This work aims to discuss how controversial themes echo transformed into verbal-imagery texts (ca... more This work aims to discuss how controversial themes echo transformed into verbal-imagery texts (cartoons, memes, comic strips etc.) in order to suggest a treatment of these as pedagogical tools for history classes. This is done in a way that humor establishes itself as an activator of multimodal reading strategies (KESSESS, van LEEUWEN, 1996) for the contribution of critical visual literacy practices (LUKE and FREEBODY, 1997; CERVETTI, 2001). The images are limited to the controversy caused by the proposal of the subject "Special Topics IV: the coup of 2016 and the future of democracy", in the course of Political Science, University of Brasilia. For this, we rely on Critical Discourse Analysis (FAIRCLOUGH, 1992; 2003). We believe that the proposal rests on the need to look at the images for the development of a theoretical instrument capable of helping teachers and students to analyze, interpret and appropriate the image content, leading from it critical discussions.

An experimental study based on the thermal analysis of 5-methyl-1H-benzotriazole and 5,6-dimethyl... more An experimental study based on the thermal analysis of 5-methyl-1H-benzotriazole and 5,6-dimethyl-1H-benzotriazole was developed, by using differential scanning calorimetry. Additionally, a summary of the experimental techniques and computational methodology being performed, in order to complement the energetic study of both compounds, is described. The knowledge of the thermochemical, thermophysical and structural properties of functionalized benzotriazoles is relevant for the evaluation of their chemical behaviour, as well as in the prediction of the reactivity of similar compounds that have not been thermodynamically characterized.

Invasive fungal infections (IFI) have significantly increased over the past years due to advances... more Invasive fungal infections (IFI) have significantly increased over the past years due to advances in medical care for the at-risk immunocompromised population. IFI are often difficult to diagnose and manage, and can be associated with substantial morbidity and mortality. This study aims to contribute to understanding the etiology of invasive and subcutaneous fungal infections, their associated risk factors, and to perceive the outcome of patients who developed invasive disease, raising awareness of these infections at a local level but also in a global context. A laboratory surveillance approach was conducted over a seven-year period and included: (i) cases of invasive and subcutaneous fungal infections caused by filamentous/dimorphic fungi, confirmed by either microscopy or positive culture from sterile samples, (ii) cases diagnosed as probable IFI according to the criteria established by EORTC/MSG when duly substantiated. Os objectivos deste estudo foi comparar por sexo, prática e frequência de actividade física ao ní... more Os objectivos deste estudo foi comparar por sexo, prática e frequência de actividade física ao nível da depressão global. A amostra deste estudo foi constituída por 183 indivíduos, sendo 38,3% do sexo masculino e 61,7% do sexo feminino. Os resultados indicam que são os homens quem mais pratica, com maior frequência e com sessões mais longas de actividade. Constatou-se a existência de efeitos significativos nas comparações por sexo ao nível da depressão global e na dimensão afectivo-somática, assim como há diferenças significativas entre sexos quanto à prática de actividade física e na depressão. Posto isto, concluímos que os homens praticam mais do que as mulheres, e elas tendem a apresentar valores de depressão superiores. As mulheres apresentam níveis mais elevados de depressão em todas as suas dimensões e de forma significativa na dimensão afectivo-somática.

