Dengue IgM capture UltraMicroELISA test with ready-to-use reagents. Dengue is currently one of th... more Dengue IgM capture UltraMicroELISA test with ready-to-use reagents. Dengue is currently one of the most important human arboviruses. It is reported in more than 100 countries, and two and a half billion people are at risk of infection. Annually, it has been estimated that there are 50 000 000 cases of infection, with more than 500 000 hospitalized cases, and 25 000 to 50 000 deaths. Because of the emergence and re-emergence of dengue and its hemorrhagic fever, the diagnosis of this disease through laboratory tests is the first objective in any country for the differentiation of dengue from other diseases, as well as, for the support of the surveillance systems. The best choice for the diagnosis of dengue fever is an immunoglobulin M-specific capture Enzyme-linked immunosorbent assay (ELISA), due to its specificity, sensitivity, and fast performance. An example of this technique is IgM dengue ultramicro ELISA. We present the results of normalization of dengue ultramicro ELISA's new format, which includes strip plates and ready-to-use reagents. The original conditions of coating and conjugate dilution of UMELISA Dengue IgM were maintained. Both, within-run and between-run precision values were below 10%, the stability of all reagents was higher than 90%. There were no significant differences when comparing the previous kinds of reagents and strips with the new ones. Compared to hemaglutination inhibition, the sensitivity was 94.3%, the specificity was 94.1%, and the kappa concordance index was 0.91. Results indicated the ultramicro ELISA's new format maintains its reliability in the early detection of dengue.
