A cumarina (1,2-benzopirona) e um composto aromatico encontrado em varias especies vegetais. Este... more A cumarina (1,2-benzopirona) e um composto aromatico encontrado em varias especies vegetais. Este trabalho objetivou avaliar as acoes da cumarina em modelos comportamentais de ansiedade, depressao e sedacao, tais como, campo aberto, rota rod, labirinto em cruz elevado (LCE), placa perfurada e suspensao da cauda, procurando esclarecer os mecanismos atraves de doseamento de aminoacidos em cortex pre-frontal e hipocampo de camundongos atraves de HPLC (High Perfomance Liquid Chomatography). Foram utilizados camundongos albinos, variedade Swiss Webster, adultos, machos, pesando entre 25-30 g, provenientes do Bioterio do Departamento de Fisiologia e Farmacologia da UFC. A cumarina (CUM) foi administrada de forma aguda por via intraperitoneal nas doses de 5, 20 ou 40 mg/kg. Os resultados mostram que a CUM, em todas as doses utilizadas, diminuiu a atividade locomotora, o numero de rearing e grooming no teste do campo aberto, sugerindo uma possivel acao sedativa. C. Smith andAnadenanthera macrocarpa(Benth.) Brenan, combined with six antimicrobial drugs against multiresistant strains ofStaphylococcus aureusandEscherichia coliisolated from humans. The antibacterial activity of the extracts was determined using the minimum inhibitory concentration (MIC). The microdilution assay was performed to verify the interactions between the natural products and the antibiotics using a subinhibitory concentration. The activity of amikacin associated with the extract ofAnadenanthera macrocarpaagainst EC 27 was enhanced, demonstrating an MIC reduction from 128 to 4 μg/mL. Among theβ-lactams, no potentiation on its activity was observed, with exception to the antagonism of the natural products with ampicillin againstS. aureus358.

O acidente vascular cerebral isquemico precipita uma cascata de eventos que envolvem excitotoxici... more O acidente vascular cerebral isquemico precipita uma cascata de eventos que envolvem excitotoxicidade, inflamacao, estresse oxidativo e morte celular. A curcumina tem demonstrado efeitos neuroprotetores, com propriedades antioxidantes e antiinflamatorias. Sendo assim, objetivou-se estudar estes efeitos da curcumina em modelo experimental de isquemia cerebral global em ratos em desenvolvimento. Metodos: O tratamento de ratas Wistar foi iniciado 24h apos o nascimento dos filhotes, durante 21 dias, com curcumina (25, 50 ou 100 mg/kg/dia, v.o.) ou agua+Cremophor 1% (10 ml/kg). No 21o dia de vida os ratos foram anestesiados com Ketamina (75 mg/kg, i.p.) e Xilazina 2% (10 m/kg, i.p.) e submetidos a isquemia cerebral global durante 15 min, reperfusao pelo mesmo tempo, seguida de corte da carotida esquerda. 24 h apos iniciou-se o tratamento dos filhotes com curcumina durante 3 ou 7 dias consecutivos. Background The species Himatanthus drasticus is popularly known in Northeast Brazil as "jana... more Background The species Himatanthus drasticus is popularly known in Northeast Brazil as "janaguba" and belongs to the family Apocynaceae. The latex collected from its stem bark is used for several purposes including anti-inflammatory properties and presents among its bioactive constituents the pentacyclic triterpene lupeol. The objective of the present work was to study in vivo and in vitro the lupeol acetate (LA) isolated from the plant latex, in several models of inflammation. Methods Male Swiss mice (25-30 g, 6-24 animals per group) were administered with LA, 30 min before the test initiation. In the evaluation of analgesic activity the formalin test was used. The anti-inflammatory activity was evaluated by the following tests: paw edema induced by carrageenan and dextran, and the carrageenan-induced neutrophil migration into peritoneal cavities. Valproic acid (VA) is an antiepileptic that is also used for the treatment of bipolar disorders. ... more Valproic acid (VA) is an antiepileptic that is also used for the treatment of bipolar disorders. The objective was to evaluate the neuroprotective effects of VA on a brain ischemia model. The groups of male Wistar rats were: SO (sham-operated), ischemic and ischemic treated with VA (25, 50 and 100 mg/kg, p.o.). After anesthesia with ketamine and xilazine, the animals were subjected to clamping of carotid arteries (30 min) and reperfusion. Except for the carotid clamping, the SO group was submitted to the same procedure. On the 7th day, the animals were behaviorally evaluated, euthanized and had their brain dissected for neurochemical and immunohistochemical assays. The data were analyzed by ANOVA and Tukey as the post hoc test. The results showed that VA reversed partly or completely the behavioral (locomotor activity and memory deficits), neurochemical (striatal DA and DOPAC levels, brain nitrite and lipid peroxidation) and immunohistochemical alterations (iNOS, COX-2, HDAC and GSK3) observed in the untreated ischemic group. VA neuroprotective effects are probably related to its anti-inflammatory and antioxidant properties, as well as to HDAC and GSK3 inhibitory effects. These findings stimulate translational studies focusing on VA as a neuroprotective drug to be potentially used in the clinic for several neurological conditions.
