The purpose of this study was to determine the clinicopathological characteristics of cervical carcinoma with pelvic lymph node metastasis that had undergone radical hysterectomy and lymphadenectomy at Dr. Hasan Sadikin General Hospital, Bandung.Methods: This study used a cross-sectional retrospective descriptive study. Irigasi usus melalui rektum atau washout merupakan suatu prosedur menggunakan sebuah pipa yang dimasukkan melalui anus dan sejumlah cairan dimasukkan melalui pipa tersebut dengan tujuan untuk membersikan usus. NaCl 0,9% sering digunakan pada pasien–pasien penderita penyakit Hirschsprung sebagai cairan washout. Ada berbagai jenis cairan washout yang digunakan, beberapa diantaranya berupa cairan siap pakai yang dapat dibeli di apotek atau toko obat, cairan lainnya dapat dibuat di rumah yaitu larutan garam. Larutan garam ini dianggap sama dengan NaCl 0,9%. Larutan ini selain mudah dibuat dengan biaya yang relatif lebih rendah, tentu saja diharapkan efektivitasnya sama dengan NaCl 0,9%. Objectives: We look for the difference of CTGF expression in urethral stricture due to infection ... more Objectives: We look for the difference of CTGF expression in urethral stricture due to infection and trauma with Immunohistochemistry (IHC) and Enzyme-linked Immunosorbent Assay (ELISA) methods. Methods: We measure the CTGF level from 5 urethral stricture tissue of patient with infection, 18 patients with traumatic urethral stricture. We used IHC and ELISA as method of CTGF measurement. We also compared the result between IHC and ELISA in our subjects. Statistical analysis was done using SPSS version 22.0. Results: From ELISA measurement, the CTGF level of traumatic urethral stricture patients was significantly higher than patients with infection (5.294 ± 7.213 pg/dl vs. 2.62 ± 2.156 pg/dl, p=0.002). From IHC examination CTGF expression in trauma group was significantly stronger than infection group (p=0.041). There was no significant difference between the result of ELISA examination and IHC examination in measuring CTGF level in urethral infection stricture tissue (p=1.000) and trauma stricture tissue (p=0.112). Conclusions: CTGF expression is higher in traumatic urethral stricture patients compared to urethral stricture patients due to infection. There were no significant difference between the result of both ELISA and IHC.

Material Method: Thirty Wistar rats were divided into three groups i.e. group A (sham procedure), group B (unilateral torsion + orchiectomy after 6 hours), and group C (unilateral torsion + orchiectomy after 24 hours). Group B and C were further divided into subgroup with and without Prednisone administration. Prednisone was given orally once a day an hour after torsion, continued till one month later. Orchiectomy of contralateral testis was performed one month later. Those testes were examined by a pathologist. Results: In group A, no abnormality on Sertoli cell quality was found. Objective: To evaluate the role of prostate basal cell staining in diagnosing Gleason score 6 pro... more Objective: To evaluate the role of prostate basal cell staining in diagnosing Gleason score 6 prostate cancer. Materials Methods: During research period, we collected 20 medical records and paraffin block specimens of Gleason score 6 prostatic adenocarcinoma patients. Specimens were taken from prostate needle biopsy. Demographic data and PSA level were extracted from medical records. Basal cells were detected by immunohistochemical staining for antikeratin 34β-E12 on paraffin block specimens analyzed by an experienced pathologist. Positive results suggest a benign lesion. Results: Mean age is 70 ± 6,5 years. Mean prostate volume and PSA level was 52±17cc and 25±21 ng/ml. Three specimens (15%) showed presence of basal cells on antikeratin 34β-E12 staining, which indicated benign lesions. Leucocyturia was found in all patients of this group. INTRODUCTION AND OBJECTIVES: Patients would greatly benefit from accurate and early identificatio... more INTRODUCTION AND OBJECTIVES: Patients would greatly benefit from accurate and early identification of clinically significant prostate cancer (PCa), leading to a reduction of over-diagnosis and over-treatment. The aim of this study was to determine the complementarity between a pre-biopsy urinary biomarker-based risk score (SelectMDx) followed by a post-biopsy epigenetic biomarker assay (ConfirmMDx) to identify men with high-grade PCa. METHODS: The study population consistent of 214 men who were undergoing a 12-core prostate biopsy. Average age was 65 years (range 50-75). Prior to biopsy, urine was collected following digital rectal examination, and urine samples were tested with two urinary RNAbased assays, SelectMDx for Prostate Cancer and PCA3. Biopsy tissue samples from all cores were subsequently tested with the epigenetic DNA assay ConfirmMDx (MDxHealth, Inc, Irvine, CA). This epigenetic test quantifies the relative DNA methylation intensities of 3 genes (GSTP1, RASSF1, and APC) in each individual biopsy core to generate an epigenetic risk score. The results were expressed as areas under the receiver-operating characteristic curve (AUC). High-grade PCa was defined as Gleason Score (GS) >¼7. RESULTS: Of 214 subjects, 95 (44%) had benign pathology at biopsy, 47 (22%) were diagnosed with GS6 cancer and 72 (34%) highgrade disease (GS >¼7). The pre-biopsy urinary tests SelectMDx and PCA3 yielded AUCs of 0.88 and 0.73 respectively, for detection of highgrade cancer at biopsy. The AUC of the post-biopsy ConfirmMDx test was 0.91. A combined model using both SelectMDx and ConfirmMDx showed a further improvement in performance compared to SelectMDx alone (AUC of 0.93). CONCLUSIONS: In this evaluation study, the pre-and postbiopsy RNA and DNA biomarker assays SelectMDx and ConfirmMDx outperformed PCA3 biomarker test. Combining the urinary RNA test with the epigenetic DNA test significantly improved accuracy for the identification of men with high-grade PCa.

