This investigation aimed to evaluate the presence of anti-Leptospira antibodies and the occurrence, antimicrobial resistance, and virulence of Salmonella spp., Yersinia enterocolitica, and Listeria monocytogenes in sera and rectal swabs collected from 42 domestic hunting dogs in the Tuscany region (Italy). Regarding Leptospira, 31 out of 42 serum samples (73.8%) were positive and serogroup Pomona was the most detected (71.4%) at titers between 1:100 and 1:400. Four Salmonella isolates (9.52%) were obtained, all belonging to serotype Infantis; two of them showed antimicrobial resistance to streptomycin, while pipB and sopE presence was assessed in all but one isolate. Wild boar (Sus scrofa) are an important Leptospira host and are increasing in population all over Europe. The aim of this investigation was to evaluate Leptospira spp. infection in the reproductive systems of wild boar hunted in two Italian regions: Tuscany and Sardinia. From 231 animals, reproductive system tissue samples (testicles, epididymides, uteri) as well as placentas and fetuses were collected. Bacteriological examination and Real-Time PCR were performed to detect pathogenic Leptospira (lipL32 gene). Leptospires were isolated from the testicles and epididymides of one adult and two subadult wild boar. As concern this important issue, the problem of plasmid-mediated colistin resistance recently emerged. Few studies reported data about colistin resistance and mcr genes in bacteria from wildlife. In this manuscript, 168 Escherichia coli isolated from hunted wild boar were tested; colistin resistance was evaluated by MIC microdilution method, and the presence of mcr-1 and mcr-2 genes was evaluated by PCR. Overall, 27.9% of isolates resulted resistant to colistin, and most of them showed a MIC value > 256 μg/mL. A percentage of 44.6% of tested E. coli scored positive for one or both genes. In details, 13.6% of isolated harbored mcr-1 and mcr-2 in combination; most of them exhibiting the highest MIC values. The effects of veterinary drugs, dietary supplements and non-protein amino acids on the European ... more The effects of veterinary drugs, dietary supplements and non-protein amino acids on the European honey bee (Apis mellifera ligustica Spinola, 1806) ventriculum microbial profile were investigated. Total viable aerobic bacteria, Enterobacteriaceae, staphylococci, Escherichia coli, lactic acid bacteria, Pseudomonas spp., aerobic bacterial endospores and Enterococcus spp. were determined using a culture-based method. Two veterinary drugs (Varromed® and Api-Bioxal®), two commercial dietary supplements (ApiHerb® and ApiGo®) and two non-protein amino acids (GABA and beta-alanine) were administered for one week to honey bee foragers reared in laboratory cages. After one week, E. coli and Staphylococcus spp. were significantly affected by the veterinary drugs (p < 0.001). This animal plays an important role as a reservoir for several pathogens, including three of the most important zoonoses: salmonellosis, yersiniosis and listeriosis. The aim of this investigation was to evaluate the occurrence of antimicrobial-resistant and virulence factor genes of Salmonella spp., Yersinia enterocolitica and Listeria monocytogenes isolated from wild boar in Tuscany (Central Italy). During two consequent hunting seasons (2018/2019 and 2019/2020), rectal swabs, spleens and livers were collected from 287 hunted wild boar to isolate strains. Swine represent typical reservoirs of this bacterium and a frequent source of human infection. Some intrinsic traits make some serovars or strains more virulent than others. Twenty-nine Salmonella spp. isolated from pigs belonging to 16 different serovars were analyzed for gastric acid environment resistance, presence of virulence genes (mgtC, rhuM, pipB, sopB, spvRBC, gipA, sodCI, sopE), antimicrobial resistance and presence of antimicrobial resistance genes (blaTEM, blaPSE-1, aadA1, aadA2, aphA1-lab, strA-strB, tetA, tetB, tetC, tetG, sul1, sul2, sul3). Fresh sausages are not always thoroughly cooked before consumption and can support the growth of ... more Fresh sausages are not always thoroughly cooked before consumption and can support the growth of pathogenic microorganisms, especially when stored at incorrect temperatures. The aim of this study was to verify the growth of