Screening for amylolytic activity and characterization of thermophilic Actinobacteria isolated from a geothermal area in West Java, Indonesia. Biodiversitas 20: 1929-1938. In this study, we describe the screening for amylolytic activity of 17 thermophilic Actinobacteria isolates obtained from the soil of Cisolok geysers, a geothermal area in West Java, Indonesia. All isolates were screened for amylolytic activity by the starch-agar plate method at various temperatures. The results showed that all of isolates were able to grow at 45 o C. The growth abilities of the isolates grown in ISP 1 medium varied at temperatures from 45 to 60 o C. Fifteen of the 17 isolates showed amylolytic activity at 45 o C, 13 showed such activity at 50 o C, and four showed activity at 55 o C. Only three isolates, designated SL1-2-R-2, SL1-2-R-3, and SL1-2-R-4, showed growth and amylolytic activity at 60 o C. These three isolates were selected for molecular identification. The nearly full-length of 16S rRNA gene sequences data showed that these three isolates have a similarity of 99.93-100% with Actinomadura keratinilytica WCC-2265 T and of 98.74-98.91% with A. miaoliensis BC 44T-5 T. Phylogenetic tree shows that all three isolates are clustered together in a monophyletic group with the type strain of A. keratinilytica WCC-2265 T as their most closely related species, with 100% bootstrap support. Based on sequencing of the 16S rRNA gene, phylogenetic comparison, and phenotypic characterization, the three isolates were identified as A. keratinilytica.

Six mycelium-forming actinomycete strains were isolated from forest soil near the Cisolok geysers... more Six mycelium-forming actinomycete strains were isolated from forest soil near the Cisolok geysers in West Java, Indonesia. The 16S rRNA gene sequences of these strains showed high similarity to members of genera in the family Pseudono-cardiaceae with values less than 96.0 %, and most closely related to the genus Thermotunica, T. guangxiensis AG2-7T (94.6–95.2%similarity). The type strain, designated SL3-2-4T, was aerobic, thermophilic, Gram-stain-positive that formed
branched, non-fragmented substrate mycelia and unbranched aerial mycelia with long-chain, oval-shaped spores on International Streptomyces Project (ISP) 3 medium. It produced light-orange substrate mycelia and light-orange diffusible pigments on ISP 3 medium with 2% gellan gum, grown at 30–55 C, with optimum growth at 45 C. The pH range for growth was 4.0–8.0, with optimum growth at pH 7.0. Strain SL3-2-4T was able to hydrolyze casein, esculin, gelatin, guanine, hypoxanthine, starch, L-tyrosine, and xanthine, but not adenine, carboxymethyl-cellulose, cellulose, chitin, Tween 20, or xylan. The major fatty acid was iso-C16 : 0, and the major menaquinone was MK-8 (H4). The detected polar lipids were diphosphatidylglycerol, phosphatidyl-glycerol, phosphatidylethanolamine, phosphatidyl-N-methylethanolamine, unidentified aminophospholipids, unidentified glycolipids, and unidentified phospholipids. The cell wall hydrolysate of SL3-2-4T contained meso-2,4-diaminopimelic acid. The whole cell sugars were arabinose and galactose. The DNA G+C content was 71.6 mol%.
Phenotypic features and phylogenetic data differentiated SL3-2-4T from members of the family Pseudonocardiaceae. Therefore, the strain SL3-2-4T is proposed as a representative of a novel species in a novel genus, Gandjariella thermophila
gen. nov., sp. nov. The type strain is SL3-2-4T (=UICC B-83T=NRRL B-67478T=InaCC A981T).

Isolation and identification of yeasts from a plant (Broussonetia papyrifera Vent., family Morace... more Isolation and identification of yeasts from a plant (Broussonetia papyrifera Vent., family Moraceae) from Trowulan, Indonesia has been carried out. A total of 1.027 yeast isolates were obtained by using direct, washing, and filter membrane methods. Based on similarity of colony morphology, 15 representative yeast isolates were selected. All representative isolates were identified based on the sequence analyses of internal transcribed spacer (ITS) regions of rDNA. Seven representative yeast isolates belong to the phylum Ascomycota, and consist of two classes, i.e. Hemiascomyetes (3 isolates of Candida), and Euascomycetes ( 1 isolate of Dothideaceae, and 3 isolates of Aureobasidium).Eight isolates belong to the phylum Basidiomycota and consist of three classes, i.e. Ustilaginomycetes (1 isolate of Pseudozyma), Urediniomycetes (2 isolates of Rhodotorula), and Hymenomycetes (5 isolates of Cryptococcus).

A total of 245 yeast isolates from Gunung Halimun National Park (GHNP) were screened for cellulol... more A total of 245 yeast isolates from Gunung Halimun National Park (GHNP) were screened for cellulolytic activity using 0.2% cellulose-azure. The results showed that 16 isolates have cellulolytic activity using cellulose-azure assay. These isolates were further screened for carboxymethyl cellulase (CMCase), avicelase and cellobiase using specific substrates (carboxymethyl cellulosa, avicel and cellobiose) with Teather and Wood method. The results showed that 7 isolates have CMCase; 6 isolates have cellobiase; 2 isolates have CMCase and cellobiase; and 1 isolate has CMCase and avicelase and cellobiase activities. Isolate S 4121 has the highest CMCase activity and identified as Trichosporon sporotrichoides (van Oorschot) van Oorschot and de Hoog UICC Y-286.
