We aim to detect gastric cancer (GC)-specific miRNAs in serum exosomes with diagnostic potential. Methods: A pair of 43 tumor and tumor-adjacent tissue biopsies obtained from GC patients, also 5 mL peripheral blood (following 12h fasting) were collected from the same patients and healthy controls (HCs). QIAGEN miRCURY LNA miRNA Focus PCR Panel applied to screen differentially expressed onco-miRNAs. The candidate miRNAs with the highest fold changes proceeded for validation by qRT-PCR in individuals. Results: We identified that exosomal miR-10a-5p, miR-19b-3p, miR-215-5p, and miR-18a-5p were significantly upregulated in GC patient’s exosomes in contrast to HCs exosomes, Roc curve analysis indicated area under the ROC curve (AUC) of 0.801, 0.721, 0.780 and 0.736 respectively. In a mature organism, tissue homeostasis is regulated by cell division and cell demise as the two... more In a mature organism, tissue homeostasis is regulated by cell division and cell demise as the two major physiological procedures. There is increasing evidence that deregulation of these processes is important in the pathogenicity of main diseases, including myocardial infarction, stroke, atherosclerosis, and inflammatory diseases. Therefore, there are ongoing efforts to discover modulating factors of the cell cycle and cell demise planners aiming at shaping innovative therapeutically modalities to the therapy of such diseases. Although the life of a cell is terminated by several modes of action, a few cell deaths exist-some of which resemble apoptosis and/or necrosis, and most of them are different from one another-that contribute to a wide range of functions to either support or disrupt the homoeostasis. Even in normal physiological conditions, cell life is severe within the cardiovascular system. Cells are persistently undergoing stretch, contraction, injurious metabolic byproducts, and hemodynamic forces, and a few of cells sustain decade-long lifetimes. The duration of vascular disease causes further exposure of vascular cells to a novel range of offences, most of which induce cell death. There is growing evidence on consequences of direct damage to a cell, as well as on responses of adjacent and infiltrating cells, which also have an effect on the pathology. In this study, by focusing on different pathways of cell death in different vascular diseases, an attempt is made to open a new perspective on the therapeutic goals associated with cell death in these diseases.

Observers report that comprehensive sanctions imposed on Iran are a barrier to Iran's coronavirus disease-2019 (COVID-19) crisis and could impede Iran's access to coronavirus vaccines. In this minireview, we discuss COVID-19 and its human rights dimensions of U.S. sanctions on Iran. We believe fighting COVID-19 with limited resources during sanctions will produce a humanitarian crisis, and coronavirus is a convincing reason to lift the sanctions on Iran.

Purpose: Here, we investigated the angiogenic potential of endothelial progenitor cells juxtapose... more Purpose: Here, we investigated the angiogenic potential of endothelial progenitor cells juxtaposed with mesenchymal stem cells (MSCs) inside alginate-gelatin microspheres with stromal derived factor-1α (SDF-1 α) for 7 days. Methods: Six encapsulated groups were allocated including endothelial progenitor cells (EPCs), EPCs/SDF-1α, MSCs, MSCs/SDF-1α, EPCs+MSCs and EPCs+MSCs/SDF-1α. Cells were encapsulated with a mixture of 1% alginate and 2% gelatin hydrogel. Cell survival was examined by MTT assay. Endothelial differentiation was determined by flow cytometry and ELISA. Tubulogenesis assay and Ac-Dil-LDL uptake were used to detect functional activity. Cell migration was analyzed by Transwell insert and gelatin zymography analyses. By using real-time polymerase chain reaction (PCR), we measured the transcription of Akt and PK1. Results: We found an increase in cell viability in MSCs/SDF-1α microspheres compared to EPCs group (P < 0.05). EPC/MSCs co-culture contributed to the increase of CD133 + cells while we found high CD31 levels in MSCs group (P < 0.05). Juxtaposition of EPC with MSCs increased tubulogenesis compared to SDF-1a-free condition (P < 0.001). SDF-1α had the potential to increase in AC-LDL uptake in MSCs and EPCs/MSCs groups. Cells migration and MMP-9 activities increased after treatment with SDF-1α. SDF-1α upregulated PK1 and Akt in encapsulated cells, especially in a co-culture system. Conclusion: Alginate-gelatin microspheres could alter the angiogenic potential of progenitor cells in the presence of SDF-1α

