Spherical co-assemblies were generated from an equivalent-molar-ratio mixture of amphiphilic poly... more Spherical co-assemblies were generated from an equivalent-molar-ratio mixture of amphiphilic poly(amidoamine) dendrons that were modified with focal carbazole and/or pyrene chromophores from the first to the third generation (DnCz I and DnPy, where n=1, 2, 3 denotes the generation of dendrons) in water at a concentration of 3.0 x 10(-6) M, lower than the individual critical aggregation concentrations (CAC) of either DnCz I or DnPy. UV-vis absorption and fluorescence emission spectra indicated that fluorescence resonance energy transfer (FRET) from carbazole to pyrene units occurred efficiently within the co-assemblies. Further study implied that part of the carbazole and/or pyrene units stacked homogeneously in the co-assemblies via pi-pi stacking and the dendritic branches served as connectors due to the hydrogen-bonding interaction.

Two series of amphiphiles composed of hydrophilic poly(amidoamine) dendrons (from the first to th... more Two series of amphiphiles composed of hydrophilic poly(amidoamine) dendrons (from the first to the third generation) as the shell and hydrophobic aromatic chromophores (3,6-di(maleimidyl)-9-phenyl carbazole and 9-(4′-maleimidyl phenyl)-3-maleimidyl carbazole) as the central scaffold were synthesized. The effect of dendritic architecture on the photophysical properties and the self-assembly behavior of these amphiphiles were studied by UV–vis absorption spectroscopy, fluorescence spectroscopy, and transmission electron microscopy (TEM) measurements. Both the generation of dendritic shell and the location of dendrons at the chromophoric scaffold had great effect on the photophysical properties of these amphiphiles. In addition, different spherical aggregates were formed from these amphiphiles in the aqueous solution at different concentrations. Because of the combined effects of steric hindrance and architecture of dendritic shells, the amphiphiles from G2 dendron with central chromophore self-organized into ordered aggregates more readily than that from G1 and G3. © 2008 Wiley Periodicals, Inc. Amphiphilic poly(amidoamine) (PAMAM) dendrimers consisting of a hydrophilic dendrimer core and hy... more Amphiphilic poly(amidoamine) (PAMAM) dendrimers consisting of a hydrophilic dendrimer core and hydrophobic aromatic dansyl or 1-(naphthalenyl)-2phenyldiazene (NPD) shells have been synthesized. These amphiphilic dendrimers from the zero generation to the third generation self-assemble into vesicular aggregates in water. The self-assembly behavior of these dendrimers strongly depends on their generations. The generation dependence has been further investigated by an exploration of their electrochemical properties. Many adult tissues are maintained by resident stem cells that elevate their proliferation in resp... more Many adult tissues are maintained by resident stem cells that elevate their proliferation in response to injury. The regulatory mechanisms underlying regenerative proliferation are still poorly understood. Here we show that injury induces Hedgehog (Hh) signaling in enteroblasts (EBs) to promote intestinal stem cell (ISC) proliferation in Drosophila melanogaster adult midgut. Elevated Hh signaling by patched (ptc) mutations drove ISC proliferation noncell autonomously. Inhibition of Hh signaling in the ISC lineage compromised injury-induced ISC proliferation but had little if any effect on homeostatic proliferation. Hh signaling acted in EBs to regulate the production of Upd2, which activated the JAK-STAT pathway to promote ISC proliferation. Furthermore, we show that Hh signaling is stimulated by DSS through the JNK pathway and that inhibition of Hh signaling in EBs prevented DSS-stimulated ISC proliferation. Hence, our study uncovers a JNK-Hh-JAK-STAT signaling axis in the regulation of regenerative stem cell proliferation.

This study evaluated the clinical factors associated with Propionibacterium acnes shoulder infect... more This study evaluated the clinical factors associated with Propionibacterium acnes shoulder infection and the standard culture procedures for isolating P. acnes from shoulder specimens by a 7-year retrospective analysis. P. acnes was incriminated as the second most common pathogen in 17 of 80 patients with positive shoulder cultures. All of the 17 patients had prior shoulder implant. The cumulative rates for isolating P. acnes were 1.9%, 1.9%, 41.9%, 96.4%, and 100% at day 1 to day 5 of incubation, respectively. The standard practice of anaerobic culture was able to detect P. acnes from shoulder specimens in patients with a clinical suspicion of infection. The sensitivity and specificity of prolonged incubation remain to be determined.

