The protective effect of medical hypothermia in ischemia/reperfusion injury is well defined. OBJECTIVES To evaluate the late results of hypothermia through a rat testicular torsion/detorsion model compatible with human testicular torsion. STUDY DESIGN Rats were divided into 5 groups (n = 7): (1)Sham (S) group, (2)T/D group: right testis was torted for 1-h, (3)T/D + H30 group: hypothermia at 4 °C was applied for 30 min before detorsion, (4)T/D + H90 group: hypothermia at 4 °C was applied for a total of 90 min (30 min before and 1-h after detorsion), (5)H group: hypothermia at 4 °C was applied to right testis for 90 min. Testicular diameters at preoperative period and 8th postoperative week were measured. Biochemically, MPO, NO, 3-NT and 4-HNE in testicular tissue and serum levels of NO, PGF 2α, 3-NT, 8-OHdG and 4-HNE were studied. Histopathologic examination and TUNEL assay were also performed. RESULTS Biochemical and macroscopical parameters of both T/D + H30 and T/D + H90 groups were statistically different from group T/D with respect to protective effects of hypothermia. Johnsen's score was also statistically different in group T/D + H90. DISCUSSION Hypothermia can easily be applied with ice bags both in perioperative period. This is the first study evaluating the effect of hypothermia applied postoperatively. Tissue level of protein oxidation marker (3-NT) and serum levels of DNA damage (8-OHdG), lipid peroxidation (4-HNE), protein oxidation (3-NT) and oxidative stress (PGF-2α) markers were measured for the first time. CONCLUSIONS Hypothermia has been shown to be macroscopically, biochemically and histopathologically beneficial in the long-term experimental testicular torsion model.

Abstract 5199 Reactive oxygen species (ROS) may play a role in ischemia, inflammation, aging, and... more Abstract 5199 Reactive oxygen species (ROS) may play a role in ischemia, inflammation, aging, and carcinogenesis. In some studies, it was shown that the metabolism of ROS and lipid peroxidation were stimulated in solid tumors and hematological malignancies. Moreover different results for the levels of protective enzymes from ROS were reported. In this prospective study, the role of ROS, lipid peroxidation, and protective enzymes from ROS were investigated in patients with newly diagnosed hematologic malignancies. Fifty-one patients (33 were male and 18 female, with mean age of 64 ± 15 years) with newly diagnosed and untreated hematological malgnancies were enrolled to this study. Local ethical commity of our university approved this study. 21 of the patients were chronic lymphocytic leukemia (CLL), 18 multiple myeloma (MM) and 12 non-Hodgkin's lymphoma (NHL). 31 healthy individuals (19 were male and 12 female, with mean age of 63±11 years) were selected as control group. The exclusion criteria were diabetes mellitus, malignancy, acute and chronic infections, and chronic inflammatory diseases. 72%, 14%, and 14% of the patients with CLL were in Stage A, Stage B and Stage C, respectively. 17%, 33%, and 50% of patients with MM were in International prognostic index-1 (IPI1), IPI2, and IPI3, respectively. The types of patients with NHL were diffuse large B-cell lymphoma (92%) and mantle cell lymphoma (8%). According to Ann-Arbor staging system, 42% of the patients with NHL were at stage 1, 8% in stage 2, 8% at stage 3, and 42% at stage 4, respectively. When diagnosed, superoxide dismutase (SOD), glutathione peroxidase (Gpx), catalase, total glutathione (GSH), and malondialdehyde (MDA) as the predictor of lipid peroxidation were evaluated in both patients and controls bu spectrophometrical. The results were compared with Student-t, One-way ANOVA, and Chi-square tests. In this study, there is no difference between patient and control groups in terms of age and sex (p>0.05). When evaluated in terms of SOD, MDA, and catalase, no difference was found between control and patient groups (p>0.05). The levels of Gpx in patients with hematological malignancies were significantly lower than control group (p< 0.001). While the levels of Gpx in all patient groups were lower than control (p< 0.001 for all groups), no significant difference was determined between patient groups (p< 0.001). Total GSH levels in patients with hematological malignancies were higher than control group (p<0.001). While GSH levels were higher only in patients with CLL (p<0.05) and MM (p<0.005), there was no difference between patients with NHL and controls (p>0.05). However, the levels of GSH were not different in patient groups (p>0.05). In conclusion, unchanged lipid peroxidation in patients with hematological malignancies may be related to decrease in Gpx and increase in GSH. The rat model of interstitial cystitis was created by biweekly intraperitoneal administration of cyclophosphamide (CYP). In group 3, TCE (a venom extracted from a brown spider known as tarantula cubensis) was administered intravesically after the model had been created. Kurkumin (diferuloilmetan), başta Çin olmak üzere Asya ülkelerinde kanser tedavisi için alternati... more Kurkumin (diferuloilmetan), başta Çin olmak üzere Asya ülkelerinde kanser tedavisi için alternatif tıpta kullanılmaktadır. Son yıllarda, kurkuminin bazı kanser tiplerine karşı doğal bir antitümöral ajan olduğu öne sürülmektedir. Bu araştırmanın amacı, Ishikawa endometriyal kanser hücre hattı üzerinde kurkuminin antitümöral etkilerini değerlendirmektir. Ishikawa hücreleri 10, 20, 40, 80 ve 100 μM konsantrasyonlarda kurkumin ile inkübe edildi. 24, 48 ve 72 saatlik inkübasyon sonrası apoptoz, kaspaz 3/7 değerleri ve hücre canlılığı değerlendirildi. Kurkuminin in vitro olarak, Ishikawa hücre canlılığını üzerinde inhibe edici etki gösterdiği saptandı. Ayrıca 48 saat kurkumin ile 10 ve 20 μM konsantrasyonlarda apoptoz en yüksek düzeyde iken, 100μM konsantrasyonlarda hücrelerin apoptoz yerine nekroza gittiği gözlendi. In hypercholesterolemia, the evidences of chronic inflammation such as the stimulation of chemokines and cytokines, increase in endothelial adhesion molecules, and the immune reactions against oxidants on lipoproteins are detected. It was shown that statins had some beneficial effects on lipids, thrombosis, endothelial dysfunction, and smooth muscle proliferation. Aim: To investigate the effects of atorvastatin on hematological and inflammatory parameters in prospective study. Objective: The aim of this article is to measure serum antioxidant melatonin, the oxidants of nit... more Objective: The aim of this article is to measure serum antioxidant melatonin, the oxidants of nitric oxide, and malondialdehyde levels to calculate the serum oxidant–antioxidant balance based on the nitric oxide/melatonin and malondialdehyde/melatonin ratios in children with ADHD. Method: The serum melatonin, nitric oxide, malondialdehyde, and the nitric oxide/melatonin and malondialdehyde/melatonin ratios were calculated and compared between the children with ADHD ( n = 103) and healthy control participants ( n = 73). Results: Serum melatonin and nitric oxide levels were higher, and the nitric oxide/melatonin and malondialdehyde/melatonin ratios were lower in ADHD children than the control group. Melatonin was found to be significantly high, and the malondialdehyde/melatonin ratio was found to be significantly low in children with a positive ADHD family history. Gerec ve yontem: Calismaya 52 hasta (periton diyalizi=16, hemodiyaliz=12, kronik bobrek hastalikli evre 3-4=24) ve 46 saglikli cocuk alindi.