Listeria monocytogenes in Italian salsiccia stored at 2 C, 7 C and 12 C for 9 days (t9) with (PCþ) and without (PC-) a commercial protective culture (Lactobacillus sakei and Staphylococcus xylosus). L. monocytogenes PC þ counts were statistically different from PC-, after 7 days (t7) at 7 C and at 12 C. At 2 C, they increased in PC þ by 0.03 and 0.36 log CFU/g vs. 0.25 and 0.91 in PC-, at t7 and t9. Moreover, quality characteristics (total aerobic counts, colour parameters, TBARS values, pH, sensory attributes) were assessed in fresh sausages stored at 7 C. Significant differences were obtained in PC þ samples at t7 for Pseudomonadaceae and Enterobacteriaceae (about 2 log CFU/g). Yeasts and moulds and Brochothrix thermosphacta were also significantly lower in PC þ samples. PC þ samples were more acidic than PC-, with statistically different colour parameters values particularly at the external surface; raw sausages resulted sensorially discernible at t7, whereas PC þ and PCcooked samples did not show any significant sensory difference. The studied protective culture proved to be a useful tool to increase safety and microbiological quality of salsiccia at abusive storage temperature, effectively limiting the growth of L. monocytogenes and Gram negative spoilage microorganisms, with some sensory drawbacks, especially at the end of the shelf life. HIGHLIGHTS The tested culture improves the microbial safety of salsiccia. It reduces the growth of spoilage microorganisms, especially Pseudomonadaceae and Enterobacteriaceae. It mainly influences colour and acidity evolution of the raw product with no sensory drawbacks in the cooked product.

In this investigation, the antibacterial activity of 41 honey samples from different Ukraine regions has been evaluated. For each honey, melissopalynological and physico-chemical analysis were performed in order to determine botanical origin, pH, glucose and fructose contents and free acidity. So, antibacterial activity against Staphylococcusaureus CCM 4223, Listeria monocytogenes ATCC 7644, Salmonella enterica serovar Typhimurium CCM 3807 and Escherichia coli ATCC 25922 was assessed through the determination of MIC (Minimum Inhibitory Concentration) and MBC (Minimum Bactericidal Concentration) values by the microdilutions method. Although this species can act as a reservoir for different pathogens, data about its role as a carrier of pathogenic and antimicrobial-resistant Escherichia coli are still scarce. The aim of this study was to evaluate the occurrence of antimicrobial-resistant and pathogenic Escherichia coli in wild boar in the Tuscany region of Italy. During the hunting season of 2018–2019, E. coli was isolated from 175 of 200 animals and subjected to antimicrobial resistance tests and PCR for detection of resistance and virulence factor genes. The highest resistance rates were against cephalothin (94.3%), amoxicillin–clavulanic acid (87.4%), ampicillin (68.6%), and tetracycline (44.6%). The most detected resistance genes were blaCMY-2 (54.3%), sul1 (38.9%), sul2 (30.9%), and tetG (24.6%). Vet. arhiv 90, 217-224, 2020. aBSTraCT This study aims to evaluate two commercial antibiotics for drying off. Ninety-five Friesian cows and 380 quarters were included. The cows were classified as Control or Subclinical Mastitis based on somatic cell count. The California Mastitis Test and the teat end score were performed. Quarters were randomly treated with Cloxacillin and Ampicillin (TA) or Cephalexin (TB). The effect of the therapy (TA vs. TB) was estimated by X2 analysis based on the Wald test. The preservative and therapeutic actions of TA vs. TB were evaluated by the Kruskall-Wallis test. TB showed a statistically significant therapeutic effect in the control group, that might be related to the pharmacological activity of the two antibiotics. Also, the subclinical mastitis group most commonly presented more quarters affected compared to the control group, leading to worse improvement despite proper therapy. In conclusion, in abrupt drying off, the California mastitis test, teat end score and somatic cell count evaluation, as well as microbial herd data, might represent key concepts for an efficient drying off standard protocol on a dairy farm. In line with the herd bacterial population, both TA and TB may be employed for drying off therapy.

