Поддержание штаммов в рабочем состоянии, сохранение их ценных свойств являются важными условиями ... more Поддержание штаммов в рабочем состоянии, сохранение их ценных свойств являются важными условиями практически любой работы с микроорганизмами – от первичного изучения до использования их в производстве различных биопрепаратов. В связи с этим в данной статье представлены результаты исследования по освежению штамма, возбудителя одного из таких инфекционных заболеваний, как парагриппа-3 крупного рогатого скота (ПГ-3 КРС) в культуре клеток ПТ и изучение патогенных свойств штамма на естественно восприимчивых животных. В результате освежения вируса в культуре клеток инфекционная активность образцов штамма составила в пределах от 6,87±0,17 до 7,33±0,14 lg ТЦД50/мл. При изучении патогенности освеженного штамма на животных установлено, что у телят, зараженных освеженным вирусом, с 3-х по 9-е сутки наблюдались клинические признаки характерные для ПГ-3 КРС, такие как повышение температуры тела до +41,2°С, потеря аппетита, а также истечение из носа и глаз. We describe the coding-complete genome sequence of a strain of severe acute respiratory syndrome ... more We describe the coding-complete genome sequence of a strain of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) obtained from a patient with symptoms of coronavirus disease 2019 (COVID-19), detected in the Republic of Kazakhstan. According to the Pangolin COVID-19 database, the studied strain, SARS-CoV-2/Human/KAZ/Delta-020/2021, belongs to lineage AY.122 and consists of 29,840 nucleotides.

This article describes the results of a preclinical safety and immunogenicity study of QazCovid-i... more This article describes the results of a preclinical safety and immunogenicity study of QazCovid-in®, the first COVID-19 vaccine developed in Kazakhstan, on BALB/c mice, rats, ferrets, Syrian hamsters and rhesus macaques (Macaca mulatta). The study's safety data suggests that this immunobiological preparation can be technically considered a Class 5 nontoxic vaccine. The series of injections that were made did not produce any adverse effect or any change in the general condition of the model animals' health, while macroscopy and histology studies identified no changes in the internal organs of the BALB/c mice and rats. This study has demonstrated that a double immunization enhances the growth of antibody titers as assessed by the microneutralization assay (MNA) and the enzyme-linked immunosorbent assay (ELISA) in a pre-clinical immunogenicity test on animal models. The best GMT results were assessed in MNA and ELISA 7 days after re-vaccination; however, we noted that GMT antibody results in ELISA were lower than in MNA. A comparative GMT assessment after the first immunization and the re-immunization identified significant differences between model animal groups and a growth of GMT antibodies in all of them; also, differences between the gender groups were statistically significant. Moreover, the most marked MNA immune response to the QazCovid-in® vaccine was seen in the Syrian hamsters, while their SARS-CoV-2-specific antibody activity as assessed with ELISA was the lowest.

Crimean–Congo hemorrhagic fever (CCHF) disease cases are registered annually in endemic regions o... more Crimean–Congo hemorrhagic fever (CCHF) disease cases are registered annually in endemic regions of Kazakhstan. To study the prevalence of various Crimean–Congo hemorrhagic fever virus (CCHFV) genotypes, a total of 694 ticks were collected from southern regions of Kazakhstan in 2021. Hyalomma marginatum (n = 323) (46.5%), Hyalomma anatolicum (n = 138) (19.9%), Hyalomma asiaticum (n = 126) (18.2%), Hyalomma scupense (n = 80) (11.5%) and Ixodes ricinus (n = 27) (3.9%) were collected using the standardized flagging technique from the environment. All the tick samples were analyzed for the presence of CCHFV RNA by RT-PCR. The CCHF-positive samples were found within three Hyalomma asiaticum and one Ixodes ricinus tick sample. For the first time in Kazakhstan, infection of the Ixodes ricinus tick with CCHFV was detected. An epidemic event capable of causing severe primary viral pneumonia in humans began to develop in the central Chinese province of Hubei. The isolated etiological agent was identified as a member of Coronaviridae. Currently, the global pandemic associated with a new coronavirus infection of acute respiratory syndrome type 2 (SARS-CoV-2 - Severe acute respiratory syndrome 2) has become a challenge for humanity. In this work, the replicative capacity and pathogenesis of the SARS-CoV-2 virus in hamsters under different infection methods was evaluated. SARS-CoV-2 virus isolates showed effective replication in hamster lungs, leading to pathological lung lesions upon intranasal infection. When infected with this virus, the clinical manifestations of the disease in hamsters were characterized by decreased body temperature and live weight, moistening and ruffling of hair, and frequent stroking of the nasal mirror. ...

