The interaction between Sn (Il)-ion and PAH was studied with the purpose to establish whether PAH could form complex with Sn (Il)-ion during the labeling with technetium-99m. HPLC, spectrophotometric and pH-metric methods were used. Dissociation constants of PAH, thermodynamic parameters of PAH dissociation, as well as stability constant of tin (II)-PAH were determined.

A modified 99mTc(Sn)-pyrophosphate (PyP) kit for the application in nuclear cardiology (radiovent... more A modified 99mTc(Sn)-pyrophosphate (PyP) kit for the application in nuclear cardiology (radioventriculography, angiocardiography, scintigraphy of blood pool) was developed. Each vial contains 12 mg PyP (Na4P207), 4 mg SnClz.2H20, 2.5 mg gentisic acid and 10 mg NaC1. The reconstitution is performed by dissolving the lyophilized kit in 3 ml 0.9% NaC1. In comparison with the standard pyrophosphate kit for bone scanning and detection of miocardial infarction, it contains an increased amount of Sn(II) so that the molar ratio ligand/rednctant is lowered from 25 to 2.5. The radiochemical analyses showed that the radiochemical purity of the labeled kit is high (~90%) during three hours after addition of 99mTc-activity. The shelf-life of the inactive freeze-dried preparation is up to four months providing that it is kept in vacuum and at appropriate temperature (2 8 ~ The biodistribution studies revealed increased accumulation in blood and low uptake by liver and kidneys. It was concluded that the modified kit performs stable and reproducible properties.

Small glucose-modified liposomes (GMLs) were loaded with magnetic Fe3O4 nanoparticles (MNPs) and ... more Small glucose-modified liposomes (GMLs) were loaded with magnetic Fe3O4 nanoparticles (MNPs) and fluorescein using a standard thin layer preparation procedure and a varying lipid/MNPs ratio. The liposomes were characterized with TEM and DLS measurements, and MNPs encapsulation rate was determined using ICP-OES. Prepared liposomes were stored at 5 °C for 30 days and subsequently exposed to an external magnetic field (20 mT) with varying exposure times (2‒20 min), at room temperature. The release of fluorescein from GMLs induced by the magnetic field exposures was quantified, showing a high release rate (25‒85%) depending on the concentration of MNPs in GMLs. EPR measurements were conducted during the liposomes storage period in order to provide semi-quantitative information of possible MNPs oxidation from Fe3O4 to Fe2O3 inside the liposomes, impacting MNPs magnetic properties. In contrast to the MNPs water dispersion, no significant change in the EPR signal of MNPs encapsulated inside GMLs was detected over the course of 30 days. The data presented in this study indicate that GMLs loaded with MNPs maintain a high stability for prolonged periods of time and that this delivery system may be used for magnetically assisted controlled drug release.

Metaiodobenzylguanidine (MIBG) is a norepinephrine analogue which labeled with radionuclides 131I and 123I is applicable in the diagnosis and treatment of neuroectodermal tumors. The main problem related to the production of 131I-MIBG is low radiochemical yield and fast in vitro and in vivo dehalogenisation, causing the binding of free 131I in thyroid glands and stomach, which further affects the misinterpretation of results of nuclear medicine imaging. Magnetic nanoparticle (MNP)-mediated hyperthermia (MH) coupled with radiation therapy (RT) is a n... more Magnetic nanoparticle (MNP)-mediated hyperthermia (MH) coupled with radiation therapy (RT) is a novel approach that has the potential to overcome various practical difficulties encountered in cancer treatment. In this work, we present recommendations for the in vitro and in vivo testing and application of the two treatment techniques. These recommendations were developed by the members of Working Group 3 of COST Action TD 1402: Multifunctional Nanoparticles for Magnetic Hyperthermia and Indirect Radiation Therapy ("Radiomag"). The purpose of the recommendations is not to provide definitive answers and directions but, rather, to outline those tests and considerations that a researcher must address in order to perform in vitro and in vivo studies. The recommendations are divided into 5 parts: (a) in vitro evaluation of MNPs; (b) in vitro evaluation of MNP-cell interactions; (c) in vivo evaluation of the MNPs; (d) MH combined with RT; and (e) pharmacokinetic studies of MNPs. ...