Background: Urethral stricture has a high recurrence rate. There is a common doctrine stating tha... more Background: Urethral stricture has a high recurrence rate. There is a common doctrine stating that "once a stricture, always a stricture". This fibrotic disease pathophysiology, pathologically characterized by excessive production, deposition and contraction of extracellular matrix is unknown. Angiotensin II type 1 (AT 1) receptor primarily induces angiogenesis, cellular proliferation and inflammatory responses. AT 1 receptors are also expressed in the fibroblasts of hypertrophic scars, whereas angiotensin II (AngII) regulates DNA synthesis in hypertrophic scar fibroblasts through a negative cross talk between AT 1 and angiotensin II type 2 (AT 2) receptors, which might contribute to the formation and maturation of human hypertrophic scars. Objective: This study was conducted to determine the expression of AT 1 receptors in urethral stricture tissues. Methods: Urethral stricture tissues were collected from patients during anastomotic urethroplasty surgery. There were 24 tissue samples collected in this study with 2 samples of normal urethra for the control group. Immunohistochemistry study was performed to detect the presence of AT 1 receptor expression. Data were analyzed using Mann-Whitney U test, and statistical analysis was performed with SPSS version 20. Results: This study showed that positive staining of AT 1 receptor was found in all urethral stricture tissues (n=24). A total of 8.33% patients had low intensity, 41.67% had moderate intensity and 50% had high intensity of AT 1 receptors, while in the control group, 100% patients had no intensity of AT 1 receptors. Using the Mann-Whitney U test, it was found that urethral stricture tissue had a higher intensity of AT 1 receptors than normal urethral tissue with a p-value = 0.012. Conclusion: The results showed that AT 1 receptor had a higher intensity in the urethral stricture tissue and that AT 1 receptor may play an important role in the development of urethral stricture.

It is commonly seen in South East Asia and found in female around 20-45 years old. The GCT mostly occurs in epiphysis around the knee. The patients normally suffer from pain, swelling, limitation of joint movement, and pathologic fracture. This study aimed to determine the epidemiology of GCT of the bone based on age, sex, location, type, metastases, and recurrence in Dr. Hasan Sadikin General Hospital Bandung. Methods: A descriptive study with retrospective cohort using total sampling method was used to obtain 33 medical records of patients with GCT of bone in Department of Orthopedic & Traumatology and Anatomical Pathology Dr. Hasan Sadikin General Hospital Bandung in the period of January 2010-December 2013. Results: The GCT of bone was mostly found in female of 20 to 29 years old and around 32 years old in male. Distal femur will be the most common site while zygoma, mandibular vertebrae, proximal tibia were rare sites. Twenty nine cases were benign lesion and the rest (4 cases) were malignant. The probability of GCT of bone to metastasize to lungs was very low. Four recurrences occurred within a year. Conclusions: The GCT of bone in male mostly occurs in the third and fourth decade of age. It is found around the knee, mostly benign, rarely metastasize to the lungs and recurrence happens within a year.