The aim of this study was to compare the prevalence of conventional CAD risk factors, as well as the plasma aminothiol profile (and its major determinants), between two groups of healthy subjects from Ponta Delgada (in Azores) and Lisbon (in mainland) cities, searching for precocious biomarker(s) of the disease. The study groups consisted of 101 healthy volunteers from Ponta Delgada (PDL) and 121 from Lisbon, aged 20–69 years. No differences in the prevalence of classical CAD risk factors were found between the study groups, except in physical inactivity and related central obesity, which were both higher in PDL men than in those from Lisbon. To evaluate the plasma aminothiol profile (PAP) and serum gamma-glutamyltransferase (GGT) activity, as well as plasma folate, vitamin B12, and vitamin B6concentrations, in 326 apparently healthy subjects from the Azores archipelago (Portugal). Also eventual relationships of PAP with conventional risk factors for atherosclerosis were investigated, aiming at the finding of early blood markers of the disease.Design and Methods. This was an observational cross-sectional study, where participants were split into two groups: one with a normal and another with an altered PAP (at least one aminothiol out of its reference concentration range).Results. About 76% of subjects had an altered PAP, mainly due to low glutathione levels (<1.5 μmol/L), mostly associated with normal GGT activity. The act of consumption is a way to structure and give meaning to the world and the relationships ... more The act of consumption is a way to structure and give meaning to the world and the relationships established among people. During the transition from modernity, i.e., from modernity to postmodernity (Rajj, 1998), in a consumer culture (Slater, 1997), the products and their signal value and consumption assume a central role in creating meaning for individuals (Solomon, 1983; Fournier, 1998), thus affecting the differentiation and integration of individuals in the social world. Today consumers are dedicated to the consumption of experiences and feelings about brands, far beyond the functionality of the product. This promotes the consumption value of the bond and connection between consumers' tastes, opinions and preferences. Thus, consumption in social and convivial contexts overlaps isolated and egocentric consumption. Given this new reality, blurring the differences between the materials products makes the concept of brand a key issue in the identification and differentiation. User groups and admirers of a brand that engage in joint activities to achieve collective goals and or to express mutual feelings, experiences and commitments are known as brand communities (Stokburger Sauer, 2010). They consume the symbolic meaning of products and not only the product features, and the image and the emotional connection is more important than functionality. In the social context of consumption of these communities should be studied both their interactions and the direct and indirect impact to organizations. It is intended to investigate and explain the interdependence between the concepts of brand community and brand value, with respect to the motivations for participation in the brand community, its characteristics and organic and ultimately the relationship with the cognitive dimensions of brand equity.

Respiratory syncytial virus (RSV) is responsible for annual respiratory infection outbreaks in in... more Respiratory syncytial virus (RSV) is responsible for annual respiratory infection outbreaks in infants and young children worldwide, frequently causing bronchiolitis and pneumonia. We evaluated clinical and epidemiological features of acute respiratory infections (ARIs) caused by respiratory syncytial virus (RSV) in children less than five years old. Nasopharyngeal aspirate samples from children with ARI symptoms, attended at the 'Hospital das Clínicas'-Federal University of Uberlândia, MG, Brazil, were collected and tested for RSV by the immunofluorescence assay (IFA). Patients' clinical and epidemiological data were also obtained. From April 2000 to June 2003, 317 nasopharyngeal samples were collected from children less than 54 months old. Seventy-six samples (24.0%) were positive for RSV, with 53% (40/76) obtained from male patients. Hospitalization occurred in 50% (38/76) of the cases, with an average period of 10.6 days, in most cases (87%, 33/38) occurring in children less than 12 months of age. Although an association between this age group and the presentation of more severe clinical symptoms was observed, such as bronchiolitis in 51% (27/53) of the patients and pneumonia in 19% (10/53), no patients died. RSV was found from February to August, with the highest incidence in May. Conclusions: RSV is an important agent that causes ARIs; the clinical manifestations varied from mild to severe and patients frequently required hospitalization; RSV mostly affected children less than one year old.
Marmosets are cooperative breeders organized as extended family groups, but breeding is generally... more Marmosets are cooperative breeders organized as extended family groups, but breeding is generally restricted to a single pair. Breeding competition is fierce in female marmosets; males, on the other hand, show low levels of intragroup aggression. We investigated male and female breeding strategies and the resulting reproductive output in 9 wild groups. Reproductive output, tenure of breeding animals, identification of the breeding system, breeding position replacements, migration and infanticide were recorded; also, we recorded grooming and aggression. Replacement of the breeding male or female was observed on nine occasions. On four occasions, the son of the breeding male inherited the breeding post, but we never observed inheritance of a breeding post by a daughter. Mostly, females attained a breeding post by immigrating to a group that had a breeding vacancy. Our results showed that Callithrix jacchus males and females use different strategies to attain a breeding position and maintain it for as long as possible. These strategies prolong the tenure of the breeding position, which is the best way to produce a large number of offspring. This article is part of a Special Issue entitled: Neotropical Behaviour.