Because of its high availability and its high lactose content, scotta could be used as an additive to improve fermentation characteristics of alfalfa silage. Silage samples were obtained for 2 years from an alfalfa second cut, collecting the forage at three growth stages: mid bud (MB), early flowering (EF), and late flowering (LF). After wilting the forage at 38% dry matter (DM), four scotta doses were added (0, 75, 150, and 300 g/kg fresh forage) and the main chemical and microbiological characteristics were evaluated after 3, 13, and 90 days of ensiling (DOE). The lowest pH (4.3) was recorded in the EF and LF growth stages, after 90 DOE and with the highest scotta dose (300 g/kg fresh forage). We studied the antimicrobial activity of an alcoholic extract of an Italian propolis in sterile skim milk, pasteurized cow’s milk, and cow’s and goat’s whey cheese (ricotta). Following the determination of the minimal inhibitory concentration on Gram+ and Gram- bacteria, the extract was employed at 2 and 5% (P2, P5), using controls with the same ethanol concentrations (E2, E5) and without any addition. In milk trials, Listeria monocytogenes, Staphylococcus aureus, Bacillus cereus, and Pseudomonas fluorescens were tested. P2 and P5 samples registered significant decreases of Gram+ bacteria in skim milk. The same was true for P5 in cows’ milk, but only with S. aureus for P2. In the present work, five different FFPs (brewery spent grains, bread and cookie leftovers, and mixes of brewer’s spent grain or bread with cookies) were employed as feeding substrates. Larvae’s growth performances, chemical composition, microbial loads, and antioxidant status were determined. Chemical compositions of the substrates affected all the tested parameters. Brewery spent grains-fed larvae showed a faster growth period and higher crude protein and carbohydrate contents. The use of cookies as a single substrate or their addition to spent grains or bread increased the lipids contents, while growth was delayed. Microbial loads were partially affected by the fed diet. The cows were classified as Control or Subclinical Mastitis based on somatic cell count. The California Mastitis Test and the teat end score were performed. Quarters were randomly treated with Cloxacillin and Ampicillin (TA) or Cephalexin (TB). The effect of the therapy (TA vs. TB) was estimated by X2 analysis based on the Wald test. The preservative and therapeutic actions of TA vs. TB were evaluated by the Kruskall-Wallis test. TB showed a statistically significant therapeutic effect in the control group, that might be related to the pharmacological activity of the two antibiotics. Also, the subclinical mastitis group most commonly presented more quarters affected compared to the control group, leading to worse improvement despite proper therapy. In conclusion, in abrupt drying off, the California mastitis test, teat end score and somatic cell count evaluation, as well as microbial herd data, might represent key concepts for an efficient drying off standard protocol on a dairy farm. In line with the herd bacterial population, both TA and TB may be employed for drying off therapy.

Because of its high availability and its high lactose content, scotta could be used as an additive to improve fermentation characteristics of alfalfa silage. Silage samples were obtained for 2 years from an alfalfa second cut, collecting the forage at three growth stages: mid bud (MB), early flowering (EF), and late flowering (LF). After wilting the forage at 38% dry matter (DM), four scotta doses were added (0, 75, 150, and 300 g/kg fresh forage) and the main chemical and microbiological characteristics were evaluated after 3, 13, and 90 days of ensiling (DOE). The lowest pH (4.3) was recorded in the EF and LF growth stages, after 90 DOE and with the highest scotta dose (300 g/kg fresh forage). We studied the antimicrobial activity of an alcoholic extract of an Italian propolis in sterile skim milk, pasteurized cow’s milk, and cow’s and goat’s whey cheese (ricotta). Following the determination of the minimal inhibitory concentration on Gram+ and Gram- bacteria, the extract was employed at 2 and 5% (P2, P5), using controls with the same ethanol concentrations (E2, E5) and without any addition. In milk trials, Listeria monocytogenes, Staphylococcus aureus, Bacillus cereus, and Pseudomonas fluorescens were tested. In the present work, five different FFPs (brewery spent grains, bread and cookie leftovers, and mixes of brewer’s spent grain or bread with cookies) were employed as feeding substrates. Larvae’s growth performances, chemical composition, microbial loads, and antioxidant status were determined. Chemical compositions of the substrates affected all the tested parameters. Brewery spent grains-fed larvae showed a faster growth period and higher crude protein and carbohydrate contents.
