Effects of fertilizer treatments on green and drug herb yields, herb essential nutrients (N, P, K, Ca, Na, Mg, Fe, Zn, Mn, Cu), essential oil yield, essential oil components (menthol, menthone, 1,8 cineole and menthofuran) and non-essential elements (heavy metal) (Pb, Ni, Co, Cr and Cd) were determined in two successive years (2011-2012). The highest green and drug herb yields were obtained from mono ammonium phosphate (MAP) treatments (24,980 kg ha–1 and 3070 kg ha–1) in the first year and from 15:15:15 treatments (16,950 kg ha–1 and 3080 kg ha–1) in the second year. Primary plant nutrients, total essential oil contents (%), and essential oil components were investigated as well. In the conventional system, nutrient inputs included 15.15.15 and monopotassium phosphate (MKP) fertilizers, and in the organic system, nutrient inputs included bone meal + zeolite (BM+Z) and rock phosphate + zeolite (RP+Z). For conventionally grown basil, the highest green herb yield was 1027.5 kg/da, and the highest drug herb yield was 191.1 kg/da. Green herb and drug herb yields were 872.6 kg/da in the MKP plots under conventional farming and 196.3 kg/da in the bone meal + zeolite plots under organic farming. Analysis of primary plant nutrients showed that nitrogen and magnesium contents in the 15.15.15 fertilizer plots and phosphorus, potassium, and calcium contents in the MKP plots were higher. Rock phosphate + zeolite was the forefront application in the organic farming system. Essential oil yields were highest in the 15.15.15 treatment in the conventional farming system and in the control plot in the organic farming system. An environmentally friendly unreported rapid and simple reverse-phase high-performance thin-layer... more An environmentally friendly unreported rapid and simple reverse-phase high-performance thin-layer chromatography (RP-HPTLC) has been designed for the simultaneous determination of bioactive sesquiterpene coumarins feselol and samarcandin in the methanol extract of five Ferula species. The method was developed using glass plates coated with RP-18 silica gel 60 F254S and a green solvent system of ethanol–water mixture (8:2 v/v) as mobile phase. After development, the plates were quantified densitometrically at 254 for feselol and samarcandin. Feselol and samarcandin peaks from methanol extract of five Ferula species were identified by comparing their single band at Rf = 0.43 ± 0.02 and Rf = 0.60 ± 0.01, respectively. Valid linear relationships between the peak areas and concentrations of feselol and samarcandin in the range of 1000–7000 ng/band respectively were obtained. Objectives: The aim of this study is to confirm the aphrodisiac potential of Ferula drudeana Korovin as listed in the Turkish traditional medicine and to isolate the active metabolites using male rats. Materials and Methods: The CHCl3soluble fraction showed promising activity. Chromatographic purification resulted in the isolation of three sesquiterpene coumarins. Isolated compounds structures were determined as feselol (1), samarcandin (2), and 3'-O-acetyl samarcandin (3) based on the physical and spectral characters. Single doses of F. drudeana CHCl3soluble fraction, aqueous fraction (200 mg/kg BW), 1 and 2 (10 mg/kg BW), and sildenafil citrate (10 mg/kg BW) were orally administered to male Wistar albino rats by gavages. Mount latency, mount frequency (MF), intromission latency, intromission frequency (IF), ejaculation latency, and postejaculatory interval (PEI) were studied. In addition, copulatory efficiency and intercopulatory efficiency were calculated. Results: Oral administration of F. drudeana roots extracts, 1 and 2 significantly increased MF and IF. The latencies of mount and intromission were reduced significantly and ELs were prolonged. Treatment with the extracts, 1 and 2 resulted in the reduction of the PEI. Conclusion: The present findings provide experimental evidence that F. drudeana roots, 1 and 2 possess aphrodisiac activities by enhancing the sexual behavior of male rats. H. Davis (Lamiaceae). The dried-in-shade at ambient temperature aerial parts of the plant were hydro-distilled for 3 h, using a modified Clevenger-type apparatus. In vitro antibacterial studies were carried out against eight bacteria strains and Candida albicans. The