The aim of this study was to investigate the therapeutic effects of propolis for the toxicity of ... more The aim of this study was to investigate the therapeutic effects of propolis for the toxicity of cypermethrin (CYP) on histopathological changes in tissues of rainbow trout (Oncorhynchus mykiss). CYP is one of the most toxic pyrethroids highly for the aquatic organisms. The fish were exposed to three sublethal concentrations of CYP (0.0041, 0.0082 and 0.0123 ppm). In addition, different concentrations of propolis (10, 20 and 30 ppm) were used in the investigation for the period of 96 h. Propolis was collected by honeybees from different plants to prevent oxidative damages as an antioxidant. The therapeutic concentration of propolis was determined at 10 ppm. 10 ppm propolis was added to three cypermethrin concentrations. Shorting the secondary lamellae , fusion of secondary lamellae, oedema, necrosis, vacuolization and cartilage damage in gill tissue of fish exposed to CYP were observed by histopatological analyses. Hepatic lesions in liver tissue of fish exposed to CYP were characterized as hydropic degeneration, necrosis, mononuclear cell infiltration and narrowing of sinusoids. The most common changes in muscle tissues at all concentrations of CYP were nuclear proliferation and congestion. Besides, 10 ppm propolis on gill, liver, muscle tissues of rainbow trouts showed significant therapeutic effects. Histological analysis revealed that propolis may serve as an antitoxic agent against pesticide toxicity.

The protective effects of propolis on biochemical and hematological parameters were appraised in ... more The protective effects of propolis on biochemical and hematological parameters were appraised in the blood of Oncorhynchus mykiss exposed to cypermethrin (CYP). The fish were subjected to three sublethal concentrations of CYP (0.0041, 0.0082 and 0.0123 ppm) and propolis (10 ppm) for 96 h. The samples were analysed to changes in the biochemical parameters, such as the metabolits: glucose, total protein, creatinine, urea, triglycerides, total cholesterol levels and the enzymes: Alanine Aminotransferase (ALT), Alanine Aminotransferase (AST), Alkaline Phosphatase (ALP), Lactate Dehydrogenase (LDH), amilase, Gamma Glutamyltransferase (GGT) and the electrolites: chloride, sodium, potassium, calcium, phosphate and ferric. Also, hematological parameters including total leucocyte count, granulocytes, agranulocytes and the erytrocyte count: Hemoglobin, hematocrit and the erytrocyte indexes: Mean corpuscular volume (MCV), mean corpuscular hemoglobin (MCH) and mean corpuscular hemoglobin concentration (MCHC) were tested in fish blood. The data of this investigation showed that CYP had a negative effect on the biochemical and hematological variables of the fish; these toxic effects were neutralised by the application of propolis. The results suggest that propolis can be effective in the prevention of CYP-induced toxicity in rainbow trout, especially hematopoiesis.

Ladik Lake (Samsun, Turkey) is a natural landscape under the threat of pollution because of urban... more Ladik Lake (Samsun, Turkey) is a natural landscape under the threat of pollution because of urban, agricultural and industrial activities. In this study, accumulation of heavy metals (Al, Ba, Cr, Cu, Fe, Mn, Pb and Zn) in the tissues (muscle, liver and gill) of sediment dwelling pike fish and in different substrates of Ladik Lake were investigated. Seasonal haematological, biochemical and histopathological parameters of the carp are also reported. In general, heavy-metal levels in water and sediment samples were found in the highest level in summer and the lowest in autumn. Histopathologic changes in the tissues of the fish (liver and gill) were at the minimum in winter while it was highest in summer. Blood biochemical parameters exhibited higher level in summer in comparison with other seasons. Al, Ba, Cr, Mn and Zn levels in the tissues of the fish were in the order gill>liver>muscle in all seasons. The levels of Cu, Fe and Pb were in the sequence liver>gill>muscle. Cr, Cu, Pb and Zn levels were determined to be in high level in the tissues of the fish with respect to the literature values. The heavy-metal levels in Ladik Lake and the fish health need to be regularly monitored for a sustainable environmental health.

The purpose of this study was to investigate the therapeutic effects of natural products like pro... more The purpose of this study was to investigate the therapeutic effects of natural products like propolis on biochemical and hematologic parameters in carp (Cyprinus carpio, Linnaeus 1758) exposed to arsenic. In this study fish were exposed to 0.01 mg/L arsenic and 10 mg/L propolis for seven days. Our results indicated that triglyceride, urea, total cholesterol, cobalt, ALT (alanine amino transferase), AST (aspartate amino transferase), LDH (lactate dehydrogenase) values increased when exposed to arsenic (P<0.05) but reduced by combination of arsenic and propolis (P<0.05). Granulocyte, erythrocyte, hemoglobin, hematocrit values were decreased by use of arsenic in comparison to control group. These parameters increased in arsenic+propolis group (P<0.05). In addition levels of leucocyte, agranulocyte, MCV (mean corpuscular volume), MCH (mean corpuscular hemoglobin) and MCHC (mean corpuscular hemoglobin concentration) increased in the arsenic group (P<0.05) and decreased in arsenic+propolis group (P<0.05). We can say that propolis can improve biochemical and hematologic functions of common carp blood, after being exposed to arsenic.