We have previously reported that exogenous bradykinin activates immature dendritic cells (DCs) vi... more We have previously reported that exogenous bradykinin activates immature dendritic cells (DCs) via the bradykinin B2 receptor (B2R), thereby stimulating adaptive immunity. In this study, we show that these premises are met in a model of s.c. infection by Trypanosoma cruzi, a protozoan that liberates kinins from kininogens through its major protease, cruzipain. Intensity of B2R-dependent paw edema evoked by trypomastigotes correlated with levels of IL-12 produced by CD11c+ dendritic cells isolated from draining lymph nodes. The IL-12 response induced by endogenously released kinins was vigorously increased in infected mice pretreated with inhibitors of angiotensin converting enzyme (ACE), a kinin-degrading metallopeptidase. Furthermore, these innate stimulatory effects were linked to B2R-dependent up-regulation of IFN-γ production by Ag-specific T cells. Here we demonstrate that the invasion of human primary umbilical vein endothelial cells (HUVECs) or Chinese hamster ovary (CHO) cells overexpressing the B2 type of bradykinin receptor (CHO-B2R) by tissue culture trypomastigotes is subtly modulated by the combined activities of kininogens, kininogenases, and kinin-degrading peptidases. The presence of captopril, an inhibitor of bradykinin degradation by kininase II, drastically potentiated parasitic invasion of HUVECs and CHO-B2R, but not of mock-transfected CHO cells, whereas the B2R antagonist HOE 140 or monoclonal antibody MBK3 to bradykinin blocked these effects. Trypanosoma cruzi activates the kinin pathway through the activity of its major cysteine proteina... more Trypanosoma cruzi activates the kinin pathway through the activity of its major cysteine proteinase, cruzipain. Because kininogen molecules may be displayed on cell surfaces by binding to glycosaminoglycans, we examined whether the ability of cruzipain to release kinins from high molecular weight kininogen (HK) is modulated by heparan sulfate (HS). Kinetic assays show that HS reduces the cysteine proteinase inhibitory activity (K i app) of HK about 10-fold. Conversely, the catalytic efficiency of cruzipain on kinin-related synthetic fluorogenic substrates is enhanced up to 6-fold in the presence of HS. Analysis of the HK breakdown products generated by cruzipain indicated that HS changes the pattern of HK cleavage products. Direct measurements of bradykinin demonstrated an up to 35fold increase in cruzipain-mediated kinin liberation in the presence of HS. Similarly, kinin release by living trypomastigotes increased up to 10-fold in the presence of HS. These studies suggest that the efficiency of T. cruzi to initiate kinin release is potently enhanced by the mutual interactions between cruzipain, HK, and heparan sulfate proteoglycans.

The major isoform of Trypanosoma cruzi cysteinyl proteinase (cruzipain) has generated Lys-bradyki... more The major isoform of Trypanosoma cruzi cysteinyl proteinase (cruzipain) has generated Lys-bradykinin (Lys-BK or kallidin), a proinflammatory peptide, by proteolysis of kininogen. The releasing of this peptide was demonstrated by mass spectrometry, radioimmunoassay, and ileum contractile responses. The kinin-releasing activity was immunoabsorbed selectively by monoclonal antibodies to the characteristic COOH-terminal domain of cruzipain. To determine the hydrolysis steps that account for the kininogenase activity of cruzipain, we synthesized a fluorogenic peptide (o-aminobenzoyl-Leu-Gly-Met-Ile-Ser-Leu-Met-Lys-Arg-Pro-Pro-Gly-Phe-Ser-Pro-Phe-Arg 389-Ser 390-Ser-Arg-Ile-NH 2) based on the sequence Leu 373 to Ile 393 of the human high molecular weight kininogen. The hydrolysis products from this peptide were isolated by high performance liquid chromatography, and Lys-BK was characterized as the major released kinin by mass spectrometry. Intramolecularly quenched fluorogenic peptides spanning the Met 379-Lys 380 and Arg 389-Ser 390 bradykinin-flanking sequences were then used to assess the substrate specificity requirements of the parasite-derived protease compared with two COOH-terminal truncated recombinant isoforms (cruzain and cruzipain 2). In contrast to the high catalytic efficiency of parasite-derived cruzipain, the recombinant proteinases cleaved the bradykinin-flanking sites at markedly different rates. In addition, we also demonstrated that cruzipain activates plasmatic prekallikrein, which would be a second and indirect way of the parasite protease to release bradykinin.