The extremely low-frequency magnetic fields (ELF-EMF) are generated by electrical devices and pow... more The extremely low-frequency magnetic fields (ELF-EMF) are generated by electrical devices and power systems (1 to 300 Hz). In recent decades, exposure to ELF-EMF has emerged potential concerns on public health. Here, we discuss recent progress in the understanding of ELF-EMF biology with a focus on mechanisms of ELF-EMF-mediated disease and summarize the results of more recent experimental and epidemiological studies of ELF-EMF exposure effects on cancer, neurological, cardiovascular, and reproductive disorders. Current views on genomic instability effects, as well as scientific evidence about ELF-EMF therapy, are put forth. According to our literature review, exposure to ELF-EMF has an adverse biological effect depending on the current intensity, strength of the magnetic field, and duration of exposure. Accumulated epidemiologic evidence indicates a correlation between exposure to ELF-EMF and childhood cancer incidence, Alzheimer's disease (AD), and miscarriage. However, adult cancer does not show augmented risk caused by the ELF-EMF. Also, no consistent evidence exists in cardiovascular disease mortality due to ELF-EMF exposure. There is a lack of comprehensive mechanisms for explaining the biological effect of ELF-EMF. Eventually, more studies are needed to clarify the mechanisms of these magnetic fields.

The extent to which Long Interspersed Element-1 (LINE-1, L1) ORF1p is expressed in cutaneous malignancies remains to be evaluated. This study aimed to assess LINE-1 ORF1p immunoreactivity in various skin cancer subtypes. Method:The expression level of LINE-1 ORF1p was evaluated in 95 skin cancer specimens comprising 36 (37.9%) BCC, 28 (29.5%) SCC, and 31 (32.6%) melanoma using the tissue microarray (TMA) technique. Then the association between expression of LINE-1 encoded protein and clinicopathological parameters was analyzed. To this end, we encapsulated human colon adenocarcinoma cell line HT29 with alginate-poly-L-lysine (Alg-PLL) microspheres and the rate of epithelial-mesenchymal transition was monitored. Methods: Cells were randomly divided into three groups; control, alginate and Alg-PLL. To encapsulate cells, we mixed HT-29 cells (1 × 106 ) with 1 mL of 0.05% PLL and 1% Alg mixture and electrosprayed into CaCl2 solution by using a high-voltage power. The proximal BIRC5 promoter possesses specific binding sites for key transcription factors such as nuclear factor κB and signal transducer and activator of transcription 3. Upregulation of survivin exacerbates the autoimmune diseases (AIDs) including multiple sclerosis and myasthenia gravis by reducing the activity threshold of survivin‐specific cytotoxic T cells. DNA damage along with upregulation or downregulation of survivin have been demonstrated in initiation and pathogenesis of cancers and AIDs. However, detailed mechanism of survivin function in pathogenesis of AIDs is not well understood. This review focuses on the structure, specificity, regulation, and function of survivin in physiologic conditions and pathogenesis of AIDs.

Pemphigus is a severe, potentially life-threatening autoimmune blistering mucocutaneous disease w... more Pemphigus is a severe, potentially life-threatening autoimmune blistering mucocutaneous disease which establishes with autoreactive IgG antibodies that target cellular adhesions, precisely extracellular domains of keratinocyte proteins. Several genetic and environmental elements are believed to contribute to the pathogenesis of the disease. The extent to which the initiation and progress of this autoimmune blistering disease may be influenced by the expression of human endogenous retroviruses (HERVs) remains to be elucidated. In this study, we evaluated the expression of HERV groups (K, W, and H) in pemphigus vulgaris (PV) patients in comparison to controls. Peripheral blood samples were collected from 24 PV patients and the corresponding age- and sex-matched healthy controls to extract total RNA for evaluation of HERV-K (HML-2), HERV-W, and HERV-H, env gene expression profile by qPCR. The mRNA expression level of HERV-K, HERV-W, and HERV-H were significantly upregulated in PV patients in comparison to healthy controls (P < 0.0001). The difference in expression of studied HERVs groups between men and women was no significant (P > 0.05). Although rituximab taking patients had a decreased expression level of studied HERVs, the results were not significant (P > 0.05). According to our obtained data, HERVs expression could be measured as a possible diagnostic tool for detection of PV and monitoring of the treatment.

