The viability testing that usually used on sesame seeds is germination testing. The aim of germination testing is to detect seeds germination viability under optimum condition. This research has purpose to determine the correlation between germination, vigour index, and maximum growth potential of sesame seeds. Seeds that used in this research were Sumberejo 4 varieties which have been stored for 6 and 7 years. Seed germination testing was held based on ISTA Rules method. The objectives of this research was to determine the radiosensitivity of Argomulyo variety of soybeans ( Glycine max (L) Merr ) by gamma ray irradiation and to estimate genetic variablity of the crop after irradiation. To determine the radiosensitivy, the seeds of Argomulyo variety w ere treated with gamma ray irradiation at 200 Gy, 400 Gy, 600 Gy, 800 Gy, and 1000 Gy. Curve-fit a nalysis was employed t o find the Lethal Doses 50 (LD 50 ) value . To evaluate the genetic variablity after irradiation with gamma ray, seeds of Argomulyo was irradiated under LD 50 at doses of 0 Gy, 50 Gy, 100 Gy, 150 Gy, and 200 Gy. Durian fruit is also called the king of fruits are very popular among various societies because of its peculiar taste. This research aimed to identify the morphological characteristic and phylogenetic relationship of durian in Sidikalang and Tanah Pinem district. The research was done at Sidikalang and Tanah Pinem district in November 2016 to February 2017 with survey method using durian descriptor of IPGRI. Sampling technique using purposive sampling with location judgement . The result of research showed that in Sidikalang and Tanah Pinem district there were 25genotipe of durian been identified. The closest relationship is obtained on G14 and G15 originating from Tanah Pinem District with a similarity value of 0,985. The furthest relationship was obtained from G13 and G15 from Tanah Pinem District with the similarity value of 6,115.

M3 generation population was thought to have wide character variability. The objective of this study was to determine the performance of agronomic characters and to estimate genetic parameters values of M3 generation in mutant populations of 150 Gy and 450 Gy. Seeds from mutant population 150 Gy and 450 Gy in M2 generation were harvested, and then their seeds were grown as M3 generation. Variations that were obtained of each character at generation M3 influences plants growth and development. The difference in character variability from the two mutant putative populations causes significant differences compared to the mean value character of control plants population. The heritability values of all characters observed range from low to high. Character canopy diameter and number of fruits per plant can be used as a character of selection in the next generation

One of the plant breeding techniques to obtain tolerant plants to salinity is the use of chemical mutations, such as colchicine. The study aimed to reveal the effect of salinity on the characters of soybean plants (mutated with colchicine) and the effect of colchicine on the number of chromosomes and ploidy level. This study was conducted from January to March 2020 and carried out in the plastic house of Faculty of Agriculture, Universitas Sumatera Utara. The experiment was arranged in a Randomized Block Design with salinity levels (0 dS.m-1, 2 dS.m-1, 4 dS.m-1, 6 dS.m-1) as treatments and consisted of ten replications in each treatment. The aim of the research was to determine the influence of oryzalin concentration on the two tomat... more The aim of the research was to determine the influence of oryzalin concentration on the two tomato varieties (Var. Permata and Tymoti). The research was conducted in Agricultural Faculty’s screen house, Universitas Sumatera Utara. The research was used Factorial Randomized Block Design with the first factor was Tomato varieties (Var. Permata and Tymoti), and the second was concentration of Oryzalin (0 µM as control, 30 µM, 60 µM, 90 µM and 120 µM). Parameters observed 30 days after sowing were the plant height, leaf number and the stem diameter of both tomato varieties. The results showed that there was no variation between var. Permata and Tymoti in parameters observed. However, there were differences among plants growth treated plants by Oryzalin concentration. In general, the higher the concentration of oryzalin, the shorter the plant height, the fewer leaf number and the smaller stem diameter for both tomato varieties.

The results showed that there are several types and numbers of soybean mutant in M 2 generation such as chlorophyll mutants (viridis, xantha, waxy and variegata), leaflet mutants (unifoliate, bifoliate, quadrifoliate, pentafoliate, narrow-rugose leaflet, wrinkled leaf) and sterile mutants (undeveloped rasim flower, half sterility and full sterility). Some types of mutants in M 2 generation clearly show that gamma-ray irradiation can be effectively used to assemble genetic diversity in plants.

Research using randomized block design with four replications, the first factor consists of soybean varieties: Tanggamus varieties, Detam 2, Anjasmoro and Detam 1, while the second factor is the media's treatment consisted of medium acid soils and limed soil. The results showed that the low level acidity of planting medium will affect the growth and development of plants. There are different mechanisms of adaptation to acidity on soybean varieties. Avoidance mechanism is indicated by an increase in pH around the roots on Tanggamus varieties, Detam2, Anjasmoro and Detam1. The research was conducted at Desa Tanjung Selamat, Kecamatan Sunggal, Kabupaten Deli Serdang from July to December 2016. The research was arranged by Randomized Block Design for one variety (Adira-1) and 6 genotype (Valencia, Malang, Malaysia, Lampung, and Putih) each treatment was replicated 4 t imes.The result showed that Pulut genotype has the highest average plant heigth (277,48 cm) that is significantly different from the others accept Malang genotype, Malang genotype has the highest average diameter (31,26 mm) wich is significantly different from the others. Penelitian ini bertujuan untuk mengetahui pengaruh kolkisin terhadap keragaman fenotipe tanaman A... more Penelitian ini bertujuan untuk mengetahui pengaruh kolkisin terhadap keragaman fenotipe tanaman Aglaonema. Penelitian ini dilaksanakan di rumah kasa dan Laboratorium Kultur Jaringan, Fakultas Pertanian USU Medan,dari bulan Juni sampai November 2016. Penelitian dilakukan dengan cara perendaman bahan tanam setek secara keseluruhan dengan 4 konsentrasi yakni 0ppm, 2ppm, 4ppm dan 6ppm.Hasil penelitian menunjukkan tanaman Aglaonema hasil perlakuan kolkisin 6ppm berpengaruh sangat nyata pada peubah amatan panjang daun terpanjang. Hasil penelitian menunjukkan tanaman Aglaonema hasil perlakuan kolkisin memiliki karakter morfologis dan kromosom yang berbeda dibandingkan dengan kontrol. Karakter fenotipe paling baik dihasilkan pada dosis kolkisin 6ppm, dan perlakuan konsentrasi kolkisin mampu menghasilkan karakter fenotipe dan kromosom yang aneh dan berbeda.

