Twenty-four pregnant rats were used in the current study. They were randomly allocated into four equal groups (n=6): control, garlic, and CP + Garlic group. At the end of the weaning period, the mothers and the offsprings of all groups were sacrificed, the kidneys were immediately excised, and processed for histological and biochemical investigations. Also, blood samples were withdrawn and processed for estimation of the assigned biochemical parameters. The renal histological sections from CP-treated mother rats displayed pronounced histopathological lesions however; their offsprings showed mild renal histopatological lesions if compared with those of their mothers. Treatment with cyclophosphamide (CTX); a widely used anticancer drug, causes leukopenia which is ... more Treatment with cyclophosphamide (CTX); a widely used anticancer drug, causes leukopenia which is the most critical associated side effect. The present work aimed to investigate the effects of treatment with bone marrow products on leukopenia and liver injury induced by CTX in Ehrlich ascetic carcinoma (EAC) mouse model. Adult female CD-1 mice were challenged with Ehrlich ascetic carcinoma (EAC) (2.5 x 105) cells on day 0 and then were inoculated 24 hours later with 100uL PBS or CTX (200 mg/kg) via intrapertioneal injection. The next day after CTX treatment, mice were treated for 5 consecutive days with granulocytes-colony stimulating factor (G-CSF; 250 µg/kg) and/or bone marrow lysate (100 µg/mouse) prepared from naive or CTX-treated mice via subcutaneous injection. Another groups of tumour-bearing mice were transferred with viable bone marrow cells (5 x 106) harvested from naive or CTX-treated mice. The results indicated that treatment with G-CSF induced recovery of the CTX-induced decreases in the numbers of bone marrow cells, splenocytes and peripheral blood leukocytes. While transfer of viable bone marrow cells resulted in recovery of bone marrow cell and spleen cell count only. Treatment of CTX-treated mice with bone marrow lysate prepared from naive or CTX-treated mice also induced recovery of the absolute number of lymphocytes. These effects were also associated with recovery from CTX-induced liver injury as observed by histopathological investigation. Of notes, treatment with bone marrow lysate prepared from naive or CTX-treated mice showed the highest recovery effects against CTX-induced lymphopenia. Finally, it is concluded that treatment with bone marrow products accelerates recovery of chemotherapy associated toxicity, opening a new avenue for further investigation to optimize their effects.

Also, the effects of honey bee and propolis extract to modulate the role of these cytokines. Methods: Fifty female Wister rats were divided into five groups, 10 per each group. After 6 w of all treatments, rats were sacrificed, whole blood and serum were collected to determine the number of white blood cells (WBCs), percentage of granulocytes, percentage of lymphocytes and the level of some cytokines such as gamma interferon(INF-γ), tumor necrosis factor(TNF-α) and interleukin-2(IL-2). Results: CCl4 administration resulted in significant increase in the number of WBCs and percentage of granulocytes (P<0.05 and P<0.01 respectively). The present study was conducted to demonstrate the histological effects of ribavirin and/or sofos... more The present study was conducted to demonstrate the histological effects of ribavirin and/or sofosbuvir on liver and kidney of rats. Rats were treated with ribavirin (0.0227 mg/kg body weight), sofosbuvir (0.01 mg/kg body weight) and mixture of ribavirin and sofosbuvir for 12 weeks. Liver and kidney were stained with H/E stain. Liver tissue also stained with AgNORs stain. The results showed that no histological alteration or abnormal counts were observed after treatment with ribavirin and / or sofosbuvir. Also the count of AgNORs is non-significantly differs between the normal non-treated rats and those treated with ribavirin and/or sofosbuvir. The present study concluded that, treatment with ribavirin and / or sofosbuvir or both has no side effects on normal rats.

The effects of artesunate and myrrh on S. mansoni infection and the levels of some Th1 and Th2 cy... more The effects of artesunate and myrrh on S. mansoni infection and the levels of some Th1 and Th2 cytokines were evaluated in the present study. Six weeks after infection, a group of mice was treated with 4 mg/kg of artesunate and other group was treated with 10 mg/kg of myrrh for 3 successive days. Worm burden was reduced with a percentage of 53.7 % and 58.78 % after treatment with myrrh and artesunate respectively as well as the levels of igG antibodies were significantly reduced compared with infected group. No obvious changes were observed in the level of interferon γ after treatment. after treatment with artesunate, interleukin 2 (il-2) level was significantly decreased, while no significant difference was observed in myrrh-treated group compared with the infected group. on the other hand, the level of il-10 was not significantly decreased after treatment with artesunate, but it was significantly increased after treatment with myrrh. however, il-12 levels were significantly decreased after treatment with artesunate. The results demonstrated that, artesunate or myrrh treatment could give a level of protection against S. mansoni infection and modulate the levels of some Th1 and Th2 cytokines in mice infected with S. mansoni.