antimicrobial activity of the essential oil was carried out with the disc diffusion method. (Helichrysum plicatum DC. subsp. plicatum, Helichrysum plicatum DC. subsp. polyphillum (Ledeb) P.H. Davis & Kupicha and Helichrysum plicatum DC. subsp. isauricum Parolly) were investigated for the essential oil chemical compositions. The volatiles were obtained by conventional hydrodistillation of aerial parts and microdistillation of inflorescences. Subsequent gas chromatography (GC-FID) and gas chromatography coupled to mass spectrometry (GC/MS) revealed chemical diversity in compositions of the volatiles analyzed. A total of 199 compounds were identified representing 73.9-98.3% of the volatiles compositions. High abundance of fatty acids and their esters (24.9-70.8%) was detected in the herb volatiles of H. plicatum subsp. polyphyllum and H. plicatum subsp. isauricum. The inflorescences of Helichrysum subspecies were found to be rich in monoterpenes (15.0-93.1%), fatty acids (0.1-36.3%) and sesquiterpenes (1.1-25.5%). Most of the species are known as "Caksır" and roots are used as aphrodisiac in Anatolia. The aim of this study is to investigate the cytotoxic potential of roots of twelve different Ferula L. species distributing in Anatolia on prostate cancer cell line PC-3. Three extracts with different polarity were prepared from grounded roots of plants by ultrasonic solvent extraction. WST assay was performed to evaluate the cytotoxic effects of extracts. High cytotoxic activity was observed in F. elaeochytris, F. drudeana chloroform extracts and F. szowitsiana hexane extract (IC50: 3.21±0.24µg/ml, 5.92±0.53µg/ml, 8.03±0.51µg/ml respectively) whereas methanol extracts of all species had no cytotoxic activity. F. tingitana chloroform extract induced high proliferation in dose dependent manner. A daucane ester "ferutinin" content was evaluated among the extracts by UPLC and it is found in highest concentrations in F. tenuissima and F. halophila hexane extracts (167380 µg/g and 157160 µg/g respectively). Although there is not a direct relationship between ferutinin content and cytotoxicity on PC-3, F. elaeochytris, F. drudeana, F. szowitsiana and F. tenuissima can be good candidates for the further studies for finding lead phytochemical compounds for prostate cancer treatment.

We investigated the effects of a methanolic extract of Helichrysum pamphylicum on mammalian DNA topoisomerase I via in vitro plasmid supercoil relaxation assays. The extracts manifested a considerable inhibition of the enzyme’s activity in a dose-dependent manner. We also performed a HPLC analysis to identify the flavonoid content of the H. pamphylicum extract and tested the identified flavonoids; luteolin, luteolin-4-glucoside, naringenin, helichrysinA and isoquercitrin, on DNA topoisomerase I activity. The measurement of the total antioxidant capacity of the flavonoid standards suggested that the topoisomerase inhibition might be correlated with the antioxidant capacity of the plant.

DNA topoisomerases are essential enzymes that regulate the conformational changes in DNA topology... more DNA topoisomerases are essential enzymes that regulate the conformational changes in DNA topology by catalyzing the concerted breakage and rejoining of DNA strands during normal cellular growth. During the past few years, there has been considerable pharmacological interest in these enzymes because the inhibitors of DNA topoisomerases represent a major class of anticancer drugs. In this study, we investigated the effects of the extracts, prepared from a number of Helichrysum taxa, on mammalian DNA topoisomerase I via in vitro supercoil relaxation assays using plasmid substrate, pBR322. Cytotoxic alkaloid, camptothecin, an inhibitor of eukaryotic topoisomerase I, was used as reference compound throughout the assays. Quantitative comparisons of the supercoiled and relaxed DNA band intensities on agarose gels suggested that the extracts from H. pallasii (Sprengel) Ledeb., H. armenium DC subsp. araxinum (Kirp.) Takht, and H. plicatum DC subsp. plicatum manifested a considerable inhibition on the enzyme in a dosedependent manner. The inhibition of mammalian DNA topoisomerase I by Helichrysum extracts is a significant result because they can be potential sources of anticancer drugs.

