This study presents an animal model and aims to study the reactions of female rats to the loss of their litters and the consequences for the next litter. Methods: Primiparious Sprague-Dawley rats were recruited into three groups: of separation-reunion, complete separation and no-separation. Maternal behaviors following separation, reunion and the birth of the subsequent litter were observed. The blood corticosterone levels of the dams in response to separation and reunion were measured. Learning, activity, anxiety and blood corticosterone response to stress of the second litters were evaluated. Results: Dams in the separation group spent significantly more time at the nest following reunion, displayed increased pup licking in their second litters compared to their first litters and did not achieve the increased duration of nursing in their second generation litters as observed for the control group. Dams' blood corticosterone levels were significantly influenced by separation and reunion. Pups born to mothers that have experienced an early loss of their previous litters had significantly impaired cognitive abilities compared to controls. Conclusion: Further analysis of the process of loss in animal models may have clinical implications for bereaved mothers.

The aim of this study was to evaluate whether VLSs are efficient learning tools and how these simulations can be integrated into laboratory practice in medical laboratory education. Methods: In this pre-test/post-test control group study, 32 volunteers were randomly assigned to either experimental or control groups. The experimental group performed laboratory simulations based on biochemistry and microbiology and then completed a self-report survey to evaluate their satisfaction and beliefs about simulations. Results: In the experimental group, post-test scores of each simulation were significantly elevated compared to pre-test scores; however, pre-and post-test scores of control group were statistically the same. The experimental group agreed that these simulations should be applied before theoretical lectures and laboratory practices. They also highlighted that translating from English to their native language creates difficulties in applying and understanding the simulation. Conclusions: We emphasized that VLSs are excellent learning tools that increase not only the knowledge but also the self-motivation and focus of the students. Based on feedbacks, native language options are necessary to enable the students to achieve equality of opportunity in education.

Methods Un-stimulated whole saliva and serum samples were collected from a total of 30 systemically healthy participants with periodontally healthy smokers (S) (n=15) and nonsmokers (n=15). Periodontal indices (plaque index, gingival index, probing depth, bleeding on probing, clinical attachment level) were recorded to confirm periodontal health. Saliva was purified, and a total of 28 amino acids and metabolites were analyzed by liquid chromatographymass spectrometry (LC-MS/MS). Smoking status was validated measuring serum cotinine levels. Intergroup comparisons were assessed using the Mann Whitney U test. Results When 28 amino acids were evaluated, smokers had statistically significantly higher cystathionine levels than non-smokers (p <0.05). Conclusions Saliva cystathionine is associated with smoking in periodontally healthy individuals, and is possibly related to altered sulfuration pathway.

Progressive compromise of antioxidant defenses and free radical-mediated lipid peroxidation, whic... more Progressive compromise of antioxidant defenses and free radical-mediated lipid peroxidation, which is one of the major mechanisms of secondary traumatic brain injury (TBI), has also been reported in pediatric head trauma. In the present study, we aimed to demonstrate the effect of melatonin, which is a potent free radical scavenger, on brain oxidative damage in 7-day-old rat pups subjected to contusion injury. Whereas TBI significantly increased thiobarbituric acid reactive substances (TBARS) levels, there was no compensatory increase in the antioxidant enzymes such as superoxide dismutase (SOD) and glutathione peroxidase (GPx) 24 hours after TBI in 7-day-old rats. Melatonin administered as a single dose of 5 mg/kg prevented the increase in TBARS levels in both non-traumatized and traumatized brain hemispheres. In conclusion, melatonin protects against oxidative damage induced by TBI in the immature brain.

Physiological Research

In a previous study we demonstrated that acute footshock stress increased glutathione peroxidase ... more In a previous study we demonstrated that acute footshock stress increased glutathione peroxidase activity in the prefrontal cortex and striatum of adult male rats. Adolescents may respond differently to stress as life stressors may be greater than at other ages. The present study examined the effects of the acute footshock stress on superoxide dismutase (SOD) and glutathione peroxidase (GPx) enzyme activities and thiobarbituric acid reactive substances (TBARS) levels in adolescent male and female rat brains. We demonstrated that acute footshock stress increased SOD activity in the prefrontal cortex, and increased GPx activity in the hippocampus in female rats. In males, acute footshock stress increased GPx activity in the prefrontal cortex and hippocampus. Footshock stress did not change TBARS levels. These results indicate a strong role of gender in the response of adolescent subjects to various aspects of stress.

Physiological Research

Melatonin has recently been suggested as an antioxidant that may protect neurons from oxidative s... more Melatonin has recently been suggested as an antioxidant that may protect neurons from oxidative stress. Acute ethanol administration produces both lipid peroxidation as an indicator of oxidative stress in the brain and impairs water-maze performance in spatial learning and memory tasks. The present study investigated the effect of melatonin against ethanol-induced oxidative stress and spatial memory impairment. The Morris water maze was used to evaluate the cognitive functions of rats. Thiobarbituric acid reactive substances (TBARS), which are the indicators of lipid peroxidation, and the activities of antioxidative enzymes (glutathione peroxidase and superoxide dismutase) were measured in the rat hippocampus and prefrontal cortex which form interconnected neural circuits for spatial memory. Acute administration of ethanol significantly increased TBARS levels in the hippocampus. Combined melatonin-ethanol treatment caused a significant increase in glutathione peroxidase activities a...

The possible modulatory effect of erythropoietin on inducible nitric oxide synthase (iNOS) mRNA expression and nitrite production were also analyzed. Erythropoietin exerted a significant protective effect against IFN-gamma and LPS-induced oligodendrocyte injury as determined by lactate dehydrogenase assay. Treatment with erythropoietin inhibited the expression of iNOS mRNA and nitrite production resulting from proinflammatory stimulation by IFN-gamma and LPS. These results suggest that erythropoietin has protective effects against inflammatory oligodendrocyte injury in vitro and may play a protective role in neurological disorders characterized by oligodendrocyte death, such as multiple sclerosis.

In this context, we created clinical integration sessions (CIS) in our first-year medical curriculum. Methods: The instructors of different disciplines wrote the clinical scenarios together. The scenarios were discussed in five sessions with 39 first-year students. The first session's scenario consisted of four brief "anemia" cases. The next four sessions included a single case, according to the feedback of the students. Students formed groups of 7-8 participants. In the first 2 h, the scenarios were discussed in the groups and questions were answered by the students. In the third hour, the instructors answered the questions together with the students. After the first CIS, written feedback obtained from the students via a survey. Results: The survey provided positive feedback on the benefits of active learning within small group discussions, and most of the students thought that their background was sufficient to solve the cases, with some literature search. Conclusion: The scenarios, which provide multidisciplinary integration of basic medical sciences and clinical medicine, can be useful educational materials.
