The discovery and development of anticancer agents were made through long processes from screenin... more The discovery and development of anticancer agents were made through long processes from screening new compounds in various cell lines, animal experimental studies, until clinical trials Majority of cancer chemotherapeutic agents have been initially screened in an in vitro cell-based system and then subsequently studied in an in vivo model using small animals destined for pharmacokinetic properties. Zebrafish (Danio rerio) has become one of the most popular animal model systems used for chemical toxicity tests and drug discovery. Thymoquinone (TQ) is the main constituent of black seed (Nigella sativa, spp) essential oil which shows anti-neoplastic activities in various tumor cells. However, to date, studies on the toxicity of TQ on normal cells or animal model are still insufficient. Here we report that TQ is selective toxic to different human cell lines and show negligible effect on the Zebrafish. The MTT assay demonstrated that TQ induced cytotoxicity on oral cancer cells (HSC-3 and HSC-4) but not on the normal cells (human oral fibroblast and HACAT cells). Moreover, no significant morphological change was observed in the Zebrafish model following TQ exposure at the concentration (0.003 mg/mL), similar to those in the cancer cells. The study suggests that TQ as a cancer chemotherapeutic candidate without apparent adverse effects.

Stres yang terjadi kemungkinan mempengaruhi kualitas hidup menjadi kurang baik dan hal tersebut banyak dialami oleh mahasiswa. Tujuan penelitian untuk menganalisis hubungan tingkatan ngkat dengan kualitas hidup terhadap kejadian SAR. Metode: Jenis penelitian adalah analitik crossectional, dengan populasi terjangkau adalah mahasiswa program studi ngkatan n sarjana ngkatan 2019 dan 2020 sebanyak 161 sampel dengan ngkat Simple Random Sampling. Menggunakan rumus slovin Analisis data univariat disajikan dalam bentuk distribusi frekuensi dan analisis bivariat menggunakan uji chi-square.

Karies gigi adalah suatu proses patologis jaringan keras gigi (email dan dentin) yang terjadi karena adanya interaksi berbagai faktor (multi-faktor) dalam rongga mulut. Karies gigi dimulai dengan terjadinya demineralisasi jaringan keras gigi yang kemudian diikuti dengan terjadinya kerusakan bahan organik gigi. Jaringan keras gigi yang termineralisasi akibat adanya asam hasil fermentasi karbohidrat oleh mikroorganisme, akan menjadi larut dan rusak. Keasaman mulut merupakan hasil metabolisme mikroorganisme pada sukrosa dapat mengakibatkan proses demineralisasi permukaan gigi. Derajat keasaman (pH) lebih rendah dan mencapai derajat keasaman 5,5. Insidensi dan kematian juga telah meningkat selama dekade terakhir (Baykul et.al., 2010; Noguti et.al.,2012; Ding et.al., 2013). Diperkirakan lebih dari 405.000 pasien per tahun menderita kanker mulut di seluruh dunia (Peng et.al., 2011) dan lebih dari 100.000 pasien meninggal per tahun karena kanker mulut (Gasche and Goel, 2012). Pada tahun 2012 ditemukan lebih dari 300.000 kasus baru dan 145.000 kematian (Min et al., 2015). Matrix metalloproteinase-9 (MMP-9) is a noteworthy protease which plays vital roles in many biolo... more Matrix metalloproteinase-9 (MMP-9) is a noteworthy protease which plays vital roles in many biological activities. MMP-9 is usually overexpressed in inflammatory reactions and malignant disorders. The aim of this research was to study the interaction between the four natural coumarins nordentatin, dentatin, clausenidin, and xanthoxyletin with crystallized MMP-9 (PDB: 2OVX) using molecular docking. Molecular docking was done using AutoDock Vina, AutoDock Tools Version 1.5.4, PyMol, and Discovery Studio Biovia 2019. Molecular docking programs showed binding affinities of nordentatin, dentatin, clausenidin and xanthoxyletin with MMP-9 protein. The optimum binding energy (ΔG) of nordentatin, dentatin, clausenidin and xanthoxyletin against the MMP-9 were found to be -7.8, -7.3, -7.6, and -7.5 kcal/mol, respectively. Mercury could be absorbed into the skin, swallowed or inhaled Being accumulatively poisoned can cause nervous system disorder such as Alzheimer, Parkinson, autism or cancer. For this reason, this research is done to analyze the impact of mercury on brain cells specimens. The Experiment is conducted to a number of white mice which are given a treatment of mercury effect periodically and the image of brain cell specimens taken through a digital microscope is processed by means of image processing.