It has been used to treat algic, inflammatory and infectious processes. The aim of this study was to evaluate the anti-inflammatory and antinociceptive action of the crude Cipura paludosa ethanolic extract at concentrations ranging between 2.0 and 4.0% in Oil and Water cream formulations for topical use. The physical-chemical stability of the formulations was monitored over a six-month period with the use of accelerated stability tests. In order to evaluate the anti-inflammatory and antinociceptive activities, we used a paw edema test induced by carrageenan and a formalin test, respectively. The paw edema test showed that there was a statistical difference in the control group in relation to the treatments. In the present study, DPPH radical scavenging assay and ferric-reducing antioxidant assay were us... more In the present study, DPPH radical scavenging assay and ferric-reducing antioxidant assay were used to estimate the potential in vitro antioxidant effect of the methanol extracts of Ferula drudeana Korovin (F. drudeana) and Ferula huber-morathii Peşmen (F. huber-morathii). The antidiabetic activity of both extracts was evaluated in streptozotocin (STZ)-induced diabetic rats. Glibenclamide was taken as the standard drug. Both extracts showed considerable antioxidant potential in the DPPH radical scavenging assay and minimum reducing power in ferric-reducing antioxidant assay. Oral administration of F. drudeana (400 mg kgG 1) and F. huber-morathii (200 and 400 mg kgG 1) extracts to diabetic rats produced a marked reduction in Fasting Blood Glucose (FBG) and elevation in insulin levels after 14 and 28 days of treatment. A meaningful reduction in the concentrations of glycosylated hemoglobin (HbA1c), triglycerides (TG), Total Cholesterol (TC), Low Density Lipoprotein (LDL) in plasma and elevations in the activities of superoxide dismutase (SOD), glutathione peroxidase (GPx), catalase (CAT) and glutathione (GSH) in liver and pancreas homogenates were observed in diabetic animals medicated with F. drudeana (200 and 400 mg kgG 1) and F. huber-morathii (400 mg kgG 1) extracts. Levels of alanine transaminase (ALT), aspartate transaminase (AST), alkaline phosphatase (ALP), bilirubin, total protein and High Density Lipoprotein (HDL) in plasma and malondialdehyde (MDA) in liver and pancreas homogenates were recovered significantly in F. drudeana and F. huber-morathii-medicated diabetic rats. Besides, biochemical results were supported by histopathological findings. These findings showed the significant antioxidant and hypoglycemic activities of F. drudeana and F. huber-morathii extracts in diabetic rats.

Bu çalışmada, nanenin (Mentha x piperita L.) verim, temel besin elementleri (N, P, K, Ca, Na, Fe,... more Bu çalışmada, nanenin (Mentha x piperita L.) verim, temel besin elementleri (N, P, K, Ca, Na, Fe, Zn, Mn, Cu ve B), uçucu yağ ve bileşenleri üzerine organik tarım sistemi ile yetiştiriciliğin etkisinin belirlenmesi amaçlanmıştır. Sertifikalı farklı organik gübre ve girdi uygulamalarının etkisinin incelendiği denemede, nanenin besin maddesi içeriği belirlenmiştir. Ayrıca farklı parametreler üzerine etkileri de saptanmıştır. Uygulamalara göre taze ve kuru herba verimi istatistiki olarak farklılaşmış ve kemik unu ile potasyumlu feldispat uygulanan parsellerde daha yüksek bulunmuştur. An environmentally friendly unreported rapid and simple reverse-phase high-performance thin-layer... more An environmentally friendly unreported rapid and simple reverse-phase high-performance thin-layer chromatography (RP-HPTLC) has been designed for the simultaneous determination of bioactive sesquiterpene coumarins feselol and samarcandin in the methanol extract of five Ferula species. The method was developed using glass plates coated with RP-18 silica gel 60 F254S and a green solvent system of ethanol–water mixture (8:2 v/v) as mobile phase. After development, the plates were quantified densitometrically at 254 for feselol and samarcandin. Feselol and samarcandin peaks from methanol extract of five Ferula species were identified by comparing their single band at Rf = 0.43 ± 0.02 and Rf = 0.60 ± 0.01, respectively. Valid linear relationships