The study of the mechanism of antiviral action and evaluation of the clinical efficacy, safety and tolerability of therapy with Fortepren® in patients with chronic recurrent herpesvirus infection of genital localization (CRHVI). Materials and methods. Clinical studies were carried out of a drug Fortepren® (0.4% sodium polyprenyl phosphate solution), which was administered to patients who underwent a basic therapeutic course of the drug Acyclovir-Acry® to relieve the acute phase of the disease. The study was performed on 80 male and female patients selected during the screening with a confirmed diagnosis of CRHVI. Two groups were formed. Patients of group 1 (experimental) were intramuscularly injected with Fortepren® at a dose of 2 ml (8 mg) three times at intervals of 21 days by 3 ± 2, 24 ± 2 and 45 ± 2 days following the 10-day basic course of treatment of the acute phase of diseases with the use of the acyclovir tablets of 400 mg - 13 ± 2, 34 ± 2 and 55 ± 2 days from the begi...

Immunogenic properties of cytolysin were studied in experiments on guinea pigs. Preliminary immun... more Immunogenic properties of cytolysin were studied in experiments on guinea pigs. Preliminary immunization with cytolysin led to the suppression of response to ConA in lymphocytes not adhering to nylon wool and to the stimulation of response to Legionella antigens in lymphocytes adhering to nylon wool. For a month after infection with L. pneumophila the suppression of the proliferative activity of lymphocytes in the spleen of the immunized animals in response to ConA and Legionella antigens was observed, while in the lungs transitory suppression of response to ConA and Legionella antigens was followed by the restoration and then stimulation of proliferation in response to T-cell mitogen and specific antigens. The data obtained in these experiments indicate the capacity of cytolysin for modulating the development of immune response.

MATERIALS AND METHODS The medical product of antiviral and immune modulating effect--fortepren (sodium polyprenyl phosphate) as a 4 mg/ml solution for injections combined with the base course of acyclic nucleoside acyclovir, 400 mg tablets, held studies. 40 male and female patients participated in the study. Введение. Интерфероны (IFN) и их индукторы эффективны в подавлении вирусной репродукции и коррекц... more Введение. Интерфероны (IFN) и их индукторы эффективны в подавлении вирусной репродукции и коррекции механизмов врождённого иммунитета. Цель исследования-проверить гипотезу возможного участия индуктора IFN ЦелАгрип (ЦА) в качестве активатора или супрессора противовирусного ответа в культурах клеток лимфомы Бёркитта (ЛБ), различающихся по способности продуцировать антигены вируса Эпштейна-Барр (ВЭБ). Материал и методы. Использовали линии человеческих клеток ЛБ Р3НR-1 и Namalva, спонтанно продуцирующие и не продуцирующие антигены ВЭБ. Выполнен кинетический анализ динамики продукции активных форм кислорода (АФК) и определена экспрессия группы генов методом ПЦР в реальном времени в ответ на обработку ЦА. Результаты и обсуждение. При обработке ЦA в клетках Namalva обнаружено снижение индекса активации АФК, в клетках Р3НR-1 наблюдали его повышение. В клетках Namalva после обработки ЦA отсутствовала достоверная активация генов IFN-α,-β и-λ, но возрастала кратность экспрессии гена ISG15 более чем в 1200 раз и гена P53 (TP53)-в 4,5 раза. При обработке ЦA клеток Р3НR-1 экспрессия генов IFN-α возрастала более чем в 200 раз, IFN-λ-в 100 раз и гена ISG15-в 2,2 раза. Предполагается взаимосвязь IFNиндуцирующего действия ЦА с активностью ISG15 и АФК в перевиваемых культурах клеток ЛБ, продуцирующих и не продуцирующих антигены ВЭБ. Заключение. В клетках Namalva, не продуцирующих антигены ВЭБ, при обработке ЦА подавляется генерация АФК и активизируется экспрессия генов ISG15 и P53 (TP53); в клетках Р3НR-1, продуцирующих антигены ВЭБ, наблюдается обратная картина: активизируются образование АФК и экспрессия генов IFN-α и-λ и супрессируется активность генов ISG15 и P53 (TP53).

A pharmacokinetic study of the absorption into the bloodstream, bioavailability and excretion of ... more A pharmacokinetic study of the absorption into the bloodstream, bioavailability and excretion of Gamavit from the body after intramuscular administration to laboratory mini-pigs was conducted. Quantitative determination was carried out by HPLC using a fluorimetric detector, for which Gamavit was labeled with Cy5 dye, which was then used for mini-pigs inoculation. The developed methods for determining Gamavit in the blood and feces were validated according to the following validation parameters: selectivity, calibration curve, accuracy, precision, limit of quantitative determination, sample transfer, and sample stability. The confirmed analytical range of the method for Gamavit detection in blood plasma and feces was 1.00…50.0 mcg/ml. Interferons (IFN) and IFN inducers are effective in suppressing viral reproduction and correcting of the innate immunity mechanisms.The aim of the study was to test the hypothesis of the possible involvement of the IFN inducer CelAgrip (CA) as an activator or suppressor of antiviral effects in Burkitt’s lymphoma (LB) cell cultures with different ability to produce Epstein-Barr virus antigens (EBV).Material and methods. The kinetic analysis of the dynamics of reactive oxygen species (ROS) production and determination of gene group expression by real-time PCR in response to CA treatment were done in human cell lines LB P3HR-1 and Namalva, spontaneously producing and not producing EBV antigens.Results and discussion. When treating CA in Namalva cells, a decrease in the ROS activation index was found; in P3HR-1 cells, an increase was observed.