Human endogenous retroviruses (HERVs) are semi-conserved subtypes of long-terminal repeats contai... more Human endogenous retroviruses (HERVs) are semi-conserved subtypes of long-terminal repeats containing retrotransposons that constitute approximately 8% of the genome. Under pathological conditions, the expression of HERVs is also affected by epigenetic modifications. The extent to which the activation of human endogenous retroviruses can be influenced upon exposure to copper remains to be evaluated. Thus, the present study was designed to evaluate the effects of CuSO administration on the transcriptional activity of three HERV families (H, K, and W) in human malignant melanoma cells. For this purpose, following the determination of less cytotoxic concentrations of copper sulfate, the human skin malignant melanoma SK-MEL-37 cells were treated with 25, 50, and 75 μM CuSO for 96 h. Then, mRNA expression of env gene of HERV-H, HERV-K, and HERV-W was evaluated by qPCR. According to the results, 96-h treatment of SK-MEL-37 cells with 75 μM CuSO could significantly downregulate HERV-H evn expression (P &lt; 0.05). Moreover, exposure of 25 μM copper significantly upregulated the expression of HERV-K env (P &lt; 0.05). Regarding HERV-W env, the expression level increased significantly in all treated concentrations (P &lt; 0.05). It seems that the expression change was decreased in both HERV-W and HERV-K by increasing doses. The study results demonstrated that copper exposure to melanoma cells might promote tumor growth by inducing HERVs and/or control tumor development by decreasing the activation of HERVs in defined levels of copper. According to the findings of this study, copper might exert a binary effect on malignant melanoma.

Here, we aimed to investigate the role of alginate-gelatin encapsulation on the dynamic of rat cardiomyoblasts in vitro. MATERIALS AND METHODS Rat cardiomyoblasts cell line H9C2 were enclosed by using alginate-gelatin microspheres and incubated for 7 days. MTT method was used to examine cell viability. The level of genes associated with cardiomyoblasts maturation MYL7, NPPA, NKX2-5, and GATA4 real-time PCR. ELISA was used to measure the protein levels of Bcl-2 and Bax factor post-encapsulation. The level of SOD, GPx, and TAC was detected by biochemical analyses. Western blotting was performed to measure the content of AMP-activated protein kinase. RESULTS We found that encapsulation was able to increase the viability of rat cardiomyocytes after 7 days. The decreased level of Bcl-2 (p < 0.001) coincided with non-significant differences in the level of Bax (p > 0.05). The transcription level of all genes MYL7, NPPA, NKX2-5, and GATA4 were found to down-regulate compared to the control non-treated cells (p < 0.05). No significant differences were found regarding the level of SOD, GPx, and TAC compared to the control (p>0.05). According to western blotting, revealed a reduced level of AMPK following 7-day incubation of rat cardiomyoblasts (p < 0.05). CONCLUSION Data confirmed that the encapsulation of rat cardiomyoblasts with alginate-gelatin microspheres maintained the cells multipotentiality.

Background and Objectives: Mycosis fungoides (MF) is the most common type of cutaneous lymphoma a... more Background and Objectives: Mycosis fungoides (MF) is the most common type of cutaneous lymphoma accompanied by neoplastic T cells. Histopathological changes after phototherapy could be an effective index for evaluation of treatment. The aim of this study was to compare histomorphological changes in Iranian MF patients before and after phototherapy. Methods: Skin biopsies of 20 diagnosed MF patients in Razi dermatology Hospital of Tehran University of Medical Sciences were evaluated based on major and minor diagnostic criteria using Guitart classification to assess histological features of phototherapy. Result: Of 20 studied patients eight patients were man and 12 were woman. Following phototherapy, major histologic criteria including density of infiltrate, epidermotropism and lymphocytic atypia along with some minor histologic criteria such as lymphocytic infiltrate and intraepidermal atypical lymphocytes were significantly alleviated after treatment (p < 0.05). Conclusion: We find a significant decrease in some important histologic parameter of MF after phototherapy, indicating these features may be important diagnostic parameters evaluating therapeutic effect of phototherapy.

