For many years, regular exercise has been recommended as an important strategy in the prevention ... more For many years, regular exercise has been recommended as an important strategy in the prevention and management of obesity. One of the most common strategies to tackle this problem is incorporating exercise into daily lifestyle. Recently, there has also been a growing body of evidence demonstrating that high-intensity interval training (HITT) can serve as an effective substitute to traditional steady-state (SS) training that is more endurance based. Furthermore, one of the mechanisms that may underlie some of the beneficial body composition effects of regular exercise in obesity, including both SS and HITT, is the effect of exercise training on substrate utilization. While carbohydrates and fatty acids are the dominant fuels utilized during exercise, the magnitude of their relative contribution can be influenced by diet, exercise duration, intensity and training status of the individual. PURPOSE: The purpose of this study was to evaluate the effects of two different types of training HITT vs. SS on body fat (BF) and fat oxidation (FAO) in recreationally active college-aged males. METHODS: 16 recreationally active college-aged male subjects completed one of two protocols lasting 12 sessions over a period of 8 weeks: SS group completed 30 minute treadmill of running, at 65% of their VO2max, while the HITT group completed 30 second sprint running followed by an immediate 4.5 minute bout of walking six times. Rating of Perceived Exertion and heart rate were recorded at each session. Subjects were tested for body composition and VO2max values pre-and post-training sessions. FAO, resting metabolic rate and BF were measured occurred 3 hours after the first session, session 6, and session 12. All subjects maintained >85% compliance with exercise. RESULTS: There was no significant difference in body composition (t= .495); however the loss of BF was 10% for HITT group while only 5% for SS. There was no significant difference in FAO between training groups (t=-0.485, p=0.643). CONCLUSION: Results showed no statistical difference in FAO between the stimuli of HITT versus SS training. However, there was a slight decrease in body fat percentage seen in the HITT group. When devising strategies to implement exercise into daily lifestyle, practitioners should consider HITT as another mode of training for clients or patients.

18F-FDG-PET/CT Imaging as Survival Predictor in Patients with Metastatic Uveal Melanoma

ABSTRACT Aims To correlate metabolic activity by 18F-FDG-PET/CT with survival in uveal melanoma p... more ABSTRACT Aims To correlate metabolic activity by 18F-FDG-PET/CT with survival in uveal melanoma patients with liver metastases (LM). Methods We undertook a retrospective study of 34 patients with uveal melanoma and LM who underwent a 18F-FDG-PET/CT at the time of metastatic diagnosis. Metabolic activity of LM was measured by standardized uptake value (SUV) and by grouping in 18F-FDG-PET-negative (lower or same metabolic activity in LM than in normal liver parenchyma (NLP) comparing to 18F-FDG-PET-positive (higher metabolic activity in LM than in NLP). These measures were correlated with overall survival (OS). Results Patients were treated in 2 institutions between 2004-2010. There were 19 women and 15 men. Median age was 62 years (30-84). Median follow-up was 16.7 months. Treatment of primary tumor was: 15 enucleation (44.1%), 3 resection (8.8%) and 16 brachytherapy (47.1%). LM were diagnosed on screening protocol and histologically confirmed if possible (85.3%). Median major diameter of LM was 20 mm (9-130). Median maximum SUV of LM was 8.7 (2-25). 18F-FDG-PET/CT was negative for metabolic activity in 6 patients and positive in 28 patients. All 6 patients with negative 18F-FDG-PET/CT were histologically confirmed. Metastatic disease treatments were: chemotherapy (27 patients), liver surgery (9 patients), liver stereotactic radiotherapy (2 patients), liver radiofrequency ablation (1 patient). Median OS was 18.8 months (CI 95% 7.2-30.4). Median OS was significantly higher (p = 0.001) in patients with 18F-FDG-PET-negative (median not reached) than in patients with 18F-FDG-PET-positive (15.1 months, CI 95% 11.5-18.7). Assessed as a continuous variable, maximum SUV in the liver was correlated with OS (p = 0.002, HR 1.1, CI 95% 1.03-1.2). Maximum SUV greater than 5 was related with poor prognosis (p = 0.025). This paper explores a fundamental connection between ductility and domain size in metallic solids... more This paper explores a fundamental connection between ductility and domain size in metallic solids under extreme conditions of cryogenic temperatures and strain rates (10 8 s-1). A series of novel experiments, backed by multiscale modelling and transmission electron microscopy (TEM) analysis, are presented that involve loading of TEM-ready single crystal nanopillar samples of Cu of varying lengths (50 nm to 1 mm) and aspect ratios (50 nm to 100 nm in diameter) by laser-generated stress waves of sub-nanosecond rise times, under extreme conditions of strain rate (>10 8 s-1) and temperature (100K). The nucleation stress for Shockley partials, which can be taken as a proxy for the onset of ductile deformation, was measured to be only 1 GPa. This is an order of magnitude lower than the previously measured values of 35 GPain bulk geometries. TEM observations show remarkable ability of the material to rearrange itself through motion of dislocations to form subgrain boundaries within a very short duration of only few nanoseconds.