This experiment used upflow anaerobic sludge blanket (UASB) with brown sugar water as fermentatio... more This experiment used upflow anaerobic sludge blanket (UASB) with brown sugar water as fermentation substrate and sewage sludge as the initiation sludge of the reaction, which was made into granular sludge by adding activated carbon. Emphasis was placed on assessing the hydrogen production performance of the UASB hydrogen bio-production system in the stable ethanol-type fermentation. Under the conditions that the influent COD was 4000 mg x L(-1), the temperature was (35 +/- 1) degrees C and the hydraulic retention time (HRT) was 8 h, a certain amount of NaHCO3 was added to adjust pH from 3.72 to 3.80, 3.85, 3.94 and 4.04, the hydrogen production rate increased correspondingly from the initial 5.5 L x d(-1) to about 7.0, 8.0, 9.5 and 6.0 L x d(-1), which was improved by 27.27%, 45.45%, 72.73% and 9.09%, respectively. The highest hydrogen production rate and gas yield were observed at pH 3.94. The maximal gas yield was 15.83 L x d(-1), which was 1.75 times as high as the initial production. Meanwhile, the hydrogen production efficiency was 58.05%. This study broke through the lower limit pH value 4.0 in fermentative hydrogen production.

To assess the willingness to utilise follow-up eye care services among participants of community ... more To assess the willingness to utilise follow-up eye care services among participants of community vision screenings in rural villages surrounding Chennai. Vision screening participants aged ≥40 years were selected by systematic sampling and were invited to respond to a pretested verbal survey with close-ended questions before undergoing screening. Two hundred and ninety-two people responded. Among the respondents, 50.3% reported experiencing an eye problem, and 53% of these individuals had never had an eye examination. Acceptance rate for eye surgery, medications, and eyeglasses among the respondents was 59.2%, 52.7% and 90.8%, respectively. These acceptances were not associated with sex, age, or employment; medication acceptance was inversely associated with literacy. Surgery acceptance and medication acceptance were associated with area of residence. Presence of another chronic disease was a predictor for surgery acceptance among respondents experiencing eye problems. Maintaining consistent quality of services delivered is crucial for increasing uptake of existing eye care services. Educational interventions may increase eye care service usage by targeting all demographic subgroups of rural populations equally. Additional interventions should be offered to patients without previous exposure to the healthcare system.

To discuss the imaging anatomy about pancreaticobiliary ductal union, occurrence rate of pancreat... more To discuss the imaging anatomy about pancreaticobiliary ductal union, occurrence rate of pancreaticobiliary maljunction (PBM) and associated diseases in a Chinese population by using magnetic resonance cholangiopancreatography (MRCP). Data were collected from 694 patients who underwent MRCP from January 2010 to December 2012. Three hundred and ninety-three patients were male and 301 patients were female. The age range was 16-92 years old and the average age was 51.8 years. The recruitment indication of all cases was patients who had clinical symptoms, such as abdominal pain, jaundice, nausea and vomiting, which thus were clinically suspected as relative pancreaticobiliary diseases. All cases were examined by MRCP using single-shot fast spin-echo sequences. In order to obtain MRCP images, the maximum intensity projection was used. According to the anatomy of pancreaticobiliary ductal union based on our analysis of MRCP images, all cases were classified into normal type and abnormal type according to the position of pancreaticobiliary ductal union. The abnormal type could be further divided into P-B type, B-P type and the duodenum type. By analyzing the incidence of biliary stone and inflammation, pancreatitis, biliary duct tumors and pancreatic tumors between normal and abnormal types, significant differences existed. The abnormal group was more likely to suffer from pancreaticobiliary diseases. Comparing three different types of PBM that were associated with pancreaticobiliary diseases by using Fisher's method, the result showed that there was no significant difference in the incidence of biliary stones, cholecystitis and pancreatic tumors. The incidence of pancreatitis in B-P type and P-B type was higher than that in duodenum type; the incidence of biliary duct tumor in B-P type was higher than that in P-B type; the incidence of biliary duct tumor in duodenum type was lower than that in P-B type. The incidence of congenital choledochus dilatation in normal type and abnormal type was similar, and there was no significant difference between the two types. Types of PBM are closely related to the occurrence of pancreaticobiliary diseases. MRCP has important clinical value in the early diagnosis and preventive treatment of pancreaticobiliary diseases.