Soybean is one of the leading strategic food crop in Indonesia. Nowadays, people's need for soybe... more Soybean is one of the leading strategic food crop in Indonesia. Nowadays, people's need for soybeans is not only seen from the production aspects but also soybean nutrient contents, especially fatty acids. One of the ways to develop superior varieties of soybean with high fatty acids is through mutations. Seven soybean mutants selected M5 and M7 generation with high production character and resistance Athelia rolfsii Curz were genetically related to high fatty acids using SSR markers. DNA quality and quantity test results showed DNA purity between 1.12-2.13 with concentration between 15.4-97.9 μg/mL. The FAD2-2D locus is the gene candidate of the result by the QTL mapping used in this study to identify soybean genes related to linoleic acid characters. The results showed that the primer used was specific and polymorphic. Mutant soybeans tested have a high genetic diversity.

(Merrill) in M5 generation based on high production character and tolerance of stem rot disease Athelia rolfsii (Curzi). This research was conducted in Plant Disease Laboratory and experimental field Faculty of Agriculture Universitas Sumatera Utara Medan, Indonesia. This research was conducted from December 2016 to June 2017. The treatments were 15 mutant lines genotypes and Anjasmoro variety. The results showed that some lines mutant genotypes can gave the good agronomic appearance character than Anjasmoro variety on inoculation treatment of stem rot disease. Selection performed on population M 5 producesselected individuals with tolerance of stem rot disease from 100 and 200 Gy population.

The results showed that there are several types and numbers of soybean mutant in M 2 generation such as chlorophyll mutants (viridis, xantha, waxy and variegata), leaflet mutants (unifoliate, bifoliate, quadrifoliate, pentafoliate, narrow-rugose leaflet, wrinkled leaf) and sterile mutants (undeveloped rasim flower, half sterility and full sterility). Some types of mutants in M 2 generation clearly show that gamma-ray irradiation can be effectively used to assemble genetic diversity in plants.

Research using randomized block design with four replications, the first factor consists of soybean varieties: Tanggamus varieties, Detam 2, Anjasmoro and Detam 1, while the second factor is the media's treatment consisted of medium acid soils and limed soil. The results showed that the low level acidity of planting medium will affect the growth and development of plants. There are different mechanisms of adaptation to acidity on soybean varieties. Avoidance mechanism is indicated by an increase in pH around the roots on Tanggamus varieties, Detam2, Anjasmoro and Detam1. Tolerant mechanism is indicated by the maturation age and high production on Tanggamus varieties

Penelitian ini bertujuan untuk mengetahui pengaruh kolkisin terhadap keragaman fenotipe tanaman A... more Penelitian ini bertujuan untuk mengetahui pengaruh kolkisin terhadap keragaman fenotipe tanaman Aglaonema. Penelitian ini dilaksanakan di rumah kasa dan Laboratorium Kultur Jaringan, Fakultas Pertanian USU Medan,dari bulan Juni sampai November 2016. Penelitian dilakukan dengan cara perendaman bahan tanam setek secara keseluruhan dengan 4 konsentrasi yakni 0ppm, 2ppm, 4ppm dan 6ppm.Hasil penelitian menunjukkan tanaman Aglaonema hasil perlakuan kolkisin 6ppm berpengaruh sangat nyata pada peubah amatan panjang daun terpanjang. Hasil penelitian menunjukkan tanaman Aglaonema hasil perlakuan kolkisin memiliki karakter morfologis dan kromosom yang berbeda dibandingkan dengan kontrol. Karakter fenotipe paling baik dihasilkan pada dosis kolkisin 6ppm, dan perlakuan konsentrasi kolkisin mampu menghasilkan karakter fenotipe dan kromosom yang aneh dan berbeda.

Nowadays, people's need for soybeans is not only seen from the production aspects but also soybean nutrient contents, especially fatty acids. One of the ways to develop superior varieties of soybean with high fatty acids is through mutations. Seven soybean mutants selected M5 and M7 generation with high production character and resistance Athelia rolfsii Curz were genetically related to high fatty acids using SSR markers. DNA quality and quantity test results showed DNA purity between 1.12-2.13 with concentration between 15.4-97.9 μg/mL. The FAD2-2D locus is the gene candidate of the result by the QTL mapping used in this study to identify soybean genes related to linoleic acid characters. The results showed that the primer used was specific and polymorphic. Mutant soybeans tested have a high genetic diversity.

(Merrill) in M5 generation based on high production character and tolerance of stem rot disease Athelia rolfsii (Curzi). This research was conducted in Plant Disease Laboratory and experimental field Faculty of Agriculture Universitas Sumatera Utara Medan, Indonesia. This research was conducted from December 2016 to June 2017. The treatments were 15 mutant lines genotypes and Anjasmoro variety. The results showed that some lines mutant genotypes can gave the good agronomic appearance character than Anjasmoro variety on inoculation treatment of stem rot disease. Selection performed on population M 5 producesselected individuals with tolerance of stem rot disease from 100 and 200 Gy population.