This is especially relevant when more than one form of radiometal/ligand complex can be formed during radiolabelling, such as complexes of 99mTc or 188Re with meso-2,3-dimercaptosuccinic acid (DMSA), where depending on the pH, metal can occur either at +3 or +5 oxidation state. The aim of our study was to evaluate possibilities for optimization of chromatographic systems leading to specific and reliable analytical method for determination of the identity and RCP of DMSA complexes with 99mTc or 188Re. ABSTRACT The purpose of this study was to compare some physicochemical characteristics as well as... more ABSTRACT The purpose of this study was to compare some physicochemical characteristics as well as pharmacokinetic behavior of 99mTc-PAH, as a novel renal agent, with 99mTc-MAG3 and 131I-OIH. 99mTc-PAH was prepared from lyophilized kit by adding 99mTcO4–. Labeled complex was stabile and high radiochemical purity radiopharmaceutical, with a low percentage of protein bound to human albumin and hydrophilic character. In spite of its smaller renal uptake, 99mTc-PAH gave satisfactory renal images. 99mTc-PAH showed faster urinary elimination than 99mTc-MAG3 and similar to those one for 131I-OIH. The comparison of pharmacokinetic parameters of 99mTc-PAH, 99mTc-MAG3 and 131I-OIH indicated the favorable characteristics of 99mTc-PAH.

A new procedure for the aggregation of human albumin and 99m Tc-labelling of the prepared macroag... more A new procedure for the aggregation of human albumin and 99m Tc-labelling of the prepared macroaggregated albumin are presented. Simple methods for quantifying of all the radiochemical impurities existing in 99m Tc-MAA were tested. Thus, 85 % methanol was used as the mobile phase in paper and ITL chromatography with Whatman N o 1 and ITLC-SA strips. A system of two solvents (acetone and 1 M NaCl or 0.9 % NaCl) was used for 3 MM paper, ITLC-SA and ITLC-SG strips and silica gel plates as the stationary phase. Low-voltage paper electrophoresis with Whatman 3 MM paper sheets soaked in barbiturate buffer and the gel chromatography column method (Sephadex G-25) were also applied. Filtration through syringe filters, proposed by European and Yugoslav Pharmacopoeia, was performed for comparison. The application of the mentioned tests lead to consistent results for the labelling efficiency (> 98.5 %) and percent radiochemical impurities of 99m Tc-MAA. Determination of the particles number in a counter chamber and their size distribution under a light microscope with a calibrated ocular scale gave the result of 300000-350000 particles per 1 mg of HA. This confirmed that the human albumin macroaggregates prepared by our new procedure is remarkably improved and convenient for routine diagnostic purposes.

Each veal of lyophilised kit contain hippuric (Hipp) or p-aminohippuric acid (PAH), diethyltriaminepentaacetic acid as calciumtrisodium salt (CaNa3DTPA) and stannouschloride (SnCl2·xH2O), in molar ratio Hipp/PAH:DTPA=4∶1. They are high radiochemical purity radiopharmaceuticals, with hydrophilic character and low percentage of protein binding. The ITL chromatography and HPLC analyses of these labeled compounds have shown almost identical results as99mTc-DTPA, but their biological behavior in rats confirm certain differences.99mTc-Hipp is a renal agent clearing by the glomerular system, with better pharmacokinetical parameters than99mTc-DTPA:t 1/2(α)=4.1 min,t 1/2(β)=198.6 min,K cl=1.2·10−2min−1 and a twofold value for blood clearance (Cl=2.07 ml/min).99mTc-PAH is a quite different renal agent, rapidly secreted by kidney as a tubular secretion agent. Its pharmacokinetical parameters:t 1/2(α)=2.5 min,t 1/2(β)=41.7 min andK cl=5.1·10−4 min−1 are almost equal to those of99mTc-MAG3, but the blood clearance of Cl=5.22 ml/min is even higher than that of IOH clearance.