The dry shrinkage and plastic shrinkage cracking evaluation test were conducted. As is shown by the results, the compressive strength of HPRPC at 28d are exceed in 60MPa and dry shrinkage at 120d are less than 500με. By contrast, the cracking -resistant performance of HPRPC-7 is superior to that of HPRPC-2.

Duzhong is historically an active ingredient commonly used in hypertensive herbal prescriptions in China. This work investigated the vasodilating effect of Duzhong and its compounds (wogonin 10 μM and oroxylin-A) in the isolated intact rat heart, perfused retrograde according the method of Langendorff and the cytoprotective effect in EA.hy926 cell lines Coronary perfusion pressure was monitored with a pressure transducer connected to a side-arm of the aortic perfusion cannula. Duzhong induced vasorelaxation in a dose dependent manner, on precontracting the vessels with endothelin-1, Duzhong 10 mg/ml, wogonin 10 μM and oroxylin-A 10 μM could significantly lower the perfusion pressure in reference to positive control SNP, Duzhong induced vasodilation was not inhibited by L-NAME (nitric oxide inhibitor), but was significantly inhibited by Tetraethyl ammonium (TEA, a K(+) channel blocker and almost abolished by potassium chloride. The underlying mechanism was carried out in EA.hy926 cell lines. When these cells were treated with H2O2, there was higher expression of NOX-4, TNF-α and COX-2 mRNA. However, wogonin treatment attenuated the mRNA of NOX-4, TNF-α and COX-2. Wogonin also upregulated the mRNA expression of CAT, SOD-1 and GSR in oxidative stress induced by H2O2 EA.hy926 cells. Duzhong and compounds can exert an in vitro relaxation effect of the coronary artery and improve the heart function in Langendorff apparatus. This action appears to be endothelium dependent but not NO mediated. Cell culture findings indicated that wogonin can exert vascular and cellular protection by scavenging Reactive Oxygen Species.

The glycoprotein IIb/IIIa receptor is the final common pathway of platelet aggregation, regardles... more The glycoprotein IIb/IIIa receptor is the final common pathway of platelet aggregation, regardless of the agonist, and thus represents an ideal therapeutic target for blocking thrombus formation. RUC-2 is a novel glycoprotein IIb/IIIa inhibitor of adenosine-5'-diphosphate (ADP)-induced platelet aggregation, importantly which exhibits a unique mode of binding with respect to classical Arg-Gly-Asp (RGD)-based glycoprotein IIb/IIIa antagonists. To identify new chemotypes that inhibit glycoprotein IIb/IIIa-mediated platelet aggregation like RUC-2, we performed a combination of structure-based pharmacophore screening and structure-based virtual screening approach to screen over 7.3 million small molecules based on the RUC-2-glycoprotein IIb/IIIa crystal structure. Three of the eleven hit compounds identified by virtual screening showed promising activity with IC50 values between 16.9 and 90.6μmolL(-1) in a human platelet aggregation assay induced by ADP and thrombin. The binding conformations of these three were analyzed to provide a rationalization of their activity profile. These compounds may serve as potential novel scaffolds for further development of glycoprotein IIb/IIIa antagonists.

MicroRNA-21 (miR-21) was reported as being overexpressed in various human cancerous tissues, but ... more MicroRNA-21 (miR-21) was reported as being overexpressed in various human cancerous tissues, but its expression in cancerous serum was not unanimous in different laboratories. On the base of optimizing experimental design and improving trial protocol, we wanted to know whether the circulating microRNA-21 was dysregulated in the common solid cancers. Using SYBR green real-time quantitative reverse transcription-PCR, we detected the expression of circulating miR-21 in 174 patients with solid cancers and 39 normal control subjects, including breast cancer, esophageal cancer, gastric cancer, colorectal cancer, lung cancer. Furthermore, we analyzed the associations between miR-21 expression and clinical features of patients. miR-21 was significantly overexpressed in human solid cancerous serum relative to normal control (P < 0.001), and its sensitivity and specificity were significantly higher than the currently used tumor markers. High miR-21 expression was not correlated with gender, age, clinical stage, and lymph node metastasis status. Circulating miR-21 could serve as a potential broad-spectrum serum-based biomarker for the detection of some solid cancers.