В настоящее время известно о циркуляции среди населения нескольких видов коронавирусов, которые, как правило, вызывают заболевания верх них дыхательных путей и легких со средней тяжестью. К этому списку прибавилась новая коронавирусная инфекция COVID-19 (SARS-CoV-2). Вирус быстро распространился во все страны мира. В последнее время в Казахстане данная инфекция наблюдается с ярко выраженной пневмонией среди населения. Следует отметить, что при работе с живым вирусом мы столкнулись с некоторыми вопросами касающихся биологической и физико-химических свойств вируса, которые остались вне внимания ученых мира. В связи с этим, в этих исследованиях нами были изучены некоторые физико-химические и биологические свойства казахстанского штамма вируса SARS-CoV-2, Проведены исследования по поиску дополнительных биологических моделей, чувствительных к вирусу SARS-CoV-2 и используемых in vitro. Установлено, наиболее чувствительными к вирусу SARS-CoV-2 были культуры клеток обезьян и свиней, в которых вирус активно репродуцировался. Были изучены влияние условий хранения образцов нуклеиновых кислот вируса SARS-CoV-2, а также влияние условий окружающей среды к самому вирусу SARS-CoV-2. Результаты проведенных нами исследований показали, что процесс замораживания при температуре минус 70 о С и размораживания при комнатной температуре (20-22 о С) не оказывает заметного влияния на качественный характер цитопатогенности, но оказывает негативное влияние на сохранность титра вируса. Данные о влиянии химических факторов при работе с возбудителями болезней необходимы для создания условий биологической безопасности для персонала и окружающей среды. В данном исследовании нами изучено влияние формальдегида к вирусу. Полученные результаты показывают, что инактивация вируса Sars-CoV-2 с помощью формалина зависит от концентрации формальдегида, температуры реакции, времени инкубации.

Progress on advanced active and passive photonic components that are required for high-speed opti... more Progress on advanced active and passive photonic components that are required for high-speed optical communications over hollow-core photonic bandgap fiber at wavelengths around 2 μm is described in this paper. Single-frequency lasers capable of operating at 10 Gb/s and covering a wide spectral range are realized. A comparison is made between waveguide and surface normal photodiodes with the latter showing good sensitivity up to 15 Gb/s. Passive waveguides, 90°optical hybrids, and arrayed waveguide grating with 100-GHz channel spacing are demonstrated on a large spot-size waveguide platform. Finally, a strong electro-optic effect using the quantum confined Stark effect in strain-balanced multiple quantum wells is demonstrated and used in a Mach-Zehnder modulator capable of operating at 10 Gb/s.

This study describes the registration of the first cases of lumpy skin disease in July 2016 in th... more This study describes the registration of the first cases of lumpy skin disease in July 2016 in the Republic of Kazakhstan. In the rural district of Makash, Kurmangazinsky district of Atyrau region, 459 cattle fell ill and 34 died (morbidity 12.9% and mortality 0.96%). To determine the cause of the disease, samples were taken from sick and dead animals, as well as from insects and ticks. LSDV DNA was detected by PCR in all samples from dead animals and ticks (Dermacentor marginatus and Hyalomma asiaticum), in 14.29% of samples from horseflies (Tabanus bromius), and in one of the samples from two Stomoxys calcitrans flies. The reproductive LSD virus was isolated from organs of dead cattle and insects in the culture of LT and MDBK cells. The virus accumulated in cell cultures of LT and MDBK at the level of the third passage with titers in the range of 5.5–5.75 log 10 TCID50/cm3. Sequencing of the GPCR gene allowed us to identify this virus as a lumpy skin disease virus.

