Objective To investigate the electrophysiological and anti-inflammatory effects of fingolimod on an experimental penicillin-induced acute epileptic seizure model in rats. Methods Thirty-two male Wistar rats were divided into four groups: control (penicillin), positive control (penicillin + diazepam [5 mg/kg]), drug (penicillin + fingolimod [0.3 mg/kg]) and synergy group (penicillin + diazepam + fingolimod). The animals were anesthetized with urethane, and epileptiform activity was induced by intracortical injection of penicillin (500,000 IU). After electrophysiological recording for 125 minutes, IL-1β, TNF-α, and IL-6 were evaluated by ELISA in the serum of sacrificed animals. Results During the experiment, animal deaths occurred in the synergy group due to the synergistic negative chronotropic effect of diazepam and fingolimod. Although not stati...

However, less is known about the effects of carvacrol administration following ischemia and reperfusion. It was hypothesized that carvacrol might have protective effects on motor neuron functionunder ischemia-and reperfusion-induced oxidative stress. A total of 24 adult Wistar rats were divided into three groups: group I (control group; n = 8), group II (spinal ischemia and reperfusion group; n = 8), and group III (spinal ischemia and reperfusion + carvacrol group; n = 8). Ischemia and reperfusion were performed by clamping the abdominal aorta for 45 min. Clamps were then removed and 100 mg/kg of carvacrol was administered to group III. In the control groups a vehicle solution was administered. Animals were then observed for motor deficit index 48 h following the ischemia. Prior to the termination of the experiment, blood serum was obtained through intracardiac puncture for analyses of total antioxidant status and total oxidative stress levels. The results did not show effects on total antioxidant status or total oxidative stress levels. However, the motor deficit index was significantly different between the carvacrol and spinal ischemia groups and between the control and spinal ischemia groups. Our study demonstrated improved motor function in spinal ischemia models following carvacrol administration. However, future studies are required to determine the mechanism which improves motor function under carvacrol administration.

Two forms of NS were used in the study (Volatile oil (NSVO) and the ethanolic extract (NSEE)). A total of 50 rats were randomly assigned to the sham group (n = 10), control group (n = 10), NSVO group (n = 10), NSEE group (n = 10), and the Seprafilm group(n = 10). After 14 days, rats were sacrificed. Adhesions were examined macroscopically, and degree of adhesions was scored. A part of horn was excised, and superoxide dismutase (SOD) and glutathione peroxidase activities as well as malondialdehyde levels were evaluated, and histological score was calculated. Total microscopic score of the NSEE group was significantly lower than the control group (p = .001) and was marginally significantly lower than the seprafilm group (p = .005). Collagen formation score was higher in the seprafilm group compared to the sham and NSEE groups (p < 0.001, p = .003, respectively). Apoptotic cells were lower in the NSEE group compared to the control group (p = .003) and also lower in the NSEE and NSVO groups compared to the seprafilm group (p = .001, p < .001, respectively). Only SOD activity was higher in the NSVO and seprafilm groups compared to the control group (p < .001). NSEE form seems to have a possible effect in the prevention of PPAs. This may occur by its effect in decreasing collagen formation and by decreasing apoptosis in the injured tissues. NSVO form seems to induce SOD. Therefore, combined use of NSVO with seprafilm may increase the adhesion preventive effect of seprafilm.

Materials and Methods: Flutamide was injected in the abdomen of pregnant rats daily from days 14 to 20 of gestation. Male offspring with cryptorchidism were randomly divided into 2 groups. Healthy male rats without undescended testes comprised the control group (group 1). Group 2 (undescended testes without N-acetylcysteine) received no treatment. Group 3 (undescended testes plus N-acetylcysteine) received intraperitoneal N-acetylcysteine daily. At 70 days after experiment initiation the testes were removed for histopathological and biochemical analysis. Results: Mean malonyl dialdehyde values were lowest in group 1 and highest in group 2. In group 3 malonyl dialdehyde levels were significantly lower than in group 2 (p <0.001). Conversely, mean glutathione peroxidase was highest in group 1 and lowest in group 2. Glutathione peroxidase levels in group 3 were significantly higher than in group 2 (p <0.001). Histopathological differences between groups 1 and 3 in the modified Johnsen score were not significant (p ¼ 0.041). However, the differences between these groups and group 2 were significant (p <0.001). The median apoptotic cell count did not differ between groups 1 and 3 but it was significantly higher in group 2 than in the other groups (p ¼ 0.03 and <0.001, respectively). Conclusions: N-acetylcysteine may alleviate undescended testis induced damage to testes through its antioxidant effects. The underlying mechanism of these effects merits further investigation. Long-term studies are also needed as well as comparative animal and human studies.