The 74 kDa target antigen was isolated from antigenic extracts of S. mansoni adult worms. Swiss and C57 BL/6J mice were immunized with 30, 50, 100 and 200 mg antigen/mouse doses with and without Freund's adjuvant. Sera of immunized mice showed high reactivity against 74 kDa antigen. The highest protection level (76.6% in Swiss mice and 50.1% in C57 BL/6J mice) was obtained using the 50 mg antigen dose with and without Freund's adjuvant. A marked reduction in granuloma number and intensity of collagen and reticular granuloma ®bers was observed. The 74 kDa antigen has the ability to protect mice of dierent strains and to modulate the host immune system.

Objectives: Argyrophilic nucleolar organizer regions (AgNOR) proteins are a set of argyrophilic n... more Objectives: Argyrophilic nucleolar organizer regions (AgNOR) proteins are a set of argyrophilic nucleolar proteins that accumulate in highly proliferating cells, whereas their expression is very low in nonproliferating cells. The present study aimed to investigate the potential of DNA flow cytometry (FCM) and AgNORs count in the assessment of cellular kinetics of liver cirrhosis and hepatocellular carcinoma. Design and methods: Small-needle liver biopsies (217) were included and were taken from 84 patients with hepatocellular carcinoma (HCC) (one biopsy from tumor lesion and the other from residual nontumor) liver tissues. Only one biopsy was taken from 49 patients with liver cirrhosis. One part of biopsy was subjected to flow cytometry, and the other, to histopathology and AgNORs counting. Results: An aneuploidy was shown in 44.5% of liver cirrhosis and in 78.6% of tumor sites. Aneuploid HCC cases showed high AgNORs count compared with diploid cases (3.407 ± 1.18 vs. 1.74 ± 0.9). An extremely significant increase in AgNORs count in tumor lesion (P b 0.001) was found compared with residual liver tissues, liver cirrhosis and normal liver (3.89 ± 0.827, 1.49 ± 0.52, 1.62 ± 0.29, and 1.3 ± 0.17, respectively). In liver cirrhosis, dysplasia showed a significant relationship with ploidy (P b 0.001) and AgNORs count (P b 0.05). Conclusion: AgNORs count and DNA ploidy analysis of core biopsy specimens are useful in the assessment of cellular kinetics of liver cirrhosis and hepatocellular carcinoma.

Day length and temperature are the two main factors that have been proved to cause changes in different hormones. The current study aimed to compare seasonal changes of some hormones such as testosterone (T), estradiol-17-β (E 2) and thyroid hormones (T 4 , thyroxine; and T 3, triiodothyronine) in selected vertebrates (amphibians, fish and mammals). These hormones were assayed by enzyme-linked immunosorbent assay (ELISA). The present results indicated that; T 3 had the highest concentration in both sexes of amphibians during winter and the lowest one was found in the investigated mammals of both sexes during summer and autumn. Regarding, T 4 also showed the highest concentration in both sexes of amphibians during winter and the lowest one was recorded in male mammals during summer. In relation to E 2 , in females; the highest and detectable concentration was observed in fishes during summer and the lowest one was found in mammals during winter. On the other hand, in males, its highest concentration was found in birds during spring, and the lowest one was recorded in amphibians during winter. Testosterone showed the highest concentration in female birds during summer, while the lowest one was recorded in amphibians during summer. In males, the highest T level was observed in fishes during summer, while the lowest one was found in amphibians in the same season. According to the results, significant changes in levels of steroid hormones, as well as thyroids in the selected animals from different classes were closely correlated to the length of the day and seasons.