Th... more The long interspersed element-1 (LINE-1 or L1) constitutes approximately 17 % of human genome. The expression of these elements is deregulated upon exposure to environmental exposures resulting to genomic instability and cancer promotion. The effect of copper as essential elements in regulation of L1 expression remained to be elucidated. Using non-cytotoxic concentrations of the copper, the expression of endogenous L1 was analyzed by qPCR after 6 days of copper pretreatment in human hepatocellular carcinoma cells (HepG2). The results indicated that the expression of active L1 elements are significantly downregulated at concentrations of 12.5, 25, and 50 μM (p < 0.005). Our data imply that low-level copper exposure may have a protective effect to suppress the induction of L1 activity and decrease incidence of cancerassociated L1 mutagenesis. If this achievement is confirmed by further studies, it can be applied in the long-term goals of cancer prevention.

We aim to detect gastric cancer (GC)-specific miRNAs in serum exosomes with diagnostic potential. Methods: A pair of 43 tumor and tumor-adjacent tissue biopsies obtained from GC patients, also 5 mL peripheral blood (following 12h fasting) were collected from the same patients and healthy controls (HCs). QIAGEN miRCURY LNA miRNA Focus PCR Panel applied to screen differentially expressed onco-miRNAs. The candidate miRNAs with the highest fold changes proceeded for validation by qRT-PCR in individuals. There is increasing evidence that deregulation of these processes is important in the pathogenicity of main diseases, including myocardial infarction, stroke, atherosclerosis, and inflammatory diseases. Therefore, there are ongoing efforts to discover modulating factors of the cell cycle and cell demise planners aiming at shaping innovative therapeutically modalities to the therapy of such diseases. Although the life of a cell is terminated by several modes of action, a few cell deaths exist-some of which resemble apoptosis and/or necrosis, and most of them are different from one another-that contribute to a wide range of functions to either support or disrupt the homoeostasis. Even in normal physiological conditions, cell life is severe within the cardiovascular system. Cells are persistently undergoing stretch, contraction, injurious metabolic byproducts, and hemodynamic forces, and a few of cells sustain decade-long lifetimes. The duration of vascular disease causes further exposure of vascular cells to a novel range of offences, most of which induce cell death. There is growing evidence on consequences of direct damage to a cell, as well as on responses of adjacent and infiltrating cells, which also have an effect on the pathology. In this study, by focusing on different pathways of cell death in different vascular diseases, an attempt is made to open a new perspective on the therapeutic goals associated with cell death in these diseases.

Observers report that comprehensive sanctions imposed on Iran are a barrier to Iran's coronavirus disease-2019 (COVID-19) crisis and could impede Iran's access to coronavirus vaccines. In this minireview, we discuss COVID-19 and its human rights dimensions of U.S. sanctions on Iran. We believe fighting COVID-19 with limited resources during sanctions will produce a humanitarian crisis, and coronavirus is a convincing reason to lift the sanctions on Iran.