between the peak areas and concentrations of feselol and samarcandin in the range of 1000–7000 ng/band respectively were obtained. Objectives: The aim of this study is to confirm the aphrodisiac potential of Ferula drudeana Korovin as listed in the Turkish traditional medicine and to isolate the active metabolites using male rats. Materials and Methods: The CHCl3soluble fraction showed promising activity. Chromatographic purification resulted in the isolation of three sesquiterpene coumarins. Isolated compounds structures were determined as feselol (1), samarcandin (2), and 3'-O-acetyl samarcandin (3) based on the physical and spectral characters. Single doses of F. drudeana CHCl3soluble fraction, aqueous fraction (200 mg/kg BW), 1 and 2 (10 mg/kg BW), and sildenafil citrate (10 mg/kg BW) were orally administered to male Wistar albino rats by gavages. Mount latency, mount frequency (MF), intromission latency, intromission frequency (IF), ejaculation latency, and postejaculatory interval (PEI) were studied. In addition, copulatory efficiency and intercopulatory efficiency were calculated. Results: Oral administration of F. drudeana roots extracts, 1 and 2 significantly increased MF and IF. The latencies of mount and intromission were reduced significantly and ELs were prolonged. Treatment with the extracts, 1 and 2 resulted in the reduction of the PEI. The highest aphrodisiac activity in male rats was exhibited by 2. Conclusion: The present findings provide experimental evidence that F. drudeana roots, 1 and 2 possess aphrodisiac activities by enhancing the sexual behavior of male rats. The obtained results supported the traditional claims about the use of Ferula species for male sexual dysfunction.

H. Davis (Lamiaceae). The dried-in-shade at ambient temperature aerial parts of the plant were hydro-distilled for 3 h, using a modified Clevenger-type apparatus. In vitro antibacterial studies were carried out against eight bacteria strains and Candida albicans. The antimicrobial activity of the essential oil was carried out with the disc diffusion method. Davis & Kupicha and Helichrysum plicatum DC. subsp. isauricum Parolly) were investigated for the essential oil chemical compositions. The volatiles were obtained by conventional hydrodistillation of aerial parts and microdistillation of inflorescences. Subsequent gas chromatography (GC-FID) and gas chromatography coupled to mass spectrometry (GC/MS) revealed chemical diversity in compositions of the volatiles analyzed. A total of 199 compounds were identified representing 73.9-98.3% of the volatiles compositions. High abundance of fatty acids and their esters (24.9-70.8%) was detected in the herb volatiles of H. plicatum subsp. polyphyllum and H. plicatum subsp. isauricum. The inflorescences of Helichrysum subspecies were found to be rich in monoterpenes (15.0-93.1%), fatty acids (0.1-36.3%) and sesquiterpenes (1.1-25.5%). Most of the species are known as "Caksır" and roots are used as aphrodisiac in Anatolia. The aim of this study is to investigate the cytotoxic potential of roots of twelve different Ferula L. species distributing in Anatolia on prostate cancer cell line PC-3. Three extracts with different polarity were prepared from grounded roots of plants by ultrasonic solvent extraction. WST assay was performed to evaluate the cytotoxic effects of extracts. High cytotoxic activity was observed in F. elaeochytris, F. drudeana chloroform extracts and F. szowitsiana hexane extract (IC50: 3.21±0.24µg/ml, 5.92±0.53µg/ml, 8.03±0.51µg/ml respectively) whereas methanol extracts of all species had no cytotoxic activity. F. tingitana chloroform extract induced high proliferation in dose dependent manner. A daucane ester "ferutinin" content was evaluated among the extracts by UPLC and it is found in highest concentrations in F. tenuissima and F. halophila hexane extracts (167380 µg/g and 157160 µg/g respectively). Although there is not a direct relationship between ferutinin content and cytotoxicity on PC-3, F. elaeochytris, F. drudeana, F. szowitsiana and F. tenuissima can be good candidates for the further studies for finding lead phytochemical compounds for prostate cancer treatment.