Conclusions: Hair dyes and PPD constitute a significant cause of contact dermatitis. There is an urgent need for creating consumer awareness regarding hair-dyes contact sensitivity and the significance of performing sensitivity testing prior to actual use.

Abstract Recently, reports on the fabrication of nonstructural configuration have generated scien... more Abstract Recently, reports on the fabrication of nonstructural configuration have generated scientific interest due to increased awareness of the oxide nanotubes in the biomedical implant field and other industrial approaches. Here, highly ordered ZrO 2 TiO 2 Nb 2 O 5 Al 2 O 3 nanotube arrays were grown by physical vapor deposition (PVD) of a zirconium layer on titanium-niobium alloy implant (IMPTi67) and succeeding anodic oxidation at a constant voltage (60 V) with exposure times ranging from 30 to 300 min. To crystallize the resulting mixed oxide nanotubes, heat treatment was also applied. The nanotubes were placed in an atmospheric furnace at temperatures ranging from 450 to 800 °C for 2 h. This treatment provided a morphological evolution was strongly influenced by the anodization time, where highly aligned bamboo-shaped oxide nanotubes (161 ± 44 nm in inside diameter and ∼1 μm in length) were formed after 300 min of anodization. After annealing at temperature below 500 °C, the nano-array configuration remained stable and the average inner diameter decreased to around 40 nm. The effects of anodically fabrication time on the adhesion strength of four-phase multilayered mixed oxide nanotubes on anodized Zr/IMPTi67 surfaces assed by microscratch analysis. From the results, increasing the annealing temperature further to 800 °C fully destroyed the nanotubes and consequently the tubular arrangement was converted to a coarse grain structure. It was revealed that annealing at 450 °C for 2 h was the optimum conditions for stable nanotubular array generation, where the highest adhesion strength (shifted from 2595.59 to 2640.12 mN), microhardness (372 HV) and hydrophilicity (15 ± 1°) were recorded. It is notify that the adhesion strength of highly ordered mixed nanotubes was remarkably decreased after thermal treatment at 800 °C (1817.51 mN). Through the laser spallation analysis, the intrinsic adhesion strength of the Zr/IMPTi67 interface was found to be at 63 MPa, while the first stages of failure occurred at a stress state of 35 MPa. The adhesion of the film with the tubular structure proved to have a significantly larger adhesion strength of 246 MPa for the complete failure and 203 MPa for incipient stages of spallation. The considered surface modification can be an effective step in determining the interface strength between biomedical implants and the nanostructured coatings.

We report here the synthesis and structure-activity relationship (SAR) of a novel series of triaz... more We report here the synthesis and structure-activity relationship (SAR) of a novel series of triazole containing mammalian target of rapamycin (mTOR) kinase inhibitors. SAR studies examining the potency, selectivity and PK parameters for a series of triazole containing 4,6- or 1,7-disubstituted-3,4-dihydropyrazino[2,3-b]pyrazine-2(1H)-ones resulted in the identification of triazole containing mTOR kinase inhibitors with improved PK properties. Potent compounds from this series were found to block both mTORC1(pS6) and mTORC2(pAktS473) signaling in PC-3 cancer cells, in vitro and in vivo. When assessed in efficacy models, analogs exhibited dose-dependent efficacy in tumor xenograft models. This work resulted in the selection of CC 115 for clinical development.