Methods: Six encapsulated groups were allocated including endothelial progenitor cells (EPCs), EPCs/SDF-1α, MSCs, MSCs/SDF-1α, EPCs+MSCs and EPCs+MSCs/SDF-1α. Cells were encapsulated with a mixture of 1% alginate and 2% gelatin hydrogel. Cell survival was examined by MTT assay. Endothelial differentiation was determined by flow cytometry and ELISA. Tubulogenesis assay and Ac-Dil-LDL uptake were used to detect functional activity. Cell migration was analyzed by Transwell insert and gelatin zymography analyses. By using real-time polymerase chain reaction (PCR), we measured the transcription of Akt and PK1. Results: We found an increase in cell viability in MSCs/SDF-1α microspheres compared to EPCs group (P < 0.05). EPC/MSCs co-culture contributed to the increase of CD133 + cells while we found high CD31 levels in MSCs group (P < 0.05). Juxtaposition of EPC with MSCs increased tubulogenesis compared to SDF-1a-free condition (P < 0.001). SDF-1α had the potential to increase in AC-LDL uptake in MSCs and EPCs/MSCs groups. Cells migration and MMP-9 activities increased after treatment with SDF-1α. SDF-1α upregulated PK1 and Akt in encapsulated cells, especially in a co-culture system. Conclusion: Alginate-gelatin microspheres could alter the angiogenic potential of progenitor cells in the presence of SDF-1α

The extremely low-frequency magnetic fields (ELF-EMF) are generated by electrical devices and pow... more The extremely low-frequency magnetic fields (ELF-EMF) are generated by electrical devices and power systems (1 to 300 Hz). In recent decades, exposure to ELF-EMF has emerged potential concerns on public health. Here, we discuss recent progress in the understanding of ELF-EMF biology with a focus on mechanisms of ELF-EMF-mediated disease and summarize the results of more recent experimental and epidemiological studies of ELF-EMF exposure effects on cancer, neurological, cardiovascular, and reproductive disorders. Current views on genomic instability effects, as well as scientific evidence about ELF-EMF therapy, are put forth. According to our literature review, exposure to ELF-EMF has an adverse biological effect depending on the current intensity, strength of the magnetic field, and duration of exposure. Accumulated epidemiologic evidence indicates a correlation between exposure to ELF-EMF and childhood cancer incidence, Alzheimer's disease (AD), and miscarriage. However, adult cancer does not show augmented risk caused by the ELF-EMF. Also, no consistent evidence exists in cardiovascular disease mortality due to ELF-EMF exposure. There is a lack of comprehensive mechanisms for explaining the biological effect of ELF-EMF. Eventually, more studies are needed to clarify the mechanisms of these magnetic fields.

The proximal BIRC5 promoter possesses specific binding sites for key transcription factors such as nuclear factor κB and signal transducer and activator of transcription 3. Upregulation of survivin exacerbates the autoimmune diseases (AIDs) including multiple sclerosis and myasthenia gravis by reducing the activity threshold of survivin‐specific cytotoxic T cells. DNA damage along with upregulation or downregulation of survivin have been demonstrated in initiation and pathogenesis of cancers and AIDs. However, detailed mechanism of survivin function in pathogenesis of AIDs is not well understood. This review focuses on the structure, specificity, regulation, and function of survivin in physiologic conditions and pathogenesis of AIDs.

Several genetic and environmental elements are believed to contribute to the pathogenesis of the disease. The extent to which the initiation and progress of this autoimmune blistering disease may be influenced by the expression of human endogenous retroviruses (HERVs) remains to be elucidated. In this study, we evaluated the expression of HERV groups (K, W, and H) in pemphigus vulgaris (PV) patients in comparison to controls. Peripheral blood samples were collected from 24 PV patients and the corresponding age- and sex-matched healthy controls to extract total RNA for evaluation of HERV-K (HML-2), HERV-W, and HERV-H, env gene expression profile by qPCR. The mRNA expression level of HERV-K, HERV-W, and HERV-H were significantly upregulated in PV patients in comparison to healthy controls (P < 0.0001). The difference in expression of studied HERVs groups between men and women was no significant (P > 0.05). Although rituximab taking patients had a decreased expression level of studied HERVs, the results were not significant (P > 0.05). According to our obtained data, HERVs expression could be measured as a possible diagnostic tool for detection of PV and monitoring of the treatment.

