Here's how I'd see a Japanese translation of Homestar Runner:

Homestar's name would have a "-kun" behind in when his full name of either Homestar Runner or Homestar Michael Runner isn't used.
Strong Bad would be named Evil-san.
Strong Mad would be named Anger-san.
Strong Sad would be named Sadness-san.
The King of Town would be named Tokyo Daimao (Great King Tokyo).
Marzipan would be named Konpeito (the name of a Japanese candy that the Star Bits of the Mario series are modeled after).
Homsar, like Homestar, would have a "-kun" at the end of his name.
Coach Z would be considered female, and speak with a Kansai accent.
Bubs would sometimes put English words in his dialogue.
The Cheat would be named Password-san.
Strong Bad's Bottom 10 would be changed to this:

1. Puking on your own keyboard.
2. Songs with very annoying vocal sounds.
3. Chocolate recipie mistakes.
4. I was dared by Homestar-kun and Sadness-san to hug an evergreen tree.
5. Not...sparrows.
6. The horrible smell of a wet Password-san.
7. Smaller versions of foodstuffs that are already small.
8. Emails with multiple Fwd: and Re: in the subject line.
9. The haunting painting in Anger-san's closet since we were children.
10. Badly-designed shirt sales.

Pom Pom would speak with splashing noises rather than bubbling noises.
The Poopsmith's name would be changed to The Refuse Shoveler, and his head would be colored dark brown.
The topic of "big white face" would instead be "fat sad-looking man".
Nebulon's signature sound would be lifted from the FDS version of Metroid rather than Star Trek for Windows 9x.
Senor Cardgage's dialogue would just be broken Japanese, and he'd just be called Cargage-sama.
A cheetah would replace Pikachu in The Cheat Music Video.
The "Eastern-European Emails" would actually be Chinese rather than Polish.
Cold Ones would be re-named "Moontori", as a parody of Suntori (used to sponsor the Japanese version of Tapper).
StrongBadia would be called Awesomeopolis.
Trogdor would be called "The Strong-Armed Dragon".
The people from England that Strong Bad offends in sbemail_22 would be Korean rather than English.
The floppy disk tray would feature the Japanese names for Mega Drive/Genesis, Famicom, and Super Famicom games.
The "No Loafing" sign would be re-written as "Computer Users Cannot Live Off Of Hentai Alone".
The poster in the Strong Bad Main Page that's revealed when highlighting "E-mail" would be of Panty & Stocking.
The titles of various Strong Bad Emails would be different such as "Very Boring Day" (boring...really), "Password-san's Birthday" (cheatday), "Alter-Egos" (secret identity), "The Boring Person At The Office" (dullard), "Fantasy Creature" (animal), "Comedy 101" (the basics), "Learning About Anger-san" (the facts), "Anime by Evil-san" (japanese cartoon), "Bizarre Animation" (crazy cartoon), "The Failed Comic" (rough copy), "Homestar-kun's Dance Party" (nightlife), and "A Message to Sadness-san" (depressio).
Many of the American pop-culture references would be replaced with Japanese pop-culture references.
