Saccharothrix algeriensis DSM 44581 T , est une actinobacterie productrice de plusieurs antibioti... more Saccharothrix algeriensis DSM 44581 T , est une actinobacterie productrice de plusieurs antibiotiques: thiolutine, tiglyl-pyrrothine, senecioyl-pyrrothine, butyryl-pyrrothine et isobutyryl-pyrrothine, appartiennent au groupe des pyrrothines connu pour ses activites biologiques interessantes. Les pyrrothines ont une structure particuliere formee d’heterocycles sur lesquels se greffe un acyl-CoA (issue d’un acide organique). Dans cette etude, plusieurs acides organiques ont ete additionnes au milieu semi-synthetique, pour eventuellement augmenter le niveau de production des cinq antibiotiques deja caracterises, et probablement pour former des nouveaux derives avec de nouvelles proprietes chimiques et/ou biologiques. Apres ensemencement du milieu avec l’actinobacterie, les fioles Erlenmeyers ont ete incubees dans un shaker. Les antibiotiques sont analyses avec un HPLC muni d’un detecteur a barrettes de diode. An actinobacterial strain named LG10 was isolated from a Saharan Atlas soil (Laghouat, Algeria). ... more An actinobacterial strain named LG10 was isolated from a Saharan Atlas soil (Laghouat, Algeria). The aerial hyphae were yellowish-white on all culture media with rectiflexibiles spore chains, suggested that this bacterium attached to Streptomyces. Furthermore, LG10 contained chemical characteristics that were diagnostic for the genus Streptomyces, such as the presence of LL-diaminopimelic acid isomer (LL-DAP) and glycine amino acid. The hydrolysates of whole-cell included non-characteristic sugars. Comparative analysis of the 16S rDNA sequence displayed a similarity level of 100% with Streptomyces puniceus NRRL ISP-5058T. The antimicrobial activity of the LG10 strain was better in the culture medium MB5. Streptomyces strains are good sources of bioactive compounds with multiple biological activities.

AC170, isolated from Algerian Saharan soil, with strong antifungal activity against pathogenic and toxinogenic fungi, was selected for further studies. The 16S rRNA results showed a distinct phylogenetic lineage from the other species within the Actinomadura genus. The production of antibiotic substances was investigated using GYEA solid medium. The butanolic extract contained four bioactive spots detected on thin layer chromatography plates. Among these antibiotics, a complex called 170A, which showed the more interesting antifungal activity, was selected and purified by reverse-phase HPLC. This complex is composed of four compounds. Ultraviolet-visible, infrared, mass and H nuclear magnetic resonance spectroscopy studies showed that these molecules contain an aromatic ring substituted by aliphatic chains. These compounds differ from the known antibiotics produced by Actinomadura species.

The strain AT37 was isolated from a Saharan soil by a dilution agar plating method using chitin-vitamin agar medium supplemented with rifampicin. The morphological and chemical studies indicated that this strain belonged to the genus Streptomyces. Its 16S rRNA gene sequence was determined and a database search indicated that it was closely associated with the type strain of Streptomyces novaecaesareae NBRC 13368(T) with 99.6% of similarity. An actinomycete strain designated G60 was isolated from a Saharan soil sample in Ghardaïa, Algeri... more An actinomycete strain designated G60 was isolated from a Saharan soil sample in Ghardaïa, Algeria, by a dilution agar plating method using chitin-vitamin agar medium supplemented with penicillin. Morphological and chemical studies indicated that this strain belonged to the genus Streptomyces. Analysis of the 16S rDNA sequence showed an identity level within Streptomyces species, with S. coerulescens ISP 5146 T and S. bellus ISP 5185 T the most closely related (100 % for each). However, the comparison of the morphological and physiological characteristics of the strain with those of the two nearest species showed significant differences. Strain G60 had a very strong activity against pathogenic staphylococci, including methicillin-resistant Staphylococcus aureus (MRSA) ATCC 43300, other clinical isolates of MRSA and vancomycin resistant S. aureus (VRSA) S1. One antimicrobial compound was extracted by n-hexane from the ISP2 culture medium at 5 days of fermentation culture and purified by HPLC. The chemical structure of the compound was determined after spectroscopic (1 H NMR, 13 C NMR, 1 H-1 H COSY and 1 H-13 C HMBC spectra), and spectrometric (mass spectrum) analyses. The bioactive compound was identified as di-(2-ethylhexyl) phthalate.

Objectif.-Étude de l'activité antifongique d'une souche d'actinomycète nommée AC 170, isolée à pa... more Objectif.-Étude de l'activité antifongique d'une souche d'actinomycète nommée AC 170, isolée à partir du sol de la palmeraie d'Adrar, oasis du Sud-Ouest algérien. Méthodes.-Sur la base des caractéristiques morphologiques et chimiques, la souche AC 170 étudiée est rattachée au genre Actinomadura. Elle a la particularité de ne pousser que sur des milieux solides. La production d'antibiotiques est testée sur cinq milieux. L'activité antimicrobienne est meilleure sur les milieux GYEA (Glucose-extrait de levure agar) et ISP1 (Tryptone-extrait de levure agar). L'extrait butanolique est testé vis-à-vis de 11 souches fongiques pathogènes et toxinogènes dont Candida albicans et sept espèces d'Aspergillus. Résultats.-Les résultats obtenus montrent que l'activité antifongique produite par cette souche est très importante. Une seule zone active est localisée par bioautographie. Le spectre UV-visible de l'extrait butanolique montre l'absence de polyènes. Les réactions chromogéniques laissent supposer que la molécule active est de nature aromatique. Conclusion.-L'ensemble de ces données préliminaires permet de penser que l'antifongique produit par la souche AC 170 est un composé aromatique. Son identification est en cours.

The taxonomic position of a soil isolate, strain SA 233T, recovered from Saharan soil from Algeri... more The taxonomic position of a soil isolate, strain SA 233T, recovered from Saharan soil from Algeria was established using a polyphasic approach. This isolate has been previously reported to produce three novel dithiolopyrrolone antibiotics, and preliminary chemotaxonomic and morphological characteristics suggested that it was representative of a member of the genus Saccharothrix. Phylogenetic analysis of the strain from 16S rDNA sequences, along with a detailed analysis of morphological, chemotaxonomic and physiological characteristics, indicates that it belongs to the genus Saccharothrix and represents a novel species that is readily distinguished from all recognized Saccharothrix species. The name Saccharothrix algeriensis sp. nov. is proposed for the isolate, with type strain SA 233T (=NRRL B-24137T=DSM 44581T).

Isolate AC104 presented the morphological and the chemical characteristics of the genus Actinomadura. On the basis of 76 physiological tests and 16S rDNA analysis, this isolate was determined to be quite different from the known species of Actinomadura. It is active against filamentous fungi and both Gram-positive and Gram-negative bacteria. The production of antibiotic substances was investigated using several culture media. The highest antimicrobial activities were obtained on ISP2 medium. The benzenic extract contained five bioactive spots detected on thin layer chromatography plates.