Colloidal particle size is an important characteristic to consider when choosing a radiopharmaceu... more Colloidal particle size is an important characteristic to consider when choosing a radiopharmaceutical for diagnosis and therapeutic purposes in nuclear medicine. Photon correlation spectroscopy (PCS) and transmission electron microscopy (TEM) were used to determine the particle-size distribution of 90 Y-and 99m Tc-labelled antimony trisulfide (Sb 2 S 3) and tin colloids (Sn-colloid). 90 Y-Sb 2 S 3 and 99m Tc-Sb 2 S 3 were found to have a diameter of 28.92 ± 0.14 and 35.61 ± 0.11 nm, respectively, by PCS. By TEM, 90 Y-Sb 2 S 3 particles were measured to be 14.33 ± 0.09 nm. 90 Y-labelled Sn colloid were found to exist with a d v(max1) of 805 nm and a d v(max2) of 2590 nm, by PCS, whereas 99m Tc-Sn colloid was shown to have more than 80% of radioactive particles of approximately 910 nm by PCS. For 90 Y-labelled Sb 2 S 3 and Sn colloid, a comparison of TEM and PCS indicates that these techniques found significantly different mean diameters. TEM has an excellent resolution necessary for radiocolloid particle-sizing analysis, and it is a desirable size-measuring technique because it is more reliable than PCS.

It was reported that novel O, O′-diethyl-(S, S)-ethylenediamine- N, N′-di-2-(3-cyclohexyl) propan... more It was reported that novel O, O′-diethyl-(S, S)-ethylenediamine- N, N′-di-2-(3-cyclohexyl) propanoate dihydrochloride (DE-EDCP) displayed in vitro antiproliferative activity on several human and mouse cancer cell lines, which was comparable to that of the prototypical anticancer drug cisplatin. In order to reveal its toxicity profile, acute and repeated-dose toxicity studies were performed in Naval Medical Research Institute (NMRI) Han mice. The intravenous LD50 values of DE-EDCP were found to be 95.3 and 101.3 mg/kg body weight in female and male mice, respectively. In the subacute toxicity study, DE-EDCP was administered intravenously at the doses of 15, 25, and 40 mg/kg/day for a period of 28 days. There were no adverse effects on general condition, growth, feed and water consumption, and hematological parameters. There was a significant increase in urea and alanine aminotransferase in female mice and aspartate aminotransferase and alkaline phosphatase in both genders in 40 mg/kg...

The purpose of this work is to assess accuracy and compare the performance of radionuclide calibr... more The purpose of this work is to assess accuracy and compare the performance of radionuclide calibrators (RNCs) used in nuclear medicine departments in Serbia. Testing of the RNCs included verification of measurement accuracy, as well as analysis of routinely used quality control protocols, by using the certified radioactivity standards (57Co, 137Cs). RNCs performances were assessed with 99mTc through comparison of reference value for radionuclide activity and RNC measurements. Results of the intercomparison revealed that RNCs, 15 in total, are accurate within 10% in vial geometry and within 15% in syringe geometry. Most of them showed similar performance. The results revealed that container geometry is an important influencing parameter in the accuracy of activity measurement. Obtained results indicate a need for regular calibration and implementation of Quality Control program in order to achieve and maintain the accuracy of activity measurements in nuclear medicine.

Each veal of lyophilised kit contain hippuric (Hipp) or p-aminohippuric acid (PAH), diethyltriaminepentaacetic acid as calciumtrisodium salt (CaNa3DTPA) and stannouschloride (SnCl2·xH2O), in molar ratio Hipp/PAH:DTPA=4∶1. They are high radiochemical purity radiopharmaceuticals, with hydrophilic character and low percentage of protein binding. The ITL chromatography and HPLC analyses of these labeled compounds have shown almost identical results as99mTc-DTPA, but their biological behavior in rats confirm certain differences.99mTc-Hipp is a renal agent clearing by the glomerular system, with better pharmacokinetical parameters than99mTc-DTPA:t 1/2(α)=4.1 min,t 1/2(β)=198.6 min,K cl=1.2·10−2min−1 and a twofold value for blood clearance (Cl=2.07 ml/min).99mTc-PAH is a quite different renal agent, rapidly secreted by kidney as a tubular secretion agent. Its pharmacokinetical parameters:t 1/2(α)=2.5 min,t 1/2(β)=41.7 min andK cl=5.1·10−4 min−1 are almost equal to those of99mTc-MAG3, but the blood clearance of Cl=5.22 ml/min is even higher than that of IOH clearance.

