The plasma membrane guard cell slow anion channel is a key element at the basis of water loss con... more The plasma membrane guard cell slow anion channel is a key element at the basis of water loss control in plants allowing prolonged osmolite efflux necessary for stomatal closure. This channel has been extensively studied by electrophysiological approaches but its molecular identification is still lacking. Recently, we described that this channel was sharing some similarities with the mammalian ATP-binding cassette protein, cystic fibrosis transmembrane conductance regulator (CFTR) chloride channel . Here, using the patch-clamp technique and a bioassay, consisting in the observation of the change in guard cell protoplasts volume, we demonstrated that a functional antibody raised against the mammalian CFTR prevented ABA-induced guard cell protoplasts shrinking and partially inhibited the slow anion current. Moreover, this antibody immunoprecipitated a polypeptide from guard cell protein extracts and immunolabeled stomata in Vicia faba leaf sections. These results indicate that the guard cell slow anion channel is, or is closely controlled by a polypeptide, exhibiting one epitope shared with the mammalian CFTR. ß

FRY1 encodes a nucleotide phosphatase that in vitro has inositol polyphosphate 1-phosphatase and 39,(29),59-bisphosphate nucleotide phosphatase activities. It is not clear which activity mediates each of the diverse biological functions of FRY1 in planta.

Plants adapt to different environmental conditions by constantly forming new organs in response t... more Plants adapt to different environmental conditions by constantly forming new organs in response to morphogenetic signals. Lateral roots branch from the main root in response to local auxin maxima. How a local auxin maximum translates into a robust pattern of gene activation ensuring the proper growth of the newly formed lateral root is largely unknown. Here, we demonstrate that miR390, TAS3-derived trans-acting short-interfering RNAs (tasiRNAs), and AUXIN RESPONSE FACTORS (ARFs) form an auxin-responsive regulatory network controlling lateral root growth. Spatial expression analysis using reporter gene fusions, tasi/miRNA sensors, and mutant analysis showed that miR390 is specifically expressed at the sites of lateral root initiation where it triggers the biogenesis of tasiRNAs. These tasiRNAs inhibit ARF2, ARF3, and ARF4, thus releasing repression of lateral root growth. In addition, ARF2, ARF3, and ARF4 affect auxin-induced miR390 accumulation. Positive and negative feedback regulation of miR390 by ARF2, ARF3, and ARF4 thus ensures the proper definition of the miR390 expression pattern. This regulatory network maintains ARF expression in a concentration range optimal for specifying the timing of lateral root growth, a function similar to its activity during leaf development. These results also show how small regulatory RNAs integrate with auxin signaling to quantitatively regulate organ growth during development.

The cDNA is 5.5 kb long and contains an ORF encoding a 1623 amino acids protein. This sequence is the first MRP-like protein found in plants. © 1998 Elsevier Science B.V.

Among the complete set of Arabidopsis ABC proteins, 15 correspond to the soluble or NBD-type (nuc... more Among the complete set of Arabidopsis ABC proteins, 15 correspond to the soluble or NBD-type (nucleotide binding domains). We have cloned cDNAs coding for three closely homologous NBD-like proteins, AtNAP2, AtNAP4 and AtNAP9 that display a characteristic ABC signature motif. In transitory expression assays, the three AtNAP proteins fused to the green-fluorescent protein (GFP) were localized in the cytosol of tobacco and Arabidopsis cell cultures. The expression of these genes is enhanced by 1% sucrose in the culture medium but not by non-assimilable sugars. This result was confirmed with an AtNAP2::GUS translational fusion derived from a promoter trap line in which the T-DNA is inserted near the 3 0 end of the AtNAP2 coding sequence. According to the GUS staining, AtNAP2 is expressed in the elongation zone and meristem of roots of white lightgrown seedlings and in the hypocotyl of dark-grown seedlings. We suggest that AtNAP2 might be involved in cell elongation processes. #

Molecular chaperones of the heat shock cognate 70 kDa (HSC70) family are highly conserved in all ... more Molecular chaperones of the heat shock cognate 70 kDa (HSC70) family are highly conserved in all living organisms and assist nascent protein folding in normal physiological conditions as well as in biotic and abiotic stress conditions. In the absence of specific inhibitors or viable knockout mutants, cytosolic/nuclear HSC70-1 overexpression (OE) and mutants in the HSC70 co-chaperone SGT1 (suppressor of G 2 /M allele of skp1) were used as genetic tools to identify HSC70/SGT1 functions in Arabidopsis development and abiotic stress responses. HSC70-1 OE caused a reduction in root and shoot meristem activities, thus explaining the dwarfism of those plants. In addition, HSC70-1 OE did not impair auxin-dependent phenotypes, suggesting that SGT1 functions previously identified in auxin signalling are HSC70 independent. While responses to abiotic stimuli such as UV-C exposure, phosphate starvation, or seedling de-etiolation were not perturbed by HSC70-1 OE, it specifically conferred g-ray hypersensitivity and tolerance to salt, cadmium (Cd), and arsenic (As). Cd and As perception was not perturbed, but plants overexpressing HSC70-1 accumulated less Cd, thus providing a possible molecular explanation for their tolerance phenotype. In summary, genetic evidence is provided for HSC70-1 involvement in a limited set of physiological processes, illustrating the essential and yet specific functions of this chaperone in development and abiotic stress responses in Arabidopsis.