The intracellular protozoan Trypanosoma cruzi causes Chagas' disease, a chronic illness assoc... more The intracellular protozoan Trypanosoma cruzi causes Chagas' disease, a chronic illness associated with cardiomyopathy and digestive disorders. This pathogen invades mammalian cells by signaling them through multiple transduction pathways. We previously showed that cruzipain, the main cysteine protease of T. cruzi , promotes host cell invasion by activating kinin receptors. Here, we report a cruzipain-mediated invasion route that is not blocked by kinin receptor antagonists. By testing different strains of T. cruzi , we observed a correlation between the level of cruzipain secreted by trypomastigotes and the capacity of the pathogen to invade host cells. Consistent with a role for cruzipain, the cysteine protease inhibitor N -methylpiperazine-urea-Phe-homophenylalanine-vinylsulfone-benzene impaired the invasion of human smooth muscle cells by strains Dm28c and X10/6 but not by the G isolate. Os objectivos deste estudo foi comparar por sexo, prática e frequência de actividade física ao ní... more Os objectivos deste estudo foi comparar por sexo, prática e frequência de actividade física ao nível da depressão global. A amostra deste estudo foi constituída por 183 indivíduos, sendo 38,3% do sexo masculino e 61,7% do sexo feminino. Os resultados indicam que são os homens quem mais pratica, com maior frequência e com sessões mais longas de actividade. Constatou-se a existência de efeitos significativos nas comparações por sexo ao nível da depressão global e na dimensão afectivo-somática, assim como há diferenças significativas entre sexos quanto à prática de actividade física e na depressão. Posto isto, concluímos que os homens praticam mais do que as mulheres, e elas tendem a apresentar valores de depressão superiores. As mulheres apresentam níveis mais elevados de depressão em todas as suas dimensões e de forma significativa na dimensão afectivo-somática.

The aim of this study was to compare the prevalence of conventional CAD risk factors, as well as the plasma aminothiol profile (and its major determinants), between two groups of healthy subjects from Ponta Delgada (in Azores) and Lisbon (in mainland) cities, searching for precocious biomarker(s) of the disease. The study groups consisted of 101 healthy volunteers from Ponta Delgada (PDL) and 121 from Lisbon, aged 20–69 years. No differences in the prevalence of classical CAD risk factors were found between the study groups, except in physical inactivity and related central obesity, which were both higher in PDL men than in those from Lisbon. Also eventual relationships of PAP with conventional risk factors for atherosclerosis were investigated, aiming at the finding of early blood markers of the disease.Design and Methods. This was an observational cross-sectional study, where participants were split into two groups: one with a normal and another with an altered PAP (at least one aminothiol out of its reference concentration range).Results. About 76% of subjects had an altered PAP, mainly due to low glutathione levels (<1.5 μmol/L), mostly associated with normal GGT activity. The act of consumption is a way to structure and give meaning to the world and the relationships ... more The act of consumption is a way to structure and give meaning to the world and the relationships established among people. During the transition from modernity, i.e., from modernity to postmodernity (Rajj, 1998), in a consumer culture (Slater, 1997), the products and their signal value and consumption assume a central role in creating meaning for individuals (Solomon, 1983; Fournier, 1998), thus affecting the differentiation and integration of individuals in the social world. Today consumers are dedicated to the consumption of experiences and feelings about brands, far beyond the functionality of the product. This promotes the consumption value of the bond and connection between consumers' tastes, opinions and preferences. Thus, consumption in social and convivial contexts overlaps isolated and egocentric consumption. Given this new reality, blurring the differences between the materials products makes the concept of brand a key issue in the identification and differentiation. User groups and admirers of a brand that engage in joint activities to achieve collective goals and or to express mutual feelings, experiences and commitments are known as brand communities (Stokburger Sauer, 2010). They consume the symbolic meaning of products and not only the product features, and the image and the emotional connection is more important than functionality. In the social context of consumption of these communities should be studied both their interactions and the direct and indirect impact to organizations. It is intended to investigate and explain the interdependence between the concepts of brand community and brand value, with respect to the motivations for participation in the brand community, its characteristics and organic and ultimately the relationship with the cognitive dimensions of brand equity.