Bcl-2 is a member of Bcl-2 family protein that is crucial for cell survival by supressing apoptosis and its overexpression is frequently detected in many type of cancers. Inactivation of Bcl-2 has been considered as an ideal strategy in anti-cancer therapies. A large number of bioactive compounds derived from natural source including Nordentatin and Quercetin have been reported to possess anticancer activities. Here we report the binding affinities of Nordentatin and Quercetin in silico against anti-apoptotic Bcl-2 protein using molecular docking programs. We demonstrate for the first time that Nordentatin showed optimum binding affinity with Bcl-2, similar to those shown by Quercetin. It can cause human disease including brain disorders. Objective: To identify the effect of mercury vapor inhalation on brain cells and the role of green tea extract (Camellia sinensis) as antioxidant on the brain cells exposed to mercury. Methods: Fourty-eight male Mus musculus were divided into 8 groups, which were given treatment for 3 and 6 weeks. Group A did not receive any treatment and served as a negative control. Group B was a positive control exposed to Mercury. Group C was exposed to Mercury and treated with 26µg/g green tea extract. Group D was exposed to mercury and treated with 52µg/g green tea extract. All animals in the Group B, C, D were exposed to mercury through inhalation for 4 hours daily. The effect of mercury on the brain cells were examined histopathologically. Results: The numbers of necrotic cells counted in the green tea-treated mice group were significantly lower than those untreated group (p<0,05). Conclusion: Mercury vapor inhalation may cause necrosis on brain cells. Administration of green tea extract as an antioxidant reduced the amount of mercury-induced necrotic brain cells in mice. ABSTRAK Pengaruh menghirup merkuri pada sel otak and efek ektrak teh hijau sebagai antioksidan. Merkuri dikenal sebagai metal yang toksik, dapat membentuk radikal bebas yang merangsang terjadinya stres oksidatif yang menyebabkan berbagai kelainan seperti kelainan otak. Tujuan: Penelitian ini bertujuan untuk mengindentifikasi pengaruh dari menghirup merkuri terhadap sel otak serta pengaruh ekstrak teh hijau (Camellia sinensis) sebagai antioksidan terhadap sel otak yang terpapar merkuri. Metode: Sampel penelitian ini adalah 48 Mus musculus jantan yang dibagi menjadi 8 grup yang diberi perlakuan selama 3 dan 6 minggu. Grup A merupakan kontrol negatif tanpa perlakuan. Grup B merupakan kontrol positif yang diberi perlakuan dengan merkuri. Grup C diberikan perlakuan dengan merkuri dan 26µg/g ekstrak teh hijau. Grup D diberi perlakuan merkuri dan 52µg/g ekstrak teh hijau. Seluruh mencit pada grup B, C, dan D diberikan perlakuan merkuri secara inhalasi selama 4 jam/hari. Selanjutnya dilakukan pemeriksaan histopatologi untuk mengevaluasi efek merkuri serta efek antioksidan dari ekstrak teh hijau. Hasil: Jumlah sel otak yang nekrotik lebih sedikit pada kelompok yang diberikan ekstrak teh hijau dibandingkan dengan kelompok yang tidak menerima ekstrak teh hijau. Perbedaan ini bermakna secara statistik (p<0,05). Simpulan: Menghirup merkuri menyebabkan nekrosis sel otak. Pemberian ekstrak teh hijau menurunkan jumlah sel nekrotik pada mencit yang telah terpapar merkuri.

Survivin is a member of the inhibitor of apoptosis (IAP) family and is reportedly overexpressed i... more Survivin is a member of the inhibitor of apoptosis (IAP) family and is reportedly overexpressed in various types of human malignancies. Because the phytochemical compounds dentatin, nordentatin, and quercetin have demonstrated antiproliferative effects in various cancer cell lines, we compared their binding affinities for survivin in silico. Molecular docking analyses were performed using PyMol, Discovery Studio Biovia 2017, AutoDock Vina, and AutoDock Tools version 1.5.4. These computations indicated greater survivin binding affinity of quercetin (∆G −7.0 kcal/mol) than nordentatin and dentatin (∆G −6.5 and −5.5 kcal/mol, respectively), but suggest that all three compounds act as ligand inhibitors of survivin. The present data warrant validation using in vitro and in vivo assays.

It was an experimental research by using 48 male white mice (Mus musculus) as the sample. The sample was divided into 8 groups i.e.: A, B, C, and D, which were given treatment for 3 and 6 weeks. MMP-9 is usually overexpressed in inflammatory reactions and malignant disorders. The aim of this research was to study the interaction between the four natural coumarins nordentatin, dentatin, clausenidin, and xanthoxyletin with crystallized MMP-9 (PDB: 2OVX) using molecular docking. Molecular docking was done using AutoDock Vina, AutoDock Tools Version 1.5.4, PyMol, and Discovery Studio Biovia 2019. Molecular docking programs showed binding affinities of nordentatin, dentatin, clausenidin and xanthoxyletin with MMP-9 protein. The optimum binding energy (ΔG) of nordentatin, dentatin, clausenidin and xanthoxyletin against the MMP-9 were found to be -7.8, -7.3, -7.6, and -7.5 kcal/mol, respectively. In medical field, amalgam which is applied on filling dental cavity is one example of mercury use. Patients, as well as a medic, are highly at risk getting poisoned by mercury. Mercury could be absorbed into the skin, swallowed or inhaled Being accumulatively poisoned can cause nervous system disorder such as Alzheimer, Parkinson, autism or cancer. For this reason, this research is done to analyze the impact of mercury on brain cells specimens. The Experiment is conducted to a number of white mice which are given a treatment of mercury effect periodically and the image of brain cell specimens taken through a digital microscope is processed by means of image processing.