This study evaluated the performance of direct mecA polymerase chain reaction (PCR) from blood cu... more This study evaluated the performance of direct mecA polymerase chain reaction (PCR) from blood culture bottles growing Gram-positive cocci in clusters and its role in optimization of antibiotic therapy. A total of 266 blood cultures including 121 methicillin-resistant and 122 methicillin-susceptible Staphylococci were tested for mecA. Compared to phenotypic testing, the overall performance of direct mecA PCR was 99% for sensitivity, specificity, positive predictive value, and negative predictive value, respectively. Assessment of antibiotic therapy upon microbiology reporting of direct mecA PCR results from 38 patients prior to (phase I) and 48 patients after implementation of testing and reporting (phase II) showed that the mean time to antibiotic optimization in phase II (0.9 ± 0.9 day) was significantly shorter than that in phase I (2.2 ± 3.2 days) (P < 0.05). Methicillin-susceptible staphylococcal bacteremias had significantly higher frequency of antibiotic adjustment upon direct mecA reporting, compared to methicillin-resistant staphylococcal bacteremias. Our study indicated that direct mecA PCR improved timely antibiotic optimization.
Increasing evidence shows that single nucleotide polymorphisms (SNPs) or mutations in microRNAs (... more Increasing evidence shows that single nucleotide polymorphisms (SNPs) or mutations in microRNAs (miRNAs) sequence may affect the processing and function of miRNAs and participate in the occurrence of diseases. Although many SNPs of miRNAs were found, their functions in the pathological process of nerve cells were only just emerging. In the present study, the effect of the SNP of one neuronal miRNA, miR-124, on miRNA biogenesis and human genetic disease was investigated using in vitro cell line model and Alzheimer's disease (AD) in the Mongolian population. Bioinformatics prediction showed that a common G/C polymorphism designated rs531564 was found in the pri-miR-124 and the G allele changed the formation of a ring-shaped structure in the predicted secondary structure of the pri-miRNA for miR-124-1. Northern blot and real-time PCR analysis showed that the amount of mature miR-124 from the C/G heterozygosity of rs531564 was increased compared with the CC or GG homozygosity of rs531564. The expression of mature miR-124 from GG homozygosity was also higher than that from CC homozygosity. But in an association study of AD patients and controls, neither genotype nor allele distribution difference was found in AD patients compared with controls. Collectively, the present study is the first to evaluate the relationship between miR-124 and AD in the Mongolian population. SNP rs531564 of miR-124 may not represent a risk factor in the development of AD among Mongolian population.

Hepatocellular carcinoma (HCC) is one of the most common malignant tumors. Clinical symptoms attr... more Hepatocellular carcinoma (HCC) is one of the most common malignant tumors. Clinical symptoms attributable to HCC are usually absent, thus often miss the best therapeutic opportunities. Traditional Chinese Medicine (TCM) plays an active role in diagnosis and treatment of HCC. In this paper, we proposed a particle swarm optimization-based hierarchical feature selection (PSOHFS) model to infer potential syndromes for diagnosis of HCC. Firstly, the hierarchical feature representation is developed by a threelayer tree. The clinical symptoms and positive score of patient are leaf nodes and root in the tree, respectively, while each syndrome feature on the middle layer is extracted from a group of symptoms. Secondly, an improved PSO-based algorithm is applied in a new reduced feature space to search an optimal syndrome subset. Based on the result of feature selection, the causal relationships of symptoms and syndromes are inferred via Bayesian networks. In our experiment, 147 symptoms were aggregated into 27 groups and 27 syndrome features were extracted. The proposed approach discovered 24 syndromes which obviously improved the diagnosis accuracy. Finally, the Bayesian approach was applied to represent the causal relationships both at symptom and syndrome levels. The results show that our computational model can facilitate the clinical diagnosis of HCC.

Data analysis in metabolomics is currently a major challenge, particularly when large sample sets... more Data analysis in metabolomics is currently a major challenge, particularly when large sample sets are analyzed. Herein, we present a novel computational platform entitled MetSign for highresolution mass spectrometry-based metabolomics. By converting the instrument raw data into mzXML format as its input data, MetSign provides a suite of bioinformatics tools to perform raw data deconvolution, metabolite putative assignment, peak list alignment, normalization, statistical significance tests, unsupervised pattern recognition, and time course analysis. MetSign uses a modular design and an interactive visual data mining approach to enable efficient extraction of useful patterns from data sets. Analysis steps, designed as containers, are presented with a wizard for the user to follow analyses. Each analysis step might contain multiple analysis procedures and/or methods and serves as a pausing point where users can interact with the system to review the results, to shape the next steps, and to return to previous steps to repeat them with different methods or parameter settings. Analysis of metabolite extract of mouse liver with spiked-in acid standards shows that MetSign outperforms the existing publically available software packages. MetSign has also been successfully applied to investigate the regulation and time course trajectory of metabolites in hepatic liver.