Under pathological conditions, the expression of HERVs is also affected by epigenetic modifications. The extent to which the activation of human endogenous retroviruses can be influenced upon exposure to copper remains to be evaluated. Thus, the present study was designed to evaluate the effects of CuSO administration on the transcriptional activity of three HERV families (H, K, and W) in human malignant melanoma cells. For this purpose, following the determination of less cytotoxic concentrations of copper sulfate, the human skin malignant melanoma SK-MEL-37 cells were treated with 25, 50, and 75 μM CuSO for 96 h. Then, mRNA expression of env gene of HERV-H, HERV-K, and HERV-W was evaluated by qPCR. According to the results, 96-h treatment of SK-MEL-37 cells with 75 μM CuSO could significantly downregulate HERV-H evn expression (P &lt; 0.05). Moreover, exposure of 25 μM copper significantly upregulated the expression of HERV-K env (P &lt; 0.05). Regarding HERV-W env, the expression level increased significantly in all treated concentrations (P &lt; 0.05). It seems that the expression change was decreased in both HERV-W and HERV-K by increasing doses. The study results demonstrated that copper exposure to melanoma cells might promote tumor growth by inducing HERVs and/or control tumor development by decreasing the activation of HERVs in defined levels of copper. According to the findings of this study, copper might exert a binary effect on malignant melanoma.

Here, we aimed to investigate the role of alginate-gelatin encapsulation on the dynamic of rat cardiomyoblasts in vitro. MATERIALS AND METHODS Rat cardiomyoblasts cell line H9C2 were enclosed by using alginate-gelatin microspheres and incubated for 7 days. MTT method was used to examine cell viability. The level of genes associated with cardiomyoblasts maturation MYL7, NPPA, NKX2-5, and GATA4 real-time PCR. ELISA was used to measure the protein levels of Bcl-2 and Bax factor post-encapsulation. The level of SOD, GPx, and TAC was detected by biochemical analyses. Western blotting was performed to measure the content of AMP-activated protein kinase. RESULTS We found that encapsulation was able to increase the viability of rat cardiomyocytes after 7 days. The decreased level of Bcl-2 (p < 0.001) coincided with non-significant differences in the level of Bax (p > 0.05). The transcription level of all genes MYL7, NPPA, NKX2-5, and GATA4 were found to down-regulate compared to the control non-treated cells (p < 0.05). No significant differences were found regarding the level of SOD, GPx, and TAC compared to the control (p>0.05). According to western blotting, revealed a reduced level of AMPK following 7-day incubation of rat cardiomyoblasts (p < 0.05). CONCLUSION Data confirmed that the encapsulation of rat cardiomyoblasts with alginate-gelatin microspheres maintained the cells multipotentiality.

Histopathological changes after phototherapy could be an effective index for evaluation of treatment. The aim of this study was to compare histomorphological changes in Iranian MF patients before and after phototherapy. Methods: Skin biopsies of 20 diagnosed MF patients in Razi dermatology Hospital of Tehran University of Medical Sciences were evaluated based on major and minor diagnostic criteria using Guitart classification to assess histological features of phototherapy. Result: Of 20 studied patients eight patients were man and 12 were woman. Following phototherapy, major histologic criteria including density of infiltrate, epidermotropism and lymphocytic atypia along with some minor histologic criteria such as lymphocytic infiltrate and intraepidermal atypical lymphocytes were significantly alleviated after treatment (p < 0.05). Conclusion: We find a significant decrease in some important histologic parameter of MF after phototherapy, indicating these features may be important diagnostic parameters evaluating therapeutic effect of phototherapy.

Th... more The long interspersed element-1 (LINE-1 or L1) constitutes approximately 17 % of human genome. The expression of these elements is deregulated upon exposure to environmental exposures resulting to genomic instability and cancer promotion. The effect of copper as essential elements in regulation of L1 expression remained to be elucidated. Using non-cytotoxic concentrations of the copper, the expression of endogenous L1 was analyzed by qPCR after 6 days of copper pretreatment in human hepatocellular carcinoma cells (HepG2). The results indicated that the expression of active L1 elements are significantly downregulated at concentrations of 12.5, 25, and 50 μM (p < 0.005). Our data imply that low-level copper exposure may have a protective effect to suppress the induction of L1 activity and decrease incidence of cancerassociated L1 mutagenesis. If this achievement is confirmed by further studies, it can be applied in the long-term goals of cancer prevention.
