhibition experiments demonstrate that ligands have easier access to the lip3 active site than to the lip1 active site, both in the presence and the absence of PEG. The presence of PEG in the crystallization medium is responsible for reported differences in the crystal structures of lip1 and lip3. A comparative analysis of crystallographic models of lip1 and lip3 suggests a role for PEG in activation of lip3 and further stabilization of the activated/open form via dimerization in aqueous media.

Reactions of N,N-diisopropylisonicotinamide (L) with anhydrous CuCl2, CuBr2 and Cu(ClO4)2·6H2O yi... more Reactions of N,N-diisopropylisonicotinamide (L) with anhydrous CuCl2, CuBr2 and Cu(ClO4)2·6H2O yielded: (a) an ionic product with the molecular composition of [CuL2(H2O)4]·2[CuLCl3] 1; (b) a coordination polymer [CuL2Br2]n2 and (c) a mononuclear complex [CuL4(C3H8O)2]·(ClO4)2] 3, respectively. Separation of daughter cells during bacterial cell division requires splitting of the septal cros... more Separation of daughter cells during bacterial cell division requires splitting of the septal cross wall by peptidoglycan hydrolases. In Streptococcus pneumoniae, PcsB is predicted to perform this operation. Recent evidence shows that PcsB is recruited to the septum by the transmembrane FtsEX complex, and that this complex is required for cell division. However, PcsB lacks detectable catalytic activity in vitro, and while it has been proposed that FtsEX activates PcsB, evidence for this is lacking. Here we demonstrate that PcsB has muralytic activity, and report the crystal structure of full-length PcsB. The protein adopts a dimeric structure in which the V-shaped coiled-coil domain of each monomer acts as a pair of molecular tweezers locking the catalytic domain of each dimeric partner in an inactive configuration. This suggests that the release of the catalytic domains likely requires an ATP-driven conformational change in the FtsEX complex, conveyed towards the catalytic domains through coordinated movements of the coiled-coil domain.

In a similar manner, the reaction of phthalimidoacetyl chloride with these imines in the presence of triethylamine afforded the corresponding 3-phthalimido β-lactams as single diastereomers

MoO 2 Cl 2 L 2 (L 0/dimethyl sulfoxide (Me 2 SO), deuterated dimethyl sulfoxide (Me 2 SOd 6), din... more MoO 2 Cl 2 L 2 (L 0/dimethyl sulfoxide (Me 2 SO), deuterated dimethyl sulfoxide (Me 2 SOd 6), din -buthyl sulfoxide (Bu 2 SO), tetramethylene sulfoxide (C 4 H 8 SO), dibenzyl sulfoxide (Bz 2 SO), diphenyl sulfoxide (Ph 2 SO), dip -tolyl sulfoxide (p-Tol 2 SO), di-pchloro-phenyl sulfoxide (p-Cl-Ph 2 SO) and methyl phenyl sulfoxide (MePhSO)) have been prepared by reacting a solution of MoO 2 Cl 2 (H 2 O) 2 in diethyl ether with the respective ligand. The molecular structure of MoO 2 Cl 2 (Me 2 SO) 2 has been established by X-ray diffraction analysis. The ability of representative complexes to work as oxotransfer catalysts has been tested.