Under the conditions that the influent COD was 4000 mg x L(-1), the temperature was (35 +/- 1) degrees C and the hydraulic retention time (HRT) was 8 h, a certain amount of NaHCO3 was added to adjust pH from 3.72 to 3.80, 3.85, 3.94 and 4.04, the hydrogen production rate increased correspondingly from the initial 5.5 L x d(-1) to about 7.0, 8.0, 9.5 and 6.0 L x d(-1), which was improved by 27.27%, 45.45%, 72.73% and 9.09%, respectively. The highest hydrogen production rate and gas yield were observed at pH 3.94. The maximal gas yield was 15.83 L x d(-1), which was 1.75 times as high as the initial production. Meanwhile, the hydrogen production efficiency was 58.05%. This study broke through the lower limit pH value 4.0 in fermentative hydrogen production.

To assess the willingness to utilise follow-up eye care services among participants of community ... more To assess the willingness to utilise follow-up eye care services among participants of community vision screenings in rural villages surrounding Chennai. Vision screening participants aged ≥40 years were selected by systematic sampling and were invited to respond to a pretested verbal survey with close-ended questions before undergoing screening. Two hundred and ninety-two people responded. Among the respondents, 50.3% reported experiencing an eye problem, and 53% of these individuals had never had an eye examination. Acceptance rate for eye surgery, medications, and eyeglasses among the respondents was 59.2%, 52.7% and 90.8%, respectively. These acceptances were not associated with sex, age, or employment; medication acceptance was inversely associated with literacy. Surgery acceptance and medication acceptance were associated with area of residence. Presence of another chronic disease was a predictor for surgery acceptance among respondents experiencing eye problems. Maintaining consistent quality of services delivered is crucial for increasing uptake of existing eye care services. Educational interventions may increase eye care service usage by targeting all demographic subgroups of rural populations equally. Additional interventions should be offered to patients without previous exposure to the healthcare system.

To discuss the imaging anatomy about pancreaticobiliary ductal union, occurrence rate of pancreat... more To discuss the imaging anatomy about pancreaticobiliary ductal union, occurrence rate of pancreaticobiliary maljunction (PBM) and associated diseases in a Chinese population by using magnetic resonance cholangiopancreatography (MRCP). Data were collected from 694 patients who underwent MRCP from January 2010 to December 2012. Three hundred and ninety-three patients were male and 301 patients were female. The age range was 16-92 years old and the average age was 51.8 years. The recruitment indication of all cases was patients who had clinical symptoms, such as abdominal pain, jaundice, nausea and vomiting, which thus were clinically suspected as relative pancreaticobiliary diseases. All cases were examined by MRCP using single-shot fast spin-echo sequences. In order to obtain MRCP images, the maximum intensity projection was used. According to the anatomy of pancreaticobiliary ductal union based on our analysis of MRCP images, all cases were classified into normal type and abnormal type according to the position of pancreaticobiliary ductal union. The abnormal type could be further divided into P-B type, B-P type and the duodenum type. By analyzing the incidence of biliary stone and inflammation, pancreatitis, biliary duct tumors and pancreatic tumors between normal and abnormal types, significant differences existed. The abnormal group was more likely to suffer from pancreaticobiliary diseases. Comparing three different types of PBM that were associated with pancreaticobiliary diseases by using Fisher's method, the result showed that there was no significant difference in the incidence of biliary stones, cholecystitis and pancreatic tumors. The incidence of pancreatitis in B-P type and P-B type was higher than that in duodenum type; the incidence of biliary duct tumor in B-P type was higher than that in P-B type; the incidence of biliary duct tumor in duodenum type was lower than that in P-B type. The incidence of congenital choledochus dilatation in normal type and abnormal type was similar, and there was no significant difference between the two types. Types of PBM are closely related to the occurrence of pancreaticobiliary diseases. MRCP has important clinical value in the early diagnosis and preventive treatment of pancreaticobiliary diseases.

As is shown by the results, the compressive strength of HPRPC at 28d are exceed in 60MPa and dry shrinkage at 120d are less than 500με. By contrast, the cracking -resistant performance of HPRPC-7 is superior to that of HPRPC-2.