In this study, hematological and biochemical investigations were carried out on Cyprinus carpio, ... more In this study, hematological and biochemical investigations were carried out on Cyprinus carpio, Leuciscus cephalus, Capoeta trutta ve Capoeta capoeta umbla fish species living in Sultansuyu Dam Lake (Malatya). The physical, chemical and microbiological features of the reservoir were specified and heavy metal analyses were also conducted. As a result of hematological investigation, a statistical increase of P<0.05 in total leukocyte count, and a decrease of P<0.05 in erytrocyte, hemoglobin and hematocrit values were observed. With respect to microbiological analysis, the presence of Enterococcus shows the drainage of domestic wastes into the reservoir. With water analyses, it was found out that the water quality was second rate and the sulphur content in the water was above standard levels. The existence of large amount of sulphur was presumed to be the result of apricot processing. Significant heavy metal pollution was not observed.

Rats are given L-NAME for 15 days and the propolis for the last 5 days with L-NAME together. The levels of aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase and gamma glutamyltransferase in the L-NAME group compared to control group have increased (P<0.05). The levels of these parameters in L-NAME+propolis group compared to the L-NAME group have decreased (P<0.05). L-NAME caused increase (P<0.05) in levels of glucose, albumin, globulin, creatinine, urea, triglyceride and cholesterol. Erythrocyte number, total leukocyte, hemoglobin, hematocrit, neutrophil and monocyte decreased (P<0.05), platelets and lymphocyte increased (P<0.05) in L-NAME+propolis group compared to the L-NAME group. The rainbow trout were exposed to heavy metal stress (Cd+2, Cr+3) at 2 ppm dosage. The present study was undertaken to determine the protective effect of selenium treatment at the same dosage (2 ppm) on some biochemical parameters. The activity of catalase (CAT), glutathione peroxidase (GSH-Px), superoxide dismutase (SOD) and the changes in levels of malondialdehyde (MDA) from biochemical parameters were determined in liver tissue of the fish groups exposed to heavy metals, especially for the selenium-applied groups. Results of this study showed that the activities of CAT, GSH-Px and SOD in the tissues of fish exposed to the stress of Cd+2 and Cr+3 were significantly lower than the control groups (P &amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;lt; 0.05). Meanwhile, the closer values to the control groups were obtained in selenium-added groups (Cr+3 + Se+4, Cd+2 + Se+4). For the level of MDA, the last production of lipid peroxidation showed increases (P &amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;lt; 0.05) in the groups exposed to the metal stress, whereas significant decreases were obtained in selenium-applied groups. The result of the statistical evaluation showed that the negative effects occurring in the biochemical parameters of the applied groups exposed to the toxicity of heavy metal were significantly eliminated (P &amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;lt; 0.05) as a result of selenium treatment.

This study was carried out to understand the preventive effect of selenium (Se4+) on heavy metal ... more This study was carried out to understand the preventive effect of selenium (Se4+) on heavy metal stress induced by lead and copper in rainbow trout (Oncorhynchus mykiss). Variation in glutathione peroxidase (Se-GSH-Px) and superoxide dismutase (SOD) activity, and in malondialdehyde (MDA) levels in liver, spleen, heart, and brain tissues of rainbow trout after 72 h of exposure to Pb2+ and Cu2+ were investigated in the presence and absence of Se4+. In the presence of Se4+, Se-GSH-Px activity and SOD activity were found to be higher and MDA levels were lower compared with in its absence. Hematological parameters were also determined and it has been observed that total leukocyte count (WBC), mean cell volume (MCV), and mean cell hemoglobin (MCH) were increased and erythrocyte number (RBC), hemoglobin (Hb), and hematocrit value (Hct; P &amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;lt; 0.05) were decreased in fish exposed to heavy metals in the absence of selenium. Selenium presence recovered hematological parameters to normal levels. In the light of our findings, it could be stated that Pb2+ and Cu2+ lead to dramatic changes in biochemical and hematological parameters and selenium caused these parameters to converge to control levels when it was administered concurrently with these heavy metals.

