𝗜𝘀 𝗶𝘁 𝘁𝗿𝘂𝗹𝘆 𝘁𝗵𝗲 𝘁𝗶𝗺𝗲 𝘁𝗼 𝘀𝗲𝗾𝘂𝗲𝗻𝗰𝗲 𝘆𝗼𝘂𝗿 𝘄𝗵𝗼𝗹𝗲 𝗴𝗲𝗻𝗼𝗺𝗲? 🧬 As genome sequencing costs have plummeted from billions to just a thousand dollars, it's becoming more accessible. But is it practical for the average person? Unfortunately, working with whole genome data still requires specialized knowledge. Reading and interpreting this data is no small task. Bioinformaticians like me often filter out hundreds of genetic mutations that, based on predictions, might seem harmful or crucial for gene function. However, many of these mutations have no real effect on the individual. This makes interpreting the data challenging, as what appears harmful in the genome might not actually be so. So, how do bioinformaticians and geneticists differentiate between significant and insignificant mutations? Here’s an oversimplified overview of how we do it: 𝟭. 𝗟𝗼𝗼𝗸 𝗮𝘁 𝗵𝗼𝘄 𝗽𝗿𝗲𝘃𝗮𝗹𝗲𝗻𝘁 𝘁𝗵𝗲 𝗺𝘂𝘁𝗮𝘁𝗶𝗼𝗻 𝗶𝘀 𝗶𝗻 𝘁𝗵𝗲 𝗽𝗼𝗽𝘂𝗹𝗮𝘁𝗶𝗼𝗻. Mutations that are predicted to have a high impact but are common in the population are less likely to be harmful. If many people have the mutation, any effect it has may be too widespread to be considered pathogenic. 𝟮. 𝗟𝗼𝗼𝗸 𝗮𝘁 𝗵𝗼𝘄 𝘄𝗲𝗹𝗹 𝘁𝗵𝗲 𝗺𝘂𝘁𝗮𝘁𝗶𝗼𝗻 𝗶𝘀 𝗰𝗼𝗻𝘀𝗲𝗿𝘃𝗲𝗱 𝗮𝗰𝗿𝗼𝘀𝘀 𝘃𝗮𝗿𝗶𝗼𝘂𝘀 𝗮𝗻𝗶𝗺𝗮𝗹 𝘀𝗽𝗲𝗰𝗶𝗲𝘀. Genes that are conserved across closely related species are likely to be important. If a mutation affects a well-conserved gene, it might be more significant. 𝟯. 𝗟𝗼𝗼𝗸 𝗶𝗳 𝘁𝗵𝗲 𝗺𝘂𝘁𝗮𝘁𝗶𝗼𝗻 𝗶𝘀 𝗽𝗿𝗲𝘀𝗲𝗻𝘁 𝗶𝗻 𝗿𝗲𝗹𝗮𝘁𝗶𝘃𝗲𝘀. If a mutation is found in family members who also show related symptoms or effects, it’s more likely to be important. Many companies offering whole genome sequencing struggle with the challenge of making this complex data understandable for the average person. It has a huge potential to make it understandable for people, and that’s why at Macromo we’re now moving further into the era of whole genome sequencing (WGS) with a new test specifically focused on the following reports, for which we have researched the most up-to-date evidence available. 𝗟𝗼𝗻𝗴𝗲𝘃𝗶𝘁𝘆 𝗮𝗻𝗱 𝗣𝗿𝗼𝘁𝗲𝗰𝘁𝗶𝘃𝗲 𝗠𝘂𝘁𝗮𝘁𝗶𝗼𝗻𝘀 - For example, beneficial mutations in the APOE gene that lower cholesterol levels, decrease Alzheimer’s risk, or mutations in a gene that cause immunity to HIV infection. 𝗠𝗲𝘁𝗮𝗯𝗼𝗹𝗶𝗰 𝗛𝗲𝗮𝗹𝘁𝗵 - Changes such as in the PCSK9 gene, which can affect blood lipid levels and thus cardiovascular risks. 𝗠𝗲𝘁𝗵𝘆𝗹𝗮𝘁𝗶𝗼𝗻 - The much discussed MTHFR, MTR, and MTRR gene mutations that are linked with higher needs of vitamin B9 and B12 intake to lower homocysteine levels and thus improve overall health. 