Introduction: The colonic epithelium is a classical aldosterone target, but the effect of the hor... more Introduction: The colonic epithelium is a classical aldosterone target, but the effect of the hormone on the oxygen consumption rate (QO2 ) of this tissue is unknown. Objectives. We aimed at assessing, in the rectal epithelium of rats fed with diets of different sodium content, the effect of epithelial sodium channel (ENaC) blockade on short-circuit current (ISC ) and QO2 , and the acute effect of aldosterone incubation on ISC and QO2 . Methods: Adult male rats were fed with normal, low or high-sodium diets for 8 days. Plasma sodium and serum aldosterone were measured. Isolated mucosa preparations from the rectal portion of the colon were mounted in Ussing chambers modified to measure ISC and QO2. Results: Baseline ISC and QO2 were highest in sodium-deprived rats. Both were proportionally reduced by amiloride (0.1 mM) in this group and in the normal sodium group, but not in sodium-loaded rats. In separate experiments, incubation with aldosterone (10 nM) for 7 h increased ISC and QO2 in all groups; increases were larger in the normal and sodium-loaded groups. Amiloride decreased both ISC and QO2 , abolishing the differences between groups. Linear regression of the decrease in QO2 and ISC after amiloride showed the steepest slope for the sodium-deprived group and the flattest one for the sodium-loaded group. Conclusions: Baseline epithelial QO2 of sodium-deprived and control rats is reduced by ENaC blockade. Aldosterone increased QO2 proportionally to ISC augmentation in all groups, but the coupling between aerobic metabolism and electrogenic transport seems more efficient in sodium-deprived animals.

Hasta comienzos del siglo XIX, el examen físico del paciente tenía un lugar secundario en el diag... more Hasta comienzos del siglo XIX, el examen físico del paciente tenía un lugar secundario en el diagnóstico médico. El enfoque diagnóstico comenzó a cambiar a partir del método anátomo-clínico desarrollado principalmente en la Escuela de Medicina de París. En la revolución allí iniciada tuvo un papel muy destacado la invención del estetoscopio por René Théophile Hyacinthe Laennec (1781-1826). Este es un relato de la vida y circunstancias de Laennec desde su nacimiento hasta el descubrimiento de la auscultación mediata por medio del estetotoscopio. En la segunda parte me referiré a la obra principal de Laennec y su impacto en la práctica de la medicina desde esta invención hasta el fallecimiento de su inspirado autor.

Osteocalcin (OC) plays a crucial role in glucose homeostasis. OC levels in the circulation appear... more Osteocalcin (OC) plays a crucial role in glucose homeostasis. OC levels in the circulation appear to be associated with glucose and fat metabolism; however, still remains controversial if it is total OC, undercarboxilated OC (ucOC) or both, which presents such association. We determined ucOC, total OC, leptin and insulin, in 226 adult non-diabetic women, having levels of glucose (80 to 110 mg/dL) and hemoglobin A1c (HbA1c b 5.7%). The presence of overweight or obesity I and II degree was determined according to BMI index. Glucose, cholesterol, triglycerides, HbA1c levels were measured by standard laboratory methods; OC (ng/mL), ucOC (ng/mL), leptin (ng/mL), insulin (uUI/L) levels by ELISA and 25hydroxyvitamin D (25OHD) (ng/mL) levels by a competitive protein-binding method. Results (mean ± SD and *: p b 0.05 vs. overweight; (**) p b 0.05 vs obesity I degree) in the next order: ucOC; OC; insulin; leptin.

According to the classical paradigm, biological effects of ionizing radiation are attributed to D... more According to the classical paradigm, biological effects of ionizing radiation are attributed to DNA damage induced in each irradiated cell. Demonstration of ionizing radiation-induced bystander effects (RIBE) has generated a deep change in current understanding of radiobiology. RIBE are radiation-induced effects produced in cells that have not been actually irradiated. Several technical advances, particularly the use of microbeams, allowed in vitro study of RIBE. There are two known ways by which irradiated cells can communícate with non-irradiated cells, namely: through gap junctions connecting the cytoplasms of adjacent cells, and through the secretion of soluble factors to the extracellular medium. These factors include several cytokines and reactive species of oxygen and nitrogen. In the affected cells, signalling pathways mostly involve activation of mitogen-activated protein kinases (MAPK), NFtranscription factor and of the enzymes cyclooxygenase 2, nitric oxide synthase 2 and NAD(P)H oxidase. RIBE induce point mutations and epigenetic changes. Effects on cellular signalling pathways can persist indefinitely and even be transmitted to the progeny of affected cells. Paradoxically, under certain conditions RIBE may be adaptive, which means that they turn affected cells more resistant to ionizing radiation. Adaptation demands protein synthesis. It enhances DNA repair mechanisms and resistance to oxidative stress. RIBE have also been demonstrated in vivo. Thus, they may have important implications for radiotherapy, both to improve therapeutic efficacy and to reduce the incidente of adverse effects. Furthermore, a better understanding of RIBE may have an influence on international radioprotection standards.