Respiratory syncytial virus (RSV) is responsible for annual respiratory infection outbreaks in in... more Respiratory syncytial virus (RSV) is responsible for annual respiratory infection outbreaks in infants and young children worldwide, frequently causing bronchiolitis and pneumonia. We evaluated clinical and epidemiological features of acute respiratory infections (ARIs) caused by respiratory syncytial virus (RSV) in children less than five years old. Nasopharyngeal aspirate samples from children with ARI symptoms, attended at the 'Hospital das Clínicas'-Federal University of Uberlândia, MG, Brazil, were collected and tested for RSV by the immunofluorescence assay (IFA). Patients' clinical and epidemiological data were also obtained. From April 2000 to June 2003, 317 nasopharyngeal samples were collected from children less than 54 months old. Seventy-six samples (24.0%) were positive for RSV, with 53% (40/76) obtained from male patients. Hospitalization occurred in 50% (38/76) of the cases, with an average period of 10.6 days, in most cases (87%, 33/38) occurring in children less than 12 months of age. Although an association between this age group and the presentation of more severe clinical symptoms was observed, such as bronchiolitis in 51% (27/53) of the patients and pneumonia in 19% (10/53), no patients died. RSV was found from February to August, with the highest incidence in May. Conclusions: RSV is an important agent that causes ARIs; the clinical manifestations varied from mild to severe and patients frequently required hospitalization; RSV mostly affected children less than one year old.
Marmosets are cooperative breeders organized as extended family groups, but breeding is generally... more Marmosets are cooperative breeders organized as extended family groups, but breeding is generally restricted to a single pair. Breeding competition is fierce in female marmosets; males, on the other hand, show low levels of intragroup aggression. We investigated male and female breeding strategies and the resulting reproductive output in 9 wild groups. Reproductive output, tenure of breeding animals, identification of the breeding system, breeding position replacements, migration and infanticide were recorded; also, we recorded grooming and aggression. Replacement of the breeding male or female was observed on nine occasions. On four occasions, the son of the breeding male inherited the breeding post, but we never observed inheritance of a breeding post by a daughter. Mostly, females attained a breeding post by immigrating to a group that had a breeding vacancy. Our results showed that Callithrix jacchus males and females use different strategies to attain a breeding position and maintain it for as long as possible. These strategies prolong the tenure of the breeding position, which is the best way to produce a large number of offspring. This article is part of a Special Issue entitled: Neotropical Behaviour.