Its incidence is 22% among all the musculoskeletal neoplasms. Despite many other studies about patient characteristics of the neoplasm in other regions, descriptive data in Bandung city is still unknown. The objective of this study is to determine the distribution of both benign and malignant neoplasms based on their characteristics of histopathological type, gender, age and anatomical site. Methods: The subjects of this descriptive study were taken from the medical records of the patients who had been examined histopathologically in Anatomical Pathology Department at Dr. Hasan Sadikin General Hospital Bandung, Indonesia within the period of 2008 to2012. The sample was obtained using total sampling technique. Patients diagnosed with cartilage neoplasms were included whereas incomplete medical records were excluded. Histopathological type, gender, age and anatomical site of each patients were collected and analyzed. Results: Seventy cartilage neoplasm cases were found. The distribution of cases comprised of 48 (67%) benign and 23 (33%) malignant. The most common benign neoplasm was osteochondroma and chondroma. Benign neoplasms were more prevalent among men and patients <30 years old, while malignant neoplasm was prevalent among women and patients >60 years old. Femur was the most common site for all neoplasms except for chondroma. Conclusions: PThere are differences in characteristic of benign and malignant cartilage neoplasm patients. Both benign and malignant cartilage neoplasms showed differences on the distribution of patient characteristics. [AMJ.2015;2(4):561-7]

Material & Method: Twenty paraffin block specimens of radical prostatectomy were analyzed looking for Gleason score distribution in each zone. Specimens were stained with Hematoxilin Eosin. Results: Among the 20 cancers, 16 (80%) specimens had more than one focus. Most foci were found in peripheral zone (95%), only one specimen contained foci solely found in transitional zone. More than half (55%) prostate specimens contain three different Gleason grades. Only one specimen contained a single grade. Conclusion: Our results confirm the heterogeneous, multifocal, and multizonal nature of prostate carcinomas. Most specimens had more than one tumor focus, with predominant location in the peripheral zone. Almost all transition zone foci were found concomitantly with peripheral zone foci.

Materials Methods: During research period, we collected 20 medical records and paraffin block specimens of Gleason score 6 prostatic adenocarcinoma patients. Specimens were taken from prostate needle biopsy. Demographic data and PSA level were extracted from medical records. Basal cells were detected by immunohistochemical staining for antikeratin 34β-E12 on paraffin block specimens analyzed by an experienced pathologist. Positive results suggest a benign lesion. Results: Mean age is 70 ± 6,5 years. Mean prostate volume and PSA level was 52±17cc and 25±21 ng/ml. Three specimens (15%) showed presence of basal cells on antikeratin 34β-E12 staining, which indicated benign lesions. Leucocyturia was found in all patients of this group. The aim of this study was to determine the complementarity between a pre-biopsy urinary biomarker-based risk score (SelectMDx) followed by a post-biopsy epigenetic biomarker assay (ConfirmMDx) to identify men with high-grade PCa. METHODS: The study population consistent of 214 men who were undergoing a 12-core prostate biopsy. Average age was 65 years (range 50-75). Prior to biopsy, urine was collected following digital rectal examination, and urine samples were tested with two urinary RNAbased assays, SelectMDx for Prostate Cancer and PCA3. Biopsy tissue samples from all cores were subsequently tested with the epigenetic DNA assay ConfirmMDx (MDxHealth, Inc, Irvine, CA). This epigenetic test quantifies the relative DNA methylation intensities of 3 genes (GSTP1, RASSF1, and APC) in each individual biopsy core to generate an epigenetic risk score. The results were expressed as areas under the receiver-operating characteristic curve (AUC). High-grade PCa was defined as Gleason Score (GS) >¼7. RESULTS: Of 214 subjects, 95 (44%) had benign pathology at biopsy, 47 (22%) were diagnosed with GS6 cancer and 72 (34%) highgrade disease (GS >¼7). The pre-biopsy urinary tests SelectMDx and PCA3 yielded AUCs of 0.88 and 0.73 respectively, for detection of highgrade cancer at biopsy. The AUC of the post-biopsy ConfirmMDx test was 0.91. A combined model using both SelectMDx and ConfirmMDx showed a further improvement in performance compared to SelectMDx alone (AUC of 0.93). CONCLUSIONS: In this evaluation study, the pre-and postbiopsy RNA and DNA biomarker assays SelectMDx and ConfirmMDx outperformed PCA3 biomarker test. Combining the urinary RNA test with the epigenetic DNA test significantly improved accuracy for the identification of men with high-grade PCa.