Methods: This experimental study included 18 male Mongolian gerbils which were divided into three groups; sham-control group, penicillin group (500 units) and exercise + penicillin (500 units) group. Each animal group was composed of six Mongolian gerbils. The epileptiform activity was verified by electrocorticographic recordings. Results: The latency of the penicillin+exercise group was longer than the penicillin group, but this difference was not statistically significant. Following the penicillin administration, spike wave frequencies of epileptiform activity in the 10, 30, and 35 minutes were significantly lower in the penicillin+exercise group, compared with the penicillin group. There were generally significant decreases in the spike wave amplitude medians in the penicillin+exercise groups compared with the penicillin group in all time periods between 0 and 5 minutes. The serum superoxide dismutase, catalase and glutathione peroxidase levels increased in the penicillin+exercise group compared with those in the penicillin group. Conclusion: The results of present study indicate that regular exercise may contribute to the amelioration of epileptic activity by increasing the antioxidant effect.

Fibroblasts and myofibroblasts, are the cells that show significant activation in the development of OSF. Bleomycin (BL) is a chemotherapeutic agent commonly used in cancer treatment that also causes inflammatory changes in the oral mucosa, initiating fibrosis in the oral submucosal tissue. Sesamol (SE) has antioxidative and anti-inflammatory properties which are abundant in sesame seeds and sesame oil, and SE has a beneficial effect on the mucosal layer. This experimental study aimed to investigate SE's effects on BL-induced OSF. Materials and Methods: The study obtained 18 healthy adult male albino rats aged 3-4 months and weighing 200-250g from Bolu Abant Izzet Baysal University's Experimental Animal Application and Research Center. The rats were divided randomly into control, BL, and BL + SE groups (n = 6 in each group). A model of OSF was established in the rats by administering 0.5 mg/mL of BL and 50 mg\kg of SE to the BL+SE group each day. The hematoxylin and eosin stain and Masson's trichrome stain were used to assess histopathological changes in the oral mucosa. Results: The results revealed SE to have beneficial effects on BL-induced OSF through its antioxidant and anti-inflammatory properties. Histopathological evaluations and biochemical analysis of oral submucosal tissue samples also revealed SE to provide statistically significant protection against fibrosis in the oral mucosa (p < 0.01). Conclusion: This study has demonstrated oral submucous fibrosis that develops due to BL as well as SE to have an antioxidant effect on OSF regarding BL-induced reactive oxygen species (ROS) activation and collagen synthesis.

Methods: A total of 28 newborn Sprague Dawley rats were used and the OIR model was created using the 50/10% oxygen model. The study composed of four groups in total. While the OIR model was not used in Group I (control group), it was created for Groups II, III, and IV. About 0.01 mL carvacrol, bevacizumab, or 0.9% NaCl was administered intraperitoneal (IP) to the rats in all groups on postnatal day (PND) 14 as follows: Group I and Group II were administered 0.9% NaCl, Group III was administered bevacizumab, and Group IV was administered carvacrol. On PND 18, rats were sacrificed and their right eyes were enucleated. Results: Histopathological and immunohistochemical studies showed that the number of vascular endothelial cells (VECs), vascular endothelial growth factor (VEGF), and tumor necrosis factor-α (TNF-α) decreased similarly in Group III and Group IV compared with Group II. VECs values for Group I, Group II, Group III, and Group IV were measured as 0 ± 0, 26.45 ± 4.57, 7.75 ± 1.98, and 5.78 ± 1.72, respectively, and it differed significantly between groups (P < 0.001). Likewise, VEGF levels were observed as 0.06 ± 0.01, 3.31 ± 0.53, 2.47 ± 0.44, and 2.49 ± 0.52, respectively, and it differed significantly between groups (P < 0.001). TNF-α levels were recorded as 0.06 ± 0.01, 3.58 ± 0.38, 2.46 ± 0.49, and 2.29 ± 0.25, respectively, and it differed significantly between groups (P < 0.001). VECs, VEGF, and TNF-α were similar between Group III and IV (range of P values were 0.486–0.998). Conclusion: The study demonstrated that carvacrol significantly reduced retinal pathological angiogenesis, NV, VEC nuclei count, VEGF, and TNF-α levels. Moreover, the observed effects were comparable to those of bevacizumab.

