Advanced Healthcare Materials, 2022

Tuberculosis (TB), including extrapulmonary TB, is responsible for more than one million deaths i... more Tuberculosis (TB), including extrapulmonary TB, is responsible for more than one million deaths in a year worldwide. Existing methods of mycobacteria detection have poor sensitivity, selectivity, and specificity, especially in human tissues. Herein, we present the synthesis of a cholic acid-derived fluorescent probe (P4) that can specifically bind and stain the mycobacterium species. We show that P4 probe specifically binds with mycobacterial lipids, trehalose monomycolate, and phosphatidylinositol mannoside 6. P4 probe detects mycobacteria in polymicrobial planktonic cultures and biofilms with high specificity, selectivity, and sensitivity. Moreover, it can detect a single mycobacterium in the presence of 10000 other bacilli. Unlike the probes that depend on active mycobacterial enzymes, the membrane-specific P4 probe can detect mycobacteria even in formalin-fixed paraffin-embedded (FFPE) mice and human tissue sections. Therefore, the ability of the P4 probe to detect mycobacteria in different biological milieu makes it a potential candidate for diagnostic and prognostic applications in clinical settings. The development of virtual communities like facebook, instagram, whatsapp, twitter, blogs etc. over the internet has increased the interaction among the global consumers. This interaction of consumers is the result of information technology revolution all around the globe from past decades. As of June 2018, 55.1% of the world"s population has internet access (Internet World Stats, 2018). This era of networking over the globe makes the consumers open to take their decisions by analysing and studying the list of available enormous options. In consumer context, Internet has provided social media platform as an interaction tool where consumers can easily interact through virtual communities like facebook, instagram, whatsapp, twitter, blogs etc. Consumer behaviour can easily understand through the interactions on virtual communities (Park and Feinberg, 2010). Virtual communities act as a platform over the internet where consumers attach emotionally and show the connections and belongingness with social media like facebook, instagram, whatsapp, twitter, blogs etc. The consumers start feeling attached with the groups by such sharing of information and interaction among each other and start to follow each other. This sort of behaviour leads to conformity among consumers. Consumers with in a group which are centrally dominant influence other members because their social prominence commands conformity from other members (Chatterjee, P. et al., 2017). Conformity occurs when an individual is influenced by society to alter their attitudes, beliefs, or actions to abide by their existing norms within the group. In online interaction, this conformity behaviour seems to affect the decision of consumers. Such Conformity behaviour of consumers through online virtual communities is termed as e-Consumer Conformity.

Research paper thumbnail of Ionic liquids as potential pretreatment agent for bioconversion of biomass to biofuels

One of the applications is pretreatment of biomass such as lignocellulosic biomass precedent to enzymatic hydrolysis for bioenergy production and other biomaterial production. Generally, the method collected of an IL pretreatment or recovery forwarded by hydrolysis of lignocellulosic biomass. Another approach came in vision in which simultaneously pretreatment process and saccharification of lignocellulosic biomass were used. However, the employment of ILs during this integrated process, within which enzymatic hydrolysis is finished within the presence of IL applied for biomass pretreatment, will simply inactivate the enzymes. Cellulases emerged as one of the important enzymes which are used to catalyze the polysaccharide from biomass and showed a higher level of stability in various ILs. Also, numerous approaches were created together with the synthesis of enzyme-compatible ILs, screening ILs-tolerant enzymes, and media engineering to boost cellulases performance. This review highlights the advancement and use of IL for the pretreatment or hydrolysis of lignocellulosic biomass. The structure of ILs and cellulase stabilization approaches are also discussed. From this review, this tends to powerfully believe that IL-compatible cellulase systems would eliminate the requirement to recover the regenerated biomass and cause simple, saccharification of cellulosic materials, which might be helpful in developing integrated bioprocesses.

However, the structure and origin of photoluminescence in these dots synthesized by the bottom-up approach are still a subject of debate. Here, using a series of separation techniques like solvent extraction, column chromatography, gel electrophoresis and dialysis, we present three distinct fluorescent materials in GQDs synthesized from pyrene, a well-known precursor molecule. The subtle balance between the extent of graphitization and molecular fluorophores determines the nature of fluorescence emission in GQDs verified using a suite of optical and vibrational spectroscopy techniques in combination with electron microscopy. The single-particle level emission properties strongly support our observation. Using cell imaging studies, we also verify that all three materials are suitable for fluorescent staining of biological samples. Our results resolve a long-standing debate o...

To achieve a similar outcome through pharmacological treatment, we improved previously identified inhibitors of B 0 AT1 by medicinal chemistry and identified second generation inhibitors by high through-put screening. Modified diarylmethine compounds inhibited B 0 AT1 with IC 50 values ranging from 8-90 mM. A second generation of inhibitors was derived from high-throughput screening and showed higher affinity (IC 50 of 1-15 mM) and strong selectivity against amino acid transporters with similar substrate specificity, such as ASCT2 (SLC1A5) and LAT1 (SLC7A5). All compounds were unrelated to B 0 AT1 substrates, but were likely to bind in the vicinity of the substrate binding site.