In this study, we show that these premises are met in a model of s.c. infection by Trypanosoma cruzi, a protozoan that liberates kinins from kininogens through its major protease, cruzipain. Intensity of B2R-dependent paw edema evoked by trypomastigotes correlated with levels of IL-12 produced by CD11c+ dendritic cells isolated from draining lymph nodes. The IL-12 response induced by endogenously released kinins was vigorously increased in infected mice pretreated with inhibitors of angiotensin converting enzyme (ACE), a kinin-degrading metallopeptidase. Furthermore, these innate stimulatory effects were linked to B2R-dependent up-regulation of IFN-γ production by Ag-specific T cells. Here we demonstrate that the invasion of human primary umbilical vein endothelial cells (HUVECs) or Chinese hamster ovary (CHO) cells overexpressing the B2 type of bradykinin receptor (CHO-B2R) by tissue culture trypomastigotes is subtly modulated by the combined activities of kininogens, kininogenases, and kinin-degrading peptidases. The presence of captopril, an inhibitor of bradykinin degradation by kininase II, drastically potentiated parasitic invasion of HUVECs and CHO-B2R, but not of mock-transfected CHO cells, whereas the B2R antagonist HOE 140 or monoclonal antibody MBK3 to bradykinin blocked these effects. Because kininogen molecules may be displayed on cell surfaces by binding to glycosaminoglycans, we examined whether the ability of cruzipain to release kinins from high molecular weight kininogen (HK) is modulated by heparan sulfate (HS). Kinetic assays show that HS reduces the cysteine proteinase inhibitory activity (K i app) of HK about 10-fold. Conversely, the catalytic efficiency of cruzipain on kinin-related synthetic fluorogenic substrates is enhanced up to 6-fold in the presence of HS. Analysis of the HK breakdown products generated by cruzipain indicated that HS changes the pattern of HK cleavage products. Direct measurements of bradykinin demonstrated an up to 35fold increase in cruzipain-mediated kinin liberation in the presence of HS. Similarly, kinin release by living trypomastigotes increased up to 10-fold in the presence of HS. These studies suggest that the efficiency of T. cruzi to initiate kinin release is potently enhanced by the mutual interactions between cruzipain, HK, and heparan sulfate proteoglycans.

The major isoform of Trypanosoma cruzi cysteinyl proteinase (cruzipain) has generated Lys-bradyki... more The major isoform of Trypanosoma cruzi cysteinyl proteinase (cruzipain) has generated Lys-bradykinin (Lys-BK or kallidin), a proinflammatory peptide, by proteolysis of kininogen. The releasing of this peptide was demonstrated by mass spectrometry, radioimmunoassay, and ileum contractile responses. The kinin-releasing activity was immunoabsorbed selectively by monoclonal antibodies to the characteristic COOH-terminal domain of cruzipain. To determine the hydrolysis steps that account for the kininogenase activity of cruzipain, we synthesized a fluorogenic peptide (o-aminobenzoyl-Leu-Gly-Met-Ile-Ser-Leu-Met-Lys-Arg-Pro-Pro-Gly-Phe-Ser-Pro-Phe-Arg 389-Ser 390-Ser-Arg-Ile-NH 2) based on the sequence Leu 373 to Ile 393 of the human high molecular weight kininogen. The hydrolysis products from this peptide were isolated by high performance liquid chromatography, and Lys-BK was characterized as the major released kinin by mass spectrometry. Intramolecularly quenched fluorogenic peptides spanning the Met 379-Lys 380 and Arg 389-Ser 390 bradykinin-flanking sequences were then used to assess the substrate specificity requirements of the parasite-derived protease compared with two COOH-terminal truncated recombinant isoforms (cruzain and cruzipain 2). In contrast to the high catalytic efficiency of parasite-derived cruzipain, the recombinant proteinases cleaved the bradykinin-flanking sites at markedly different rates. In addition, we also demonstrated that cruzipain activates plasmatic prekallikrein, which would be a second and indirect way of the parasite protease to release bradykinin.

This pathogen invades mammalian cells by signaling them through multiple transduction pathways. We previously showed that cruzipain, the main cysteine protease of T. cruzi , promotes host cell invasion by activating kinin receptors. Here, we report a cruzipain-mediated invasion route that is not blocked by kinin receptor antagonists. By testing different strains of T. cruzi , we observed a correlation between the level of cruzipain secreted by trypomastigotes and the capacity of the pathogen to invade host cells. Consistent with a role for cruzipain, the cysteine protease inhibitor N -methylpiperazine-urea-Phe-homophenylalanine-vinylsulfone-benzene impaired the invasion of human smooth muscle cells by strains Dm28c and X10/6 but not by the G isolate.