We investigated the effects of a methanolic extract of Helichrysum pamphylicum on mammalian DNA topoisomerase I via in vitro plasmid supercoil relaxation assays. The extracts manifested a considerable inhibition of the enzyme’s activity in a dose-dependent manner. We also performed a HPLC analysis to identify the flavonoid content of the H. pamphylicum extract and tested the identified flavonoids; luteolin, luteolin-4-glucoside, naringenin, helichrysinA and isoquercitrin, on DNA topoisomerase I activity. The measurement of the total antioxidant capacity of the flavonoid standards suggested that the topoisomerase inhibition might be correlated with the antioxidant capacity of the plant.

During the past few years, there has been considerable pharmacological interest in these enzymes because the inhibitors of DNA topoisomerases represent a major class of anticancer drugs. In this study, we investigated the effects of the extracts, prepared from a number of Helichrysum taxa, on mammalian DNA topoisomerase I via in vitro supercoil relaxation assays using plasmid substrate, pBR322. Cytotoxic alkaloid, camptothecin, an inhibitor of eukaryotic topoisomerase I, was used as reference compound throughout the assays. Quantitative comparisons of the supercoiled and relaxed DNA band intensities on agarose gels suggested that the extracts from H. pallasii (Sprengel) Ledeb., H. armenium DC subsp. araxinum (Kirp.) Takht, and H. plicatum DC subsp. plicatum manifested a considerable inhibition on the enzyme in a dosedependent manner. The inhibition of mammalian DNA topoisomerase I by Helichrysum extracts is a significant result because they can be potential sources of anticancer drugs.

It has been used to treat algic, inflammatory and infectious processes. The aim of this study was to evaluate the anti-inflammatory and antinociceptive action of the crude Cipura paludosa ethanolic extract at concentrations ranging between 2.0 and 4.0% in Oil and Water cream formulations for topical use. The physical-chemical stability of the formulations was monitored over a six-month period with the use of accelerated stability tests. In order to evaluate the anti-inflammatory and antinociceptive activities, we used a paw edema test induced by carrageenan and a formalin test, respectively. The paw edema test showed that there was a statistical difference in the control group in relation to the treatments. In the present study, DPPH radical scavenging assay and ferric-reducing antioxidant assay were us... more In the present study, DPPH radical scavenging assay and ferric-reducing antioxidant assay were used to estimate the potential in vitro antioxidant effect of the methanol extracts of Ferula drudeana Korovin (F. drudeana) and Ferula huber-morathii Peşmen (F. huber-morathii). The antidiabetic activity of both extracts was evaluated in streptozotocin (STZ)-induced diabetic rats. Glibenclamide was taken as the standard drug. Both extracts showed considerable antioxidant potential in the DPPH radical scavenging assay and minimum reducing power in ferric-reducing antioxidant assay. Oral administration of F. drudeana (400 mg kgG 1) and F. huber-morathii (200 and 400 mg kgG 1) extracts to diabetic rats produced a marked reduction in Fasting Blood Glucose (FBG) and elevation in insulin levels after 14 and 28 days of treatment. A meaningful reduction in the concentrations of glycosylated hemoglobin (HbA1c), triglycerides (TG), Total Cholesterol (TC), Low Density Lipoprotein (LDL) in plasma and elevations in the activities of superoxide dismutase (SOD), glutathione peroxidase (GPx), catalase (CAT) and glutathione (GSH) in liver and pancreas homogenates were observed in diabetic animals medicated with F. drudeana (200 and 400 mg kgG 1) and F. huber-morathii (400 mg kgG 1) extracts. Levels of alanine transaminase (ALT), aspartate transaminase (AST), alkaline phosphatase (ALP), bilirubin, total protein and High Density Lipoprotein (HDL) in plasma and malondialdehyde (MDA) in liver and pancreas homogenates were recovered significantly in F. drudeana and F. huber-morathii-medicated diabetic rats. Besides, biochemical results were supported by histopathological findings. These findings showed the significant antioxidant and hypoglycemic activities of F. drudeana and F. huber-morathii extracts in diabetic rats.