Duzhong is historically an active ingredient commonly used in hypertensive herbal prescriptions in China. This work investigated the vasodilating effect of Duzhong and its compounds (wogonin 10 μM and oroxylin-A) in the isolated intact rat heart, perfused retrograde according the method of Langendorff and the cytoprotective effect in EA.hy926 cell lines Coronary perfusion pressure was monitored with a pressure transducer connected to a side-arm of the aortic perfusion cannula. Duzhong induced vasorelaxation in a dose dependent manner, on precontracting the vessels with endothelin-1, Duzhong 10 mg/ml, wogonin 10 μM and oroxylin-A 10 μM could significantly lower the perfusion pressure in reference to positive control SNP, Duzhong induced vasodilation was not inhibited by L-NAME (nitric oxide inhibitor), but was significantly inhibited by Tetraethyl ammonium (TEA, a K(+) channel blocker and almost abolished by potassium chloride. The underlying mechanism was carried out in EA.hy926 cell lines. When these cells were treated with H2O2, there was higher expression of NOX-4, TNF-α and COX-2 mRNA. However, wogonin treatment attenuated the mRNA of NOX-4, TNF-α and COX-2. Wogonin also upregulated the mRNA expression of CAT, SOD-1 and GSR in oxidative stress induced by H2O2 EA.hy926 cells. Duzhong and compounds can exert an in vitro relaxation effect of the coronary artery and improve the heart function in Langendorff apparatus. This action appears to be endothelium dependent but not NO mediated. Cell culture findings indicated that wogonin can exert vascular and cellular protection by scavenging Reactive Oxygen Species.

The glycoprotein IIb/IIIa receptor is the final common pathway of platelet aggregation, regardles... more The glycoprotein IIb/IIIa receptor is the final common pathway of platelet aggregation, regardless of the agonist, and thus represents an ideal therapeutic target for blocking thrombus formation. RUC-2 is a novel glycoprotein IIb/IIIa inhibitor of adenosine-5'-diphosphate (ADP)-induced platelet aggregation, importantly which exhibits a unique mode of binding with respect to classical Arg-Gly-Asp (RGD)-based glycoprotein IIb/IIIa antagonists. To identify new chemotypes that inhibit glycoprotein IIb/IIIa-mediated platelet aggregation like RUC-2, we performed a combination of structure-based pharmacophore screening and structure-based virtual screening approach to screen over 7.3 million small molecules based on the RUC-2-glycoprotein IIb/IIIa crystal structure. Three of the eleven hit compounds identified by virtual screening showed promising activity with IC50 values between 16.9 and 90.6μmolL(-1) in a human platelet aggregation assay induced by ADP and thrombin. The binding conformations of these three were analyzed to provide a rationalization of their activity profile. These compounds may serve as potential novel scaffolds for further development of glycoprotein IIb/IIIa antagonists.

MicroRNA-21 (miR-21) was reported as being overexpressed in various human cancerous tissues, but ... more MicroRNA-21 (miR-21) was reported as being overexpressed in various human cancerous tissues, but its expression in cancerous serum was not unanimous in different laboratories. On the base of optimizing experimental design and improving trial protocol, we wanted to know whether the circulating microRNA-21 was dysregulated in the common solid cancers. Using SYBR green real-time quantitative reverse transcription-PCR, we detected the expression of circulating miR-21 in 174 patients with solid cancers and 39 normal control subjects, including breast cancer, esophageal cancer, gastric cancer, colorectal cancer, lung cancer. Furthermore, we analyzed the associations between miR-21 expression and clinical features of patients. miR-21 was significantly overexpressed in human solid cancerous serum relative to normal control (P < 0.001), and its sensitivity and specificity were significantly higher than the currently used tumor markers. High miR-21 expression was not correlated with gender, age, clinical stage, and lymph node metastasis status. Circulating miR-21 could serve as a potential broad-spectrum serum-based biomarker for the detection of some solid cancers.