The invention of Rhizoginin-S was only possible after development of a simple and effective method of preparation PI, U.S. patent (4,977,091). Development of a new culture system without protein supplements from animal sources will generate safer vaccines. The purpose of our work was to investigate the effect of Rhizoginin-S on the growth of mammalian cells in tissue culture. It was established that BHK-21 cell incubated with Rhizoginin-S yielded higher cell numbers than cells incubated with medium containing buffer and serum or with medium containing nutrient medium and serum. The proliferation indexes of BHK-21 cells incubated with Rhizoginin-S, with medium or with medium containing serum were 1.2, 0.8 and 1.0 accordingly. We isolated the SARS-CoV-2 virus from clinical materials from some of these patients. Subsequently, a whole virion inactivated candidate vaccine, QazCovid-in, was developed based on this virus. To develop the vaccine, a virus grown in Vero cell culture was used, which was inactivated with formaldehyde, purified, concentrated, sterilized by filtration, and then adsorbed on aluminum hydroxide gel particles. The formula virus and adjuvant in buffer saline solution were used as the vaccine. The safety and protective effectiveness of the developed vaccine were studied in Syrian hamsters. The results of the studies showed the absolute safety of the candidate vaccine in the Syrian hamsters. Frequency analysis of structural (chromosomal aberrations) and genomic (in aneuploid and polyploid cells) chromosomal changes has demonstrated that chromosome mutation rate of the assayed HSCs is within the limits of the normal spontaneous variability of the human chromosomes in the somatic cells of adults.. Finally, a strong electro-optic effect using the quantum confined Stark effect in strain-balanced multiple quantum wells is demonstrated and used in a Mach-Zehnder modulator capable of operating at 10 Gb/s.

This study describes the registration of the first cases of lumpy skin disease in July 2016 in th... more This study describes the registration of the first cases of lumpy skin disease in July 2016 in the Republic of Kazakhstan. In the rural district of Makash, Kurmangazinsky district of Atyrau region, 459 cattle fell ill and 34 died (morbidity 12.9% and mortality 0.96%). To determine the cause of the disease, samples were taken from sick and dead animals, as well as from insects and ticks. LSDV DNA was detected by PCR in all samples from dead animals and ticks (Dermacentor marginatus and Hyalomma asiaticum), in 14.29% of samples from horseflies (Tabanus bromius), and in one of the samples from two Stomoxys calcitrans flies. The reproductive LSD virus was isolated from organs of dead cattle and insects in the culture of LT and MDBK cells. The virus accumulated in cell cultures of LT and MDBK at the level of the third passage with titers in the range of 5.5–5.75 log 10 TCID50/cm3. Sequencing of the GPCR gene allowed us to identify this virus as a lumpy skin disease virus.

The invention of Rhizoginin-S was only possible after development of a simple and effective method of preparation PI, U.S. patent (4,977,091). Development of a new culture system without protein supplements from animal sources will generate safer vaccines. The purpose of our work was to investigate the effect of Rhizoginin-S on the growth of mammalian cells in tissue culture. It was established that BHK-21 cell incubated with Rhizoginin-S yielded higher cell numbers than cells incubated with medium containing buffer and serum or with medium containing nutrient medium and serum. The proliferation indexes of BHK-21 cells incubated with Rhizoginin-S, with medium or with medium containing serum were 1.2, 0.8 and 1.0 accordingly. We isolated the SARS-CoV-2 virus from clinical materials from some of these patients. Subsequently, a whole virion inactivated candidate vaccine, QazCovid-in, was developed based on this virus. To develop the vaccine, a virus grown in Vero cell culture was used, which was inactivated with formaldehyde, purified, concentrated, sterilized by filtration, and then adsorbed on aluminum hydroxide gel particles. The formula virus and adjuvant in buffer saline solution were used as the vaccine. The safety and protective effectiveness of the developed vaccine were studied in Syrian hamsters. The results of the studies showed the absolute safety of the candidate vaccine in the Syrian hamsters. Frequency analysis of structural (chromosomal aberrations) and genomic (in aneuploid and polyploid cells) chromosomal changes has demonstrated that chromosome mutation rate of the assayed HSCs is within the limits of the normal spontaneous variability of the human chromosomes in the somatic cells of adults..
