PHARMACOL BIOCHEM BEHAV 21(2) 177-180, 1984.-The effects of 3 beta-adrenergic agonists (clenbuter... more PHARMACOL BIOCHEM BEHAV 21(2) 177-180, 1984.-The effects of 3 beta-adrenergic agonists (clenbuterol, isoproterenol and salbutamol) on the spontaneous motor activity of mice were studied. The present research indicated that motor activity was significantly decreased 30 minutes after IP injection of either clenbuterol (0.06 mg/kg), isoproterenol (0.5 mg/kg) or salbutamol (2 mg/kg). Hypomotility induced by clenbuterol was also significantly antagonized by propranolol in doses ranging from I to 8 mg/kg and by penbutolol in doses from 0.03 to 0.5 mg/kg. However, practolol, which does not cross the blood brain barrier, did not antagonize the effect of clenbuterol. Therefore, it may be hypothesized that beta adrenergic agonists decrease motor activity by a central mechanism. It was also found that tachyphylaxis or resistance to treatment, observed in cardiovascular and bronchopulmonary systems with beta-adrenergic agonists, developed after 7 injections of clenbuterol (0.25 mg/kg IP, twice daily) in the behavioral model of spontaneous motor activity in mice.

The effect in mice of chronic administration of the beta-adrenergic agonist clenbuterol was studi... more The effect in mice of chronic administration of the beta-adrenergic agonist clenbuterol was studied using four pharmacological tests, two of which are predictive of antidepressant activity in man. The suppressive effect of acute administration of the beta-adrenergic agonist in two tests unrelated to antidepressant activity, locomotor activity and investigation of food, was attenuated after chronic treatment. However, in the two tests which are predictive of antidepressant activity, antagonism of reserpine induced hypothermia and potentiation of yohimbine toxicity, the effect of clenbuterol was greater following chronic treatment.

Clenbuterol, a β‐adrenergic agonist known to enter the brain, has been studied in mice with a set... more Clenbuterol, a β‐adrenergic agonist known to enter the brain, has been studied in mice with a set of psychopharmacological tests. Clenbuterol has an ambiguous sedative effect: it reduces motor activity in a dose‐dependent manner without modifying exploratory behavior in the open field; it does not potentiate the effect of pentobarbital. Clenbuterol decreases the search for food in starved mice. Regarding the tests in animals which are expected to reflect antidepressant activity in man (hypothermias induced by reserpine, oxotremorine or a high dose of apomorphine, potentiation of yohimbine toxicity) clenbuterol acts at doses much lower than those required with two other β‐adrenergic agonists, isoproterenol and salbutamol. Moreover, the active doses of clenbuterol and salbutamol being nearly the same at a peripheral level, the ratio of the centrally and peripherally active doses is highly in favor of clenbuterol. This suggests that the effects of clenbuterol on depressed patients should be studied.

One component of control programmes to eliminate trachoma is the treatment of Chlamydia trachomat... more One component of control programmes to eliminate trachoma is the treatment of Chlamydia trachomatis infection. A diagnosis of trachoma is based on clinical grounds, but the signs of active trachoma do not always correlate with the presence of C. trachomatis. During a therapeutic trial, the level of C. trachomatis infection in children with active trachoma in Guinea and Pakistan was assessed using a qualitative commercially available PCR that targeted the C. trachomatis plasmid. The influence of the quality of specimens on the efficiency of the PCR was investigated using two quantitative real-time PCRs targeting the specific omp1 gene of C. trachomatis and human chromosomal DNA, respectively. C. trachomatis was detected in c. 23% of children (aged 1-10 years) who presented with clinically active trachoma. Controls showed that PCR-related problems did not influence this detection rate. For 14% of the positive samples, C. trachomatis was detected in only one eye, with a significantly lower mean load of bacteria. These results suggest that epidemiological and therapeutic surveys should be conducted by sampling and testing both eyes. Moreover, the high variability of the cell load observed in the conjunctival swabs suggests that the effectiveness of swabbing may be questionable.

Une femme âgée de 68 ans s'est présentée aux urgences de l'hôpital avec une canaliculite (tuméfac... more Une femme âgée de 68 ans s'est présentée aux urgences de l'hôpital avec une canaliculite (tuméfaction indolore de la paupière supérieure droite) qui avait débuté plusieurs semaines auparavant. Lors de la canaliculotomie postérieure, des concrétions solides ont été extraites, leur culture a permis l'isolement du Fusobacterium nucleatum sp. nucleatum (bactérie anaérobie). L'extraction chirurgicale des concrétions, associée à un traitement per os par amoxicilline + acide clavulanique et tobramycine topique, ont permis une régression des signes cliniques en deux semaines et leur disparition complète deux mois après.