Bcl-2 is a member of Bcl-2 family protein that is crucial for cell survival by supressing apoptosis and its overexpression is frequently detected in many type of cancers. Inactivation of Bcl-2 has been considered as an ideal strategy in anti-cancer therapies. A large number of bioactive compounds derived from natural source including Nordentatin and Quercetin have been reported to possess anticancer activities. Here we report the binding affinities of Nordentatin and Quercetin in silico against anti-apoptotic Bcl-2 protein using molecular docking programs. We demonstrate for the first time that Nordentatin showed optimum binding affinity with Bcl-2, similar to those shown by Quercetin. It can cause human disease including brain disorders. Objective: To identify the effect of mercury vapor inhalation on brain cells and the role of green tea extract (Camellia sinensis) as antioxidant on the brain cells exposed to mercury. Methods: Fourty-eight male Mus musculus were divided into 8 groups, which were given treatment for 3 and 6 weeks. Group A did not receive any treatment and served as a negative control. Group B was a positive control exposed to Mercury. Group C was exposed to Mercury and treated with 26µg/g green tea extract. Group D was exposed to mercury and treated with 52µg/g green tea extract. All animals in the Group B, C, D were exposed to mercury through inhalation for 4 hours daily. The effect of mercury on the brain cells were examined histopathologically. Results: The numbers of necrotic cells counted in the green tea-treated mice group were significantly lower than those untreated group (p<0,05). Conclusion: Mercury vapor inhalation may cause necrosis on brain cells. Administration of green tea extract as an antioxidant reduced the amount of mercury-induced necrotic brain cells in mice. ABSTRAK Pengaruh menghirup merkuri pada sel otak and efek ektrak teh hijau sebagai antioksidan. Merkuri dikenal sebagai metal yang toksik, dapat membentuk radikal bebas yang merangsang terjadinya stres oksidatif yang menyebabkan berbagai kelainan seperti kelainan otak. Tujuan: Penelitian ini bertujuan untuk mengindentifikasi pengaruh dari menghirup merkuri terhadap sel otak serta pengaruh ekstrak teh hijau (Camellia sinensis) sebagai antioksidan terhadap sel otak yang terpapar merkuri. Metode: Sampel penelitian ini adalah 48 Mus musculus jantan yang dibagi menjadi 8 grup yang diberi perlakuan selama 3 dan 6 minggu. Grup A merupakan kontrol negatif tanpa perlakuan. Grup B merupakan kontrol positif yang diberi perlakuan dengan merkuri. Grup C diberikan perlakuan dengan merkuri dan 26µg/g ekstrak teh hijau. Grup D diberi perlakuan merkuri dan 52µg/g ekstrak teh hijau. Seluruh mencit pada grup B, C, dan D diberikan perlakuan merkuri secara inhalasi selama 4 jam/hari. Selanjutnya dilakukan pemeriksaan histopatologi untuk mengevaluasi efek merkuri serta efek antioksidan dari ekstrak teh hijau. Hasil: Jumlah sel otak yang nekrotik lebih sedikit pada kelompok yang diberikan ekstrak teh hijau dibandingkan dengan kelompok yang tidak menerima ekstrak teh hijau. Perbedaan ini bermakna secara statistik (p<0,05). Simpulan: Menghirup merkuri menyebabkan nekrosis sel otak. Pemberian ekstrak teh hijau menurunkan jumlah sel nekrotik pada mencit yang telah terpapar merkuri.

Survivin is a member of the inhibitor of apoptosis (IAP) family and is reportedly overexpressed i... more Survivin is a member of the inhibitor of apoptosis (IAP) family and is reportedly overexpressed in various types of human malignancies. Because the phytochemical compounds dentatin, nordentatin, and quercetin have demonstrated antiproliferative effects in various cancer cell lines, we compared their binding affinities for survivin in silico. Molecular docking analyses were performed using PyMol, Discovery Studio Biovia 2017, AutoDock Vina, and AutoDock Tools version 1.5.4. These computations indicated greater survivin binding affinity of quercetin (∆G −7.0 kcal/mol) than nordentatin and dentatin (∆G −6.5 and −5.5 kcal/mol, respectively), but suggest that all three compounds act as ligand inhibitors of survivin. The present data warrant validation using in vitro and in vivo assays.

It was an experimental research by using 48 male white mice (Mus musculus) as the sample. The sample was divided into 8 groups i.e.: A, B, C, and D, which were given treatment for 3 and 6 weeks. Each group consisted of 6 mice. A group was a negative control which did not get any treatment. Group B as a positive control group was exposed to mercury.