During a period of 4−6 weeks after CRT, clinical response rate (cRR) was evaluated by control exa... more During a period of 4−6 weeks after CRT, clinical response rate (cRR) was evaluated by control examinations and patients were sent to the Institute of digestive disease at department for surgery if their tumor were achieved complete or partial regression. Results: All patients finished radiotherapy course. In 22 pts (47.38%) chemotherapy was interrupted due to toxicity: in 10 pts after 2 cycles of chemotherapy and in 12 pts after 3 cycles. Toxicity grade 1 and 2 had 24 pts (52.17%) Grade 3 and 4 toxicity was noted: cardiotoxicity in 3 pts, leucopenia gr 4 in 5 pts, neutropenia gr 3 in 6 pts, neutropenia gr 4 in 8 pts. cRR was achieved in 27/46 patients (58.7%): complete response in 3 pts (6.52%), partial response in 24 pts (52.17%), stabile disease in 13 pts (28.26%) and early progression in 6 pts (13.04%). According to histopathological assessment by Mandard, complete tumor regression TRG1 was noted in 5 pts (41.67%), partial tumor regression TRG2 in 1 pt (8.46%), TRG3 in 3 pts (25%), minimal tumor regression TRG4 in 3 pts (25%). Conclusion: Our preliminary results show promising treatment results on neoadjuvant CRT with Cisplatin and LV5FU2. We have to enroll more patients, to reach statistical power of more than 80%. RATIONALE: It's thought that aeroallergen sensitization is infrequent in preschool and breast-fed... more RATIONALE: It's thought that aeroallergen sensitization is infrequent in preschool and breast-fed children. We expect to find out the frequency and type of allergen sensitization in children under the age of five. METHODS: 250 medical histories of children under the age of five who were derived from primary care centers located in our healthcare area with suspicion of asthma were reviewed, selecting 174 with asthma. We used SPSS version 12 to create a database and analyse the impact in asthma of several variables: age, date of consultation and birth, atopic dermatitis, food allergy, asthma severity, type of sensitization, and celiac disease. RESULTS: 41% of the entire children studied had aeroallergen sensitization. 39% of those who suffered from mild intermittent asthma had at least one positive aeroallergen. 51% of mild persistent asthma children were sensitized. 37% of moderate persistent asthma was explained by aeroallergens. 17% of the population sensitized had egg allergy. 10% showed present or overcame milk allergy. One important finding is that 22% of children showed atopic dermatitis and asthma due to aeroallergens compared to 21% of children under the age of five who had a history of atopic dermatitis but no aeroallergen sensitization. We observed no aeroallergen sensitization under 12 months of age. CONCLUSIONS: Precocious neumoallergen sensitization in breast-fed and preschool children suffering from asthma is frequent in our healthcare area currently. Therefore, allergologic study is suitable.

Conclusion: These studies demonstrate the successful use of NIR fluorescence and ICG in sentinel lymph node mapping of breast cancer patients. The optimal parameters are a dose of 500 mM ICG that is not premixed with HSA, and the use of patent blue can be omitted.

The aim of this study was to prospectively evaluate 18 F-FDG PET/CT in predicting response to neo... more The aim of this study was to prospectively evaluate 18 F-FDG PET/CT in predicting response to neoadjuvant chemotherapy in large primary breast cancer. Methods Fifty consecutive patients underwent PET/CT at baseline and after the second cycle. Baseline MRI was performed to establish tumour size. All findings were confirmed by histopathological analysis. Changes in maximum standardized uptake value (SUV max) between baseline study and after two cycles of neoadjuvant chemotherapy (epirubicin + cyclophosphamide + taxanes) were compared using response evaluation criteria in solid tumours (RECIST) criteria and the Miller and Payne (M&P) scale. Results The mean tumour size was 4.3±1.4 cm. Forty patients were considered responders and ten as nonresponders. SUV max changes in patients with good prognosis (M&P grades 4-5) were higher than in patients with bad prognosis (M&P grades 1-3) (p=0.025). SUV max changes between responders and non-responders following RECIST criteria were also statistically significant (p= 0.0028). A cutoff ΔSUV value of 40% differentiates both groups, with a sensitivity of 77% and a specificity of 80%. Conclusion 18 F-FDG PET/CT can predict response to neoadjuvant chemotherapy at an early stage.

The considered surface modification can be an effective step in determining the interface strength between biomedical implants and the nanostructured coatings.

We report here the synthesis and structure-activity relationship (SAR) of a novel series of triaz... more We report here the synthesis and structure-activity relationship (SAR) of a novel series of triazole containing mammalian target of rapamycin (mTOR) kinase inhibitors. SAR studies examining the potency, selectivity and PK parameters for a series of triazole containing 4,6- or 1,7-disubstituted-3,4-dihydropyrazino[2,3-b]pyrazine-2(1H)-ones resulted in the identification of triazole containing mTOR kinase inhibitors with improved PK properties. Potent compounds from this series were found to block both mTORC1(pS6) and mTORC2(pAktS473) signaling in PC-3 cancer cells, in vitro and in vivo. When assessed in efficacy models, analogs exhibited dose-dependent efficacy in tumor xenograft models. This work resulted in the selection of CC 115 for clinical development.