This study evaluated the performance of direct mecA polymerase chain reaction (PCR) from blood cu... more This study evaluated the performance of direct mecA polymerase chain reaction (PCR) from blood culture bottles growing Gram-positive cocci in clusters and its role in optimization of antibiotic therapy. A total of 266 blood cultures including 121 methicillin-resistant and 122 methicillin-susceptible Staphylococci were tested for mecA. Compared to phenotypic testing, the overall performance of direct mecA PCR was 99% for sensitivity, specificity, positive predictive value, and negative predictive value, respectively. Assessment of antibiotic therapy upon microbiology reporting of direct mecA PCR results from 38 patients prior to (phase I) and 48 patients after implementation of testing and reporting (phase II) showed that the mean time to antibiotic optimization in phase II (0.9 ± 0.9 day) was significantly shorter than that in phase I (2.2 ± 3.2 days) (P < 0.05). Methicillin-susceptible staphylococcal bacteremias had significantly higher frequency of antibiotic adjustment upon direct mecA reporting, compared to methicillin-resistant staphylococcal bacteremias. Our study indicated that direct mecA PCR improved timely antibiotic optimization.
Increasing evidence shows that single nucleotide polymorphisms (SNPs) or mutations in microRNAs (... more Increasing evidence shows that single nucleotide polymorphisms (SNPs) or mutations in microRNAs (miRNAs) sequence may affect the processing and function of miRNAs and participate in the occurrence of diseases. Although many SNPs of miRNAs were found, their functions in the pathological process of nerve cells were only just emerging. In the present study, the effect of the SNP of one neuronal miRNA, miR-124, on miRNA biogenesis and human genetic disease was investigated using in vitro cell line model and Alzheimer's disease (AD) in the Mongolian population. Bioinformatics prediction showed that a common G/C polymorphism designated rs531564 was found in the pri-miR-124 and the G allele changed the formation of a ring-shaped structure in the predicted secondary structure of the pri-miRNA for miR-124-1. Northern blot and real-time PCR analysis showed that the amount of mature miR-124 from the C/G heterozygosity of rs531564 was increased compared with the CC or GG homozygosity of rs531564. The expression of mature miR-124 from GG homozygosity was also higher than that from CC homozygosity. But in an association study of AD patients and controls, neither genotype nor allele distribution difference was found in AD patients compared with controls. Collectively, the present study is the first to evaluate the relationship between miR-124 and AD in the Mongolian population. SNP rs531564 of miR-124 may not represent a risk factor in the development of AD among Mongolian population.

Hepatocellular carcinoma (HCC) is one of the most common malignant tumors. Clinical symptoms attr... more Hepatocellular carcinoma (HCC) is one of the most common malignant tumors. Clinical symptoms attributable to HCC are usually absent, thus often miss the best therapeutic opportunities. Traditional Chinese Medicine (TCM) plays an active role in diagnosis and treatment of HCC. In this paper, we proposed a particle swarm optimization-based hierarchical feature selection (PSOHFS) model to infer potential syndromes for diagnosis of HCC. Firstly, the hierarchical feature representation is developed by a threelayer tree. The clinical symptoms and positive score of patient are leaf nodes and root in the tree, respectively, while each syndrome feature on the middle layer is extracted from a group of symptoms. Secondly, an improved PSO-based algorithm is applied in a new reduced feature space to search an optimal syndrome subset. Based on the result of feature selection, the causal relationships of symptoms and syndromes are inferred via Bayesian networks. In our experiment, 147 symptoms were aggregated into 27 groups and 27 syndrome features were extracted. The proposed approach discovered 24 syndromes which obviously improved the diagnosis accuracy. Finally, the Bayesian approach was applied to represent the causal relationships both at symptom and syndrome levels. The results show that our computational model can facilitate the clinical diagnosis of HCC.

Herein, we present a novel computational platform entitled MetSign for highresolution mass spectrometry-based metabolomics. By converting the instrument raw data into mzXML format as its input data, MetSign provides a suite of bioinformatics tools to perform raw data deconvolution, metabolite putative assignment, peak list alignment, normalization, statistical significance tests, unsupervised pattern recognition, and time course analysis. MetSign uses a modular design and an interactive visual data mining approach to enable efficient extraction of useful patterns from data sets. Analysis steps, designed as containers, are presented with a wizard for the user to follow analyses. Each analysis step might contain multiple analysis procedures and/or methods and serves as a pausing point where users can interact with the system to review the results, to shape the next steps, and to return to previous steps to repeat them with different methods or parameter settings. Analysis of metabolite extract of mouse liver with spiked-in acid standards shows that MetSign outperforms the existing publically available software packages. MetSign has also been successfully applied to investigate the regulation and time course trajectory of metabolites in hepatic liver.