Dexosomes were isolated purely from small-scale-up production by using t25-cell-culture flasks. Total RNA was measured as 1.43 ± 0.33 ng/106 cell. Despite the fact that they possessed a surface that is highly abundant in protein, this did not become a significant effect on the DOX loading amount. Ultrasonication was used for doxorubicin (DOX) loading into the IDc dexosomes. In accordance with the literature, three candidate DOX formulations were designed as IC50 values; dExoIII, 1.8 µg/mL, dExoII, 1.2 µg/mL, and dExoI, 0.6 µg/mL, respectively. Formulations were evaluated by MTT test against highly metastatic A549 (CCL-185; ATTC) cell line. Confocal images of unloaded (naïve) were obtained by CellMaskTM membrane staining before DOX loading. Although, dexosome membranes were highly durable subsequent to ultrasonication, it was observed that dexosomes could not be stable ab...

With that in mind, we investigate whether running exercises performed during pregnancy can contribute to the development of tolerance to neonatal hyperoxic brain damage. Method: While two female rats maintained a sedentary pregnancy, one female rat performed the mandatory running exercise for 30 minutes for five days a week throughout the pregnancy. Following delivery, the sedentary rats and the exercised rat were kept together with their offspring for five days at oxygen concentrations above 80 percent in order to induce brain damage. The offspring were sacrificed on postnatal Day 7 and brain/body ratio measurements were obtained. Results: The brain/body ratios in the control, hyperoxia and exercise-hyperoxia groups were found to be median (IQR) 0.

Research paper thumbnail of Improvement of antibacterial and biocompatibility properties of electrospray biopolymer films by ZnO and MCM-41

Polymer Bulletin, 2019

This study aims the improvement of antibacterial and biocompatibility properties of electrospray ... more This study aims the improvement of antibacterial and biocompatibility properties of electrospray ternary blends of chitosan/poly(ethylene glycol)/hyaluronic acid. It conserves microscale particle structure even after incorporating zinc oxide (ZnO), the zeolite Mobil Composition of Matter No. 41 (MCM41) and penicillin G during this technique. Three different electrospray (ESP) blend compositions (ESPI, ESPII and ESPIII) have been produced in order to improve both antibacterial activity against to both gram-positive and gram-negative bacteria and biocompatibility. Results of FTIR spectroscopy and microscopy verified with SEM, EDS and AFM analyses. Hyaluronic acid surface has been specified definitely through ZnO-based ESPI surface composed of heterogeneously dispersed microparticles. Surface structures of ESPII and ESPIII have more homogenously dispersed microparticles as hill-valley surface by the aid of MCM 41-PEN. Antibacterial activity has been performed by Kirby-Bauer method. ESPI has good antibacterial activity against both gram-positive (S. aureus and S. epidermidis) and gram-negative bacteria (E. cloacea). Each electrospray film displayed good biocompatibility against to mouse fibroblast cell line L929 (ATTC number CCL-1). The highest amount of cell proliferation has been detected on ESPIII surface.

Research paper thumbnail of Effects of gestational exercise on hyperoxia-induced brain damage in the newborn

Preterm infants encounter hyperoxia relatively early on as they leave the intrauterine environmen... more Preterm infants encounter hyperoxia relatively early on as they leave the intrauterine environment earlier than expected, while also being exposed to a higher level of hyperoxic stress due to insufficiencies in their antioxidant defense mechanisms.