During a period of 4−6 weeks after CRT, clinical response rate (cRR) was evaluated by control exa... more During a period of 4−6 weeks after CRT, clinical response rate (cRR) was evaluated by control examinations and patients were sent to the Institute of digestive disease at department for surgery if their tumor were achieved complete or partial regression. Results: All patients finished radiotherapy course. In 22 pts (47.38%) chemotherapy was interrupted due to toxicity: in 10 pts after 2 cycles of chemotherapy and in 12 pts after 3 cycles. Toxicity grade 1 and 2 had 24 pts (52.17%) Grade 3 and 4 toxicity was noted: cardiotoxicity in 3 pts, leucopenia gr 4 in 5 pts, neutropenia gr 3 in 6 pts, neutropenia gr 4 in 8 pts. cRR was achieved in 27/46 patients (58.7%): complete response in 3 pts (6.52%), partial response in 24 pts (52.17%), stabile disease in 13 pts (28.26%) and early progression in 6 pts (13.04%). According to histopathological assessment by Mandard, complete tumor regression TRG1 was noted in 5 pts (41.67%), partial tumor regression TRG2 in 1 pt (8.46%), TRG3 in 3 pts (25%), minimal tumor regression TRG4 in 3 pts (25%). Conclusion: Our preliminary results show promising treatment results on neoadjuvant CRT with Cisplatin and LV5FU2. We have to enroll more patients, to reach statistical power of more than 80%. RATIONALE: It's thought that aeroallergen sensitization is infrequent in preschool and breast-fed... more RATIONALE: It's thought that aeroallergen sensitization is infrequent in preschool and breast-fed children. We expect to find out the frequency and type of allergen sensitization in children under the age of five. METHODS: 250 medical histories of children under the age of five who were derived from primary care centers located in our healthcare area with suspicion of asthma were reviewed, selecting 174 with asthma. We used SPSS version 12 to create a database and analyse the impact in asthma of several variables: age, date of consultation and birth, atopic dermatitis, food allergy, asthma severity, type of sensitization, and celiac disease. RESULTS: 41% of the entire children studied had aeroallergen sensitization. 39% of those who suffered from mild intermittent asthma had at least one positive aeroallergen. 51% of mild persistent asthma children were sensitized. 37% of moderate persistent asthma was explained by aeroallergens. 17% of the population sensitized had egg allergy. 10% showed present or overcame milk allergy. One important finding is that 22% of children showed atopic dermatitis and asthma due to aeroallergens compared to 21% of children under the age of five who had a history of atopic dermatitis but no aeroallergen sensitization. We observed no aeroallergen sensitization under 12 months of age. CONCLUSIONS: Precocious neumoallergen sensitization in breast-fed and preschool children suffering from asthma is frequent in our healthcare area currently. Therefore, allergologic study is suitable.

The sentinel lymph node was detected in all patients using NIR fluorescence before the blue dye could be observed. Conclusion: These studies demonstrate the successful use of NIR fluorescence and ICG in sentinel lymph node mapping of breast cancer patients. The optimal parameters are a dose of 500 mM ICG that is not premixed with HSA, and the use of patent blue can be omitted.

The aim of this study was to prospectively evaluate 18 F-FDG PET/CT in predicting response to neo... more The aim of this study was to prospectively evaluate 18 F-FDG PET/CT in predicting response to neoadjuvant chemotherapy in large primary breast cancer. Methods Fifty consecutive patients underwent PET/CT at baseline and after the second cycle. Baseline MRI was performed to establish tumour size. All findings were confirmed by histopathological analysis. Changes in maximum standardized uptake value (SUV max) between baseline study and after two cycles of neoadjuvant chemotherapy (epirubicin + cyclophosphamide + taxanes) were compared using response evaluation criteria in solid tumours (RECIST) criteria and the Miller and Payne (M&P) scale. Results The mean tumour size was 4.3±1.4 cm. Forty patients were considered responders and ten as nonresponders. SUV max changes in patients with good prognosis (M&P grades 4-5) were higher than in patients with bad prognosis (M&P grades 1-3) (p=0.025). SUV max changes between responders and non-responders following RECIST criteria were also statistically significant (p= 0.0028). A cutoff ΔSUV value of 40% differentiates both groups, with a sensitivity of 77% and a specificity of 80%. Conclusion 18 F-FDG PET/CT can predict response to neoadjuvant chemotherapy at an early stage.