A 30 kDa antigen was characterized as a hydrophobic polypeptide containing 16 amino acids and eva... more A 30 kDa antigen was characterized as a hydrophobic polypeptide containing 16 amino acids and evaluated as a potential candidate vaccine against infection by Schistosoma mansoni. CD1 albino mice immunized at 0, 14, and 21 days with 25 or 50 microg of the 30 kDa antigen per mouse with and without alum developed high levels of IgG antibodies (predominantly IgG2a and IgG2b isotypes). When immunized mice were infected with 200 S. mansoni cercariae, the highest protection levels (61% and 65% reduction in worm burden in two separate experiments) were obtained using the 50-microg antigen without alum adjuvant. The granuloma size decreased to 10%, a non-significant level in mice immunized using alum adjuvant. The results demonstrate the ability of the 30 kDa antigen with and without alum adjuvant to protect mice against S. mansoni infection.

Objectives: Argyrophilic nucleolar organizer regions (AgNOR) proteins are a set of argyrophilic n... more Objectives: Argyrophilic nucleolar organizer regions (AgNOR) proteins are a set of argyrophilic nucleolar proteins that accumulate in highly proliferating cells, whereas their expression is very low in nonproliferating cells. The present study aimed to investigate the potential of DNA flow cytometry (FCM) and AgNORs count in the assessment of cellular kinetics of liver cirrhosis and hepatocellular carcinoma. Design and methods: Small-needle liver biopsies (217) were included and were taken from 84 patients with hepatocellular carcinoma (HCC) (one biopsy from tumor lesion and the other from residual nontumor) liver tissues. Only one biopsy was taken from 49 patients with liver cirrhosis. One part of biopsy was subjected to flow cytometry, and the other, to histopathology and AgNORs counting. Results: An aneuploidy was shown in 44.5% of liver cirrhosis and in 78.6% of tumor sites. Aneuploid HCC cases showed high AgNORs count compared with diploid cases (3.407 ± 1.18 vs. 1.74 ± 0.9). An extremely significant increase in AgNORs count in tumor lesion (P b 0.001) was found compared with residual liver tissues, liver cirrhosis and normal liver (3.89 ± 0.827, 1.49 ± 0.52, 1.62 ± 0.29, and 1.3 ± 0.17, respectively). In liver cirrhosis, dysplasia showed a significant relationship with ploidy (P b 0.001) and AgNORs count (P b 0.05). Conclusion: AgNORs count and DNA ploidy analysis of core biopsy specimens are useful in the assessment of cellular kinetics of liver cirrhosis and hepatocellular carcinoma.

Day length and temperature are the two main factors that have been proved to cause changes in different hormones. The current study aimed to compare seasonal changes of some hormones such as testosterone (T), estradiol-17-β (E 2) and thyroid hormones (T 4 , thyroxine; and T 3, triiodothyronine) in selected vertebrates (amphibians, fish and mammals). These hormones were assayed by enzyme-linked immunosorbent assay (ELISA). The present results indicated that; T 3 had the highest concentration in both sexes of amphibians during winter and the lowest one was found in the investigated mammals of both sexes during summer and autumn. Regarding, T 4 also showed the highest concentration in both sexes of amphibians during winter and the lowest one was recorded in male mammals during summer. In relation to E 2 , in females; the highest and detectable concentration was observed in fishes during summer and the lowest one was found in mammals during winter. On the other hand, in males, its highest concentration was found in birds during spring, and the lowest one was recorded in amphibians during winter. Testosterone showed the highest concentration in female birds during summer, while the lowest one was recorded in amphibians during summer. In males, the highest T level was observed in fishes during summer, while the lowest one was found in amphibians in the same season. According to the results, significant changes in levels of steroid hormones, as well as thyroids in the selected animals from different classes were closely correlated to the length of the day and seasons.

A 30 kDa antigen was characterized as a hydrophobic polypeptide containing 16 amino acids and eva... more A 30 kDa antigen was characterized as a hydrophobic polypeptide containing 16 amino acids and evaluated as a potential candidate vaccine against infection by Schistosoma mansoni. CD1 albino mice immunized at 0, 14, and 21 days with 25 or 50 microg of the 30 kDa antigen per mouse with and without alum developed high levels of IgG antibodies (predominantly IgG2a and IgG2b isotypes). When immunized mice were infected with 200 S. mansoni cercariae, the highest protection levels (61% and 65% reduction in worm burden in two separate experiments) were obtained using the 50-microg antigen without alum adjuvant. The granuloma size decreased to 10%, a non-significant level in mice immunized using alum adjuvant. The results demonstrate the ability of the 30 kDa antigen with and without alum adjuvant to protect mice against S. mansoni infection.