Chemical toxic pollutants (especially heavy metals) are important sources of reactive oxygen spec... more Chemical toxic pollutants (especially heavy metals) are important sources of reactive oxygen species (ROS) in biological systems. Membrane phospholipids of aerobic organisms are continually subjected to oxidant challenges from endogenous and exogenous sources, while peroxidized membranes and lipid peroxidation products represent constant threats to aerobic cells. The primary antioxidant protection against free radical and ROS is provided by the enzymes glutathione peroxidase (GSH-Px), superoxide dismutase (SOD) and catalase (CAT), respectively. The trace element selenium has been implicated in chemo-prevention and drug-resistance through reduction of oxidative stress. Selenium could prevent damage to the unsaturated fatty acid of subcellular membranes by lipid peroxidation induced by free radicals. The results reported here show that sodium selenite has an important contribution to antioxidative defense for the spleen and heart of rainbow trout. The ability of sodium selenite to prevent the oxidative stress induced by heavy metals (Cd(2+), Cr(3+)) in fish was rationalized.

Propolis, a resinous material produced by worker bees from the leaf buds and exudates of plants, ... more Propolis, a resinous material produced by worker bees from the leaf buds and exudates of plants, is reported to possess various therapeutic properties. The aim of this study was to investigate the effects of propolis on biochemical parameters and histopathologic findings in carp Cyprinus carpio L. exposed to arsenic. A sublethal concentration of arsenic (0.01 mg l −1) and/or 10 mg l −1 propolis were administered to fish for 1 wk. Catalase (CAT) activities and malondialdehyde (MDA) levels were determined in liver, gill and muscle tissues in control, arsenic only, propolis only and arsenic+propolis treatment groups. Results showed that CAT activity decreased in the arsenic group compared to the control and propolis groups. CAT activity in the arsenic+ propolis group was significantly higher compared to the arsenic group. MDA levels in fish exposed to 0.01 mg l −1 arsenic significantly increased compared to the control group. However, MDA levels in the arsenic+propolis group were significantly lower compared to the arsenic group. Histopathological changes in the liver, gill and muscle tissues of carp were examined by light microscopy: various changes were observed in all tissues of fish in the arsenic group. Propolis showed important antioxidant effects against arsenic toxicity in all fish tissues.

Variation in glutathione peroxidase (Se-GSH-Px) and superoxide dismutase (SOD) activity, and in malondialdehyde (MDA) levels in liver, spleen, heart, and brain tissues of rainbow trout after 72 h of exposure to Pb2+ and Cu2+ were investigated in the presence and absence of Se4+. In the presence of Se4+, Se-GSH-Px activity and SOD activity were found to be higher and MDA levels were lower compared with in its absence. Hematological parameters were also determined and it has been observed that total leukocyte count (WBC), mean cell volume (MCV), and mean cell hemoglobin (MCH) were increased and erythrocyte number (RBC), hemoglobin (Hb), and hematocrit value (Hct; P &amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;amp;lt; 0.05) were decreased in fish exposed to heavy metals in the absence of selenium. Selenium presence recovered hematological parameters to normal levels. In the light of our findings, it could be stated that Pb2+ and Cu2+ lead to dramatic changes in biochemical and hematological parameters and selenium caused these parameters to converge to control levels when it was administered concurrently with these heavy metals.

Membrane phospholipids of aerobic organisms are continually subjected to oxidant challenges from endogenous and exogenous sources, while peroxidized membranes and lipid peroxidation products represent constant threats to aerobic cells. The primary antioxidant protection against free radical and ROS is provided by the enzymes glutathione peroxidase (GSH-Px), superoxide dismutase (SOD) and catalase (CAT), respectively. The trace element selenium has been implicated in chemo-prevention and drug-resistance through reduction of oxidative stress. Selenium could prevent damage to the unsaturated fatty acid of subcellular membranes by lipid peroxidation induced by free radicals. The results reported here show that sodium selenite has an important contribution to antioxidative defense for the spleen and heart of rainbow trout. The ability of sodium selenite to prevent the oxidative stress induced by heavy metals (Cd(2+), Cr(3+)) in fish was rationalized.

Propolis, a resinous material produced by worker bees from the leaf buds and exudates of plants, ... more Propolis, a resinous material produced by worker bees from the leaf buds and exudates of plants, is reported to possess various therapeutic properties. The aim of this study was to investigate the effects of propolis on biochemical parameters and histopathologic findings in carp Cyprinus carpio L. exposed to arsenic. A sublethal concentration of arsenic (0.01 mg l −1) and/or 10 mg l −1 propolis were administered to fish for 1 wk. Catalase (CAT) activities and malondialdehyde (MDA) levels were determined in liver, gill and muscle tissues in control, arsenic only, propolis only and arsenic+propolis treatment groups. Results showed that CAT activity decreased in the arsenic group compared to the control and propolis groups. CAT activity in the arsenic+ propolis group was significantly higher compared to the arsenic group. MDA levels in fish exposed to 0.01 mg l −1 arsenic significantly increased compared to the control group. However, MDA levels in the arsenic+propolis group were significantly lower compared to the arsenic group. Histopathological changes in the liver, gill and muscle tissues of carp were examined by light microscopy: various changes were observed in all tissues of fish in the arsenic group. Propolis showed important antioxidant effects against arsenic toxicity in all fish tissues.
