Figure S2 Breast cancer cell lines respond to Sema4D. (A) Analyses of plexin B1 receptor in TSA and MLP29 cell lines. Cells were lysed with EB buffer, as previously described (Giordano, S., S. Corso, P. Conrotto, S. Artigiani, G. Gilestro, D. Barberis, L. Tamagnone, and P.M. Comoglio. 2002. Nat. Cell Biol. 4:720–724), and immunoprecipitated with anti–plexin B1 antibodies. Proteins were then run on an 8% SDS-PAGE. MLP29 cells were used as positive control. IP, immunoprecipitation; WB, Western blot. (B and C) Transwell motility and invasion assays. The ability of the indicated breast cancer cell lines to migrate or invade Matrigel-coated Transwells in response to different chemoattractants (HGF and Sema4D) was evaluated. The graphs indicate the number of migrated cells. Error bars represent mean ± SD (n = 6). Nt, not treated.