Colloidal particle size is an important characteristic to consider when choosing a radiopharmaceu... more Colloidal particle size is an important characteristic to consider when choosing a radiopharmaceutical for diagnosis and therapeutic purposes in nuclear medicine. Photon correlation spectroscopy (PCS) and transmission electron microscopy (TEM) were used to determine the particle-size distribution of 90 Y-and 99m Tc-labelled antimony trisulfide (Sb 2 S 3) and tin colloids (Sn-colloid). 90 Y-Sb 2 S 3 and 99m Tc-Sb 2 S 3 were found to have a diameter of 28.92 ± 0.14 and 35.61 ± 0.11 nm, respectively, by PCS. By TEM, 90 Y-Sb 2 S 3 particles were measured to be 14.33 ± 0.09 nm. 90 Y-labelled Sn colloid were found to exist with a d v(max1) of 805 nm and a d v(max2) of 2590 nm, by PCS, whereas 99m Tc-Sn colloid was shown to have more than 80% of radioactive particles of approximately 910 nm by PCS. For 90 Y-labelled Sb 2 S 3 and Sn colloid, a comparison of TEM and PCS indicates that these techniques found significantly different mean diameters. TEM has an excellent resolution necessary for radiocolloid particle-sizing analysis, and it is a desirable size-measuring technique because it is more reliable than PCS.

It was reported that novel O, O′-diethyl-(S, S)-ethylenediamine- N, N′-di-2-(3-cyclohexyl) propan... more It was reported that novel O, O′-diethyl-(S, S)-ethylenediamine- N, N′-di-2-(3-cyclohexyl) propanoate dihydrochloride (DE-EDCP) displayed in vitro antiproliferative activity on several human and mouse cancer cell lines, which was comparable to that of the prototypical anticancer drug cisplatin. In order to reveal its toxicity profile, acute and repeated-dose toxicity studies were performed in Naval Medical Research Institute (NMRI) Han mice. The intravenous LD50 values of DE-EDCP were found to be 95.3 and 101.3 mg/kg body weight in female and male mice, respectively. In the subacute toxicity study, DE-EDCP was administered intravenously at the doses of 15, 25, and 40 mg/kg/day for a period of 28 days. There were no adverse effects on general condition, growth, feed and water consumption, and hematological parameters. There was a significant increase in urea and alanine aminotransferase in female mice and aspartate aminotransferase and alkaline phosphatase in both genders in 40 mg/kg...

A protein integrity and purity were proved by applying of reducing sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). Vibrational spectroscopy (Infra-red (IR) and Raman spectroscopy) provide molecular structure information and are convenient for verification of changes in the secondary structure. The purpose of this work is to assess accuracy and compare the performance of radionuclide calibr... more The purpose of this work is to assess accuracy and compare the performance of radionuclide calibrators (RNCs) used in nuclear medicine departments in Serbia. Testing of the RNCs included verification of measurement accuracy, as well as analysis of routinely used quality control protocols, by using the certified radioactivity standards (57Co, 137Cs). RNCs performances were assessed with 99mTc through comparison of reference value for radionuclide activity and RNC measurements. Results of the intercomparison revealed that RNCs, 15 in total, are accurate within 10% in vial geometry and within 15% in syringe geometry. Most of them showed similar performance. The results revealed that container geometry is an important influencing parameter in the accuracy of activity measurement. Obtained results indicate a need for regular calibration and implementation of Quality Control program in order to achieve and maintain the accuracy of activity measurements in nuclear medicine.