There is a common doctrine stating that "once a stricture, always a stricture". This fibrotic disease pathophysiology, pathologically characterized by excessive production, deposition and contraction of extracellular matrix is unknown. Angiotensin II type 1 (AT 1) receptor primarily induces angiogenesis, cellular proliferation and inflammatory responses. AT 1 receptors are also expressed in the fibroblasts of hypertrophic scars, whereas angiotensin II (AngII) regulates DNA synthesis in hypertrophic scar fibroblasts through a negative cross talk between AT 1 and angiotensin II type 2 (AT 2) receptors, which might contribute to the formation and maturation of human hypertrophic scars. Objective: This study was conducted to determine the expression of AT 1 receptors in urethral stricture tissues. Methods: Urethral stricture tissues were collected from patients during anastomotic urethroplasty surgery. There were 24 tissue samples collected in this study with 2 samples of normal urethra for the control group. Immunohistochemistry study was performed to detect the presence of AT 1 receptor expression. Data were analyzed using Mann-Whitney U test, and statistical analysis was performed with SPSS version 20. Results: This study showed that positive staining of AT 1 receptor was found in all urethral stricture tissues (n=24). A total of 8.33% patients had low intensity, 41.67% had moderate intensity and 50% had high intensity of AT 1 receptors, while in the control group, 100% patients had no intensity of AT 1 receptors. Using the Mann-Whitney U test, it was found that urethral stricture tissue had a higher intensity of AT 1 receptors than normal urethral tissue with a p-value = 0.012. Conclusion: The results showed that AT 1 receptor had a higher intensity in the urethral stricture tissue and that AT 1 receptor may play an important role in the development of urethral stricture.

It is commonly seen in South East Asia and found in female around 20-45 years old. The GCT mostly occurs in epiphysis around the knee. The patients normally suffer from pain, swelling, limitation of joint movement, and pathologic fracture. This study aimed to determine the epidemiology of GCT of the bone based on age, sex, location, type, metastases, and recurrence in Dr. Hasan Sadikin General Hospital Bandung. Methods: A descriptive study with retrospective cohort using total sampling method was used to obtain 33 medical records of patients with GCT of bone in Department of Orthopedic & Traumatology and Anatomical Pathology Dr. Hasan Sadikin General Hospital Bandung in the period of January 2010-December 2013. Results: The GCT of bone was mostly found in female of 20 to 29 years old and around 32 years old in male. Distal femur will be the most common site while zygoma, mandibular vertebrae, proximal tibia were rare sites. Twenty nine cases were benign lesion and the rest (4 cases) were malignant. The probability of GCT of bone to metastasize to lungs was very low. Four recurrences occurred within a year. Conclusions: The GCT of bone in male mostly occurs in the third and fourth decade of age. It is found around the knee, mostly benign, rarely metastasize to the lungs and recurrence happens within a year.

Its incidence is 22% among all the musculoskeletal neoplasms. Despite many other studies about patient characteristics of the neoplasm in other regions, descriptive data in Bandung city is still unknown. The objective of this study is to determine the distribution of both benign and malignant neoplasms based on their characteristics of histopathological type, gender, age and anatomical site. Methods: The subjects of this descriptive study were taken from the medical records of the patients who had been examined histopathologically in Anatomical Pathology Department at Dr. Hasan Sadikin General Hospital Bandung, Indonesia within the period of 2008 to2012. The sample was obtained using total sampling technique. Patients diagnosed with cartilage neoplasms were included whereas incomplete medical records were excluded. Histopathological type, gender, age and anatomical site of each patients were collected and analyzed. Results: Seventy cartilage neoplasm cases were found. The distribution of cases comprised of 48 (67%) benign and 23 (33%) malignant. The most common benign neoplasm was osteochondroma and chondroma. Benign neoplasms were more prevalent among men and patients <30 years old, while malignant neoplasm was prevalent among women and patients >60 years old. Femur was the most common site for all neoplasms except for chondroma. Conclusions: PThere are differences in characteristic of benign and malignant cartilage neoplasm patients. Both benign and malignant cartilage neoplasms showed differences on the distribution of patient characteristics. [AMJ.2015;2(4):561-7]

Material & Method: Twenty paraffin block specimens of radical prostatectomy were analyzed looking for Gleason score distribution in each zone. Specimens were stained with Hematoxilin Eosin. Results: Among the 20 cancers, 16 (80%) specimens had more than one focus. Most foci were found in peripheral zone (95%), only one specimen contained foci solely found in transitional zone. More than half (55%) prostate specimens contain three different Gleason grades. Only one specimen contained a single grade. Conclusion: Our results confirm the heterogeneous, multifocal, and multizonal nature of prostate carcinomas. Most specimens had more than one tumor focus, with predominant location in the peripheral zone. Almost all transition zone foci were found concomitantly with peripheral zone foci.