𝗜𝗻𝗳𝗹𝗮𝗺𝗺𝗮𝘁𝗶𝗼𝗻 - Certain genetic changes in IL-6, IL-1A, IL-1B, and other interleukin group genes can cause a predisposition to higher levels of systemic inflammation. Our customer support team at Macromo tells me that many people are asking about the MTHFR gene, methylation, and homocysteine. We're curious about why this topic is suddenly so popular. The gene is important and interesting to study, but most people don't look beyond the hype surrounding it. 𝗪𝗵𝗮𝘁 𝗶𝘀 𝗺𝗲𝘁𝗵𝘆𝗹𝗮𝘁𝗶𝗼𝗻 𝗮𝗻𝗱 𝗵𝗼𝘄 𝗱𝗼𝗲𝘀 𝗶𝘁 𝗮𝗳𝗳𝗲𝗰𝘁 𝗵𝗼𝗺𝗼𝗰𝘆𝘀𝘁𝗲𝗶𝗻𝗲? People talk about methylation, both positively and negatively, and often mention genes like MTHFR, MTR, and MTRR. Methylation is a chemical process that happens in your body all the time. It adds small "tags" (called methyl groups) to molecules, helping your body control genes and do other important things. The MTHFR gene provides instructions for making an enzyme that changes vitamin B9 into a form needed for methylation. If this gene has mutations, the enzyme may not work as well, leading to fewer methyl groups. Having fewer of those available means that the body cannot convert homocysteine, a naturally occurring amino acid, into its less harmful form. High levels of homocysteine in the blood are linked to heart disease. 𝗣𝗲𝗼𝗽𝗹𝗲 𝗮𝗿𝗲 𝗶𝗻𝘁𝗲𝗿𝗲𝘀𝘁𝗲𝗱 𝗷𝘂𝘀𝘁 𝗶𝗻 𝘁𝗵𝗲 𝘀𝗽𝗲𝗰𝗶𝗳𝗶𝗰 𝗴𝗲𝗻𝗲𝘀, 𝗯𝘂𝘁 𝗱𝗼𝗻'𝘁 𝘀𝗲𝗲 𝗮 𝗱𝗶𝗳𝗳𝗲𝗿𝗲𝗻𝗰𝗲 𝗯𝗲𝘁𝘄𝗲𝗲𝗻 𝗮 𝗴𝗲𝗻𝗲 𝗮𝗻𝗱 𝗶𝘁𝘀 𝘃𝗮𝗿𝗶𝗮𝗻𝘁. There is a tendency for people to focus on specific genes without distinguishing between the gene itself and its particular variants. People often reach out, wanting us to “measure a specific gene”. The longevity community often highlights multiple genes as influential, but it is essential to recognize that we all possess these genes. Actually, if someone didn’t have some of the genes that the longevity community labels as “bad”, their absence would likely result in serious inborn disease. One example could be the APOE gene, specifically with its APOE4 variant. Unfortunately, the changes in the gene that result in the APOE4 variant do increase various health risks, especially Alzheimer’s, but if someone didn't have the APOE gene at all, it would lead to very serious health problems. This shows that while certain variants of a gene might be linked to health risks, thus people label it as a "bad gene", the gene itself is essential for overall health. 𝗠𝗮𝗰𝗿𝗼𝗺𝗼 𝗶𝘀 𝗺𝗼𝘃𝗶𝗻𝗴 𝗶𝗻 𝘁𝗼 𝗰𝗹𝗮𝗿𝗶𝗳𝘆 𝘁𝗵𝗶𝘀 𝘁𝗼𝗽𝗶𝗰 At Macromo, we're about to launch a new product with more clear, evidence-based reports.