Objective: Trachoma (Chlamydia-triggered blinding infection) provoked irreversible visual impairm... more Objective: Trachoma (Chlamydia-triggered blinding infection) provoked irreversible visual impairment in about 8 million people in 2011, and the prevalence among children with dirty faces is more than three fold that among children with clean faces. In 250 villages with a high prevalence of trachoma (Kolofata district, Far North Region, Cameroon), the lack of water for facial cleanliness was reported during trachoma awareness campaigns. The objective of this study was to determine if the lack of water was linked with the absence of means to dig wells. Methods: Wells, waterholes, motorcycles, irrigation pumps, electricity, goats and oxen, cell phones and distance from waterholes were recorded in January 2011 in 50 randomized villages of Kolofata's district. Results: The number of villages with <25 goats and <5 oxen was 0 and the number of adults owning <1 goat was 0. The cost of a pail of water was 0.01 USD. Motorcycles, cell phones and televisions have been reported in more than 66% of villages. The cost for the construction of lined shaft wells ranged between 15-35 goats and 0.5-3 oxen; the cost for drinking water wells ranged between 50-200 goats and 3-30 oxen. Discussion: No link between the means for digging wells at the village level and access to water was found. Social solidarity, which refers to a social debt owed by each person to his/her group, should be added to training guides to gauge its ability to release people from the dead end of having to wait for external assistance to gain access to water.
Helicobacter pylori infection is most prevalent and known to cause chronic gastritis and peptic u... more Helicobacter pylori infection is most prevalent and known to cause chronic gastritis and peptic ulcer disease in Ethiopia. To date, simple and rapid point-of-care tests are commercially available; however, information is limited regarding their diagnostic significance. This cross-sectional study was conducted to evaluate the diagnostic performance of SD BIOLINE H. pylori rapid antibody test. A consecutive 203 enzyme linked immunosorbent assay (ELISA) confirmed sera (148 H. pylori positive and 55 negative) from dyspeptic patients were tested using SD BIOLINE H. pylori kit (Standard Diagnostic Inc, Korea) at Hawassa Teaching and Referral Hospital, southern Ethiopia from October, 2012 to January, 2013. Individuals under 15 years old, who were on anti-H. pylori treatment during the month prior to the study, those with discordant ELISA results, and refused to participate, were excluded. The sensitivity, specificity, positive predictive value, negative predictive value, and accuracy of the SD BIOLINE H. pylori test kit were 95.3, 94.5, 97.9, 88.1 and 95.1%, respectively. Therefore, this point-of-care test could be used as alternative to ELISA testing and best fit our context.

Methodologie/Principaux resultats La population du district tout entier (120 000 personnes) a ete traite avec de l'azithromycine collyre 1,5 % en fevrier 2008, janvier 2009 et janvier 2010. Pour evaluer l'effet du traitement sur la prevalence du trachome actif, quatre etudes epidemiologiques ont ete menees sur un echantillon representatif d'enfants âges de 1 a 10 ans. La premiere etude a ete realisee juste avant le premier traitement, la deuxieme juste avant le deuxieme traitement, la troisieme juste avant le troisieme traitement et la quatrieme juste un an plus tard. Terminology of fungus is confusing and adds to the difficulty. Current management is still based on traditional diagnosis and limited options of treatment. Fortunately, there are newer ways to diagnose and treat fungal keratitis which I call new horizons to look for in fungal keratitis.

Diagnosis of Acanthamoeba by microscopic examination, culture, and polymerase chain reactions (PC... more Diagnosis of Acanthamoeba by microscopic examination, culture, and polymerase chain reactions (PCRs) has several limitations (sensitivity, specificity, lack of detection of several strains, cost of testing for discrimination among strains). We developed a new high-resolution melting real-time PCR (HRM) to detect and characterize Acanthamoeba infections. HRM performances were evaluated with strains from the American Type Culture Collection (ATCC) and with 20 corneal scrapings. The DNA extracted from specimens were amplified, detected, and characterized in 1 run using 2 original primers diluted in a solution containing an intercalating dye. Detection and molecular characterization of Acanthamoeba infections could be achieved in less than 2.5 h with a dramatic reduction in cost of reactants (postamplification procedures and radioactive or fluorescentlabeled molecular probes were unnecessary). HRM detection limits were 0.1 cyst/μL or less (including genotypes T5 and T11), and its sensitivity and specificity were higher than other molecular tests. For the tested strains from the ATCC, the HRM drafted 4 different profiles: Type I (genotypes T2 and T4), Type II (T5 and T7), Type III (T8), and Type IV (T1, T3, T6, T9, T11, T12, and T13).
