Morphological examination of the highly polarized protozoan parasite Toxoplasma gondii suggests t... more Morphological examination of the highly polarized protozoan parasite Toxoplasma gondii suggests that secretory traffic in this organism progresses from the endoplasmic reticulum to the Golgi apparatus using the nuclear envelope as an intermediate compartment. While the endoplasmic reticulum is predominantly located near the basal end of the parasite, the Golgi is invariably adjacent to the apical end of the nucleus, and the space between the Golgi and nuclear envelope is filled with numerous coatomer-coated vesicles. Staining with antiserum raised against recombinant T. gondii beta-COP confirms its association with the apical juxtanuclear region. Perturbation of protein secretion using brefeldin A, microtubule inhibitors or dithiothreitol disrupts the Golgi, causing swelling of the nuclear envelope, particularly at its basal end. Most species of the protozoan phylum Apicomplexa harbor an endosymbiotic organelle—the apicoplast... more Most species of the protozoan phylum Apicomplexa harbor an endosymbiotic organelle—the apicoplast— acquired when an ancestral parasite engulfed a eukaryotic plastid-containing alga. Several hundred proteins are encoded in the parasite nucleus and are posttranslationally targeted to the apicoplast by a distinctive bipartite signal. The N-terminal 20 to 30 amino acids of nucleus-encoded apicoplast targeted proteins function as a classical signal sequence, mediating entry into the secretory pathway. Cleavage of the signal sequence exposes a transit peptide of variable length (50 to 200 amino acids) that is required for directing proteins to the apicoplast. Apicomplexan parasites release factors via specialized secretory organelles (rhoptries, microneme... more Apicomplexan parasites release factors via specialized secretory organelles (rhoptries, micronemes) that are thought to control host cell responses. In order to explore parasite-mediated modulation of host cell signaling pathways, we exploited a phylogenomic approach to characterize the Toxoplasma gondii kinome, defining a 44 member family of coccidian-specific secreted kinases, some of which have been previously implicated in virulence. Comparative genomic analysis suggests that ‘ROPK ’ genes are under positive selection, and expression profiling demonstrates that most are differentially expressed between strains and/or during differentiation. Integrating diverse genomic-scale analyses points to ROP38 as likely to be particularly important in parasite biology. Polyethylene glycol (PEG) induces rapid fusion of LM cells. Membrane fusion, as detected by forma... more Polyethylene glycol (PEG) induces rapid fusion of LM cells. Membrane fusion, as detected by formation of pentalaminar membrane arrays, occurs as early as 1 min after PEG treatment. The entire cell surface arrears to be capable of fusion since fusion occurs in regions where pseudopodia make contact with each other or with a neighbouring cell body and also in areas where cells are in contact along their entire periphery. Cytoskeletal components showed no apparent deleterious effect from PEG treatment or subsequent cell fusion as determined by thin-section EM. Freeze-fracture of monolayer cultures reveals a thermotropic rearrangement of intramembranous particles following PEG treatment.

We have used drugs to examine the role(s) of the actin and microtubule cytoskeletons in the intra... more We have used drugs to examine the role(s) of the actin and microtubule cytoskeletons in the intracellular growth and replication of the intracellular protozoan parasite, Toxoplasma gondii. By using a 5 minute infection period and adding the drugs shortly after entry we can treat parasites at the start of intracellular development and 6–8 hours prior to the onset of daughter cell budding. Using this approach we found, somewhat surprisingly, that reagents that perturb the actin cytoskeleton in different ways (cytochalasin D, latrunculin A and jasplakinolide) had little effect on parasite replication although they had the expected effects on the host cells. These actin inhibitors did, however, disrupt the orderly turnover of the mother cell organelles leading to the formation of a large residual body at the posterior end of each pair of budding parasites. Проанализировать эволюцию концептуальных подходов к зонированию территорий по степени опасности, а также выработать предложения по их уточнению с учетом законов войны, положений Военной доктрины, Национальной стратегии по снижению риска возникновения чрезвычайных ситуаций в Республике Беларусь на 2019-2030 годы. Методы. В статье использованы общенаучные методы исследования: индукции и дедукции, анализа и синтеза, сравнения и обобщения, аналогии. Результаты. Определены роль и место зонирования территорий по степени опасности в общей системе оборонных мероприятий страны, выработаны предложения по совершенствованию требований технических нормативных правовых актов в сфере гражданской обороны. Область применения исследований. Результаты исследования вносят вклад в развитие теории гражданской обороны и могут использоваться при переработке требований технических нормативных правовых актов в части уточнения концептуальных подходов к зонированию территорий по степени опасности.

Application of Fourier analysis techniques to images of isolated, frozen-hydrated subpellicular m... more Application of Fourier analysis techniques to images of isolated, frozen-hydrated subpellicular microtubules from the protozoan parasite Toxoplasma gondii demonstrates a distinctive 32 nm periodicity along the length of the microtubules. A 32 nm longitudinal repeat is also observed in the double rows of intramembranous particles seen in freeze-fracture images of the parasite's pellicle; these rows are thought to overlie the subpellicular microtubules. Remarkably, the 32 nm intramembranous particle periodicity is carried over laterally to the single rows of particles that lie between the microtubule-associated double rows. This creates a two-dimensional particle lattice, with the second dimension at an angle of approximately 75 degrees to the longitudinal rows (depending on position along the length of the parasite). PV1 is structurally distinctive due to its large size, yet it lacks a tubulovesicular network (C. A. Speer, M. Tilley, M. Temple, J. A. Blixt, J. P. Dubey, and M. W. White, Mol. Biochem. Parasitol. 75:75-86, 1995). Confirming the finding that sporozoites have a different electron-dense-granule composition, we have now found that sporozoites within oocysts lack the mRNAs encoding the 5' nucleoside triphosphate hydrolases (NTPase). This rate of growth is intrinsic, as neither the number of host cells invaded nor host cell type appears to influence emergent tachyzoite replication. Fast tachyzoite growth was not persistent, and following ∼20 divisions, the population uniformly shifted to slower growth. Parasites 10 days post-sporozoite infection doubled only once every 15 h and, unlike emergent tachyzoites, they grew at this slower rate over several months of continuous cell culture. The spontaneous change in tachyzoite growth rate preceded the expression of the bradyzoite-specific marker,BAG1. Here, we identify ASP5 substrates by selectively enriching N-terminally derived peptides from wild-type and parasites. We reveal more than 2,000 unique N-terminal peptides, mapping to both natural N termini and protease cleavage sites. Several of these peptides mapped directly downstream of the characterized ASP5 cleavage site, arginine-arginine-leucine (RRL). We validate candidates as true ASP5 substrates, revealing they are not processed in parasites lacking ASP5 or in wild-type parasites following mutation of the motif from RRL to ARL.
