The aim of this study was to investigate the phenotypic variations in birth and body weights at v... more The aim of this study was to investigate the phenotypic variations in birth and body weights at various ages of local sheep, total of 37 heads, born as singletons n =13, twins n = 19, triplets n =5. The lambs were reared at two locations Purwakarta and Jatinangor under extensive feeding system and grazing. All animals were weighed monthly till six months of age by hang scale (kg). Data analyzed by SAS (version 9.0). Means ± SE for the birth and body weights were performed. Analysis of variance GLM was done to examine the effects of sex, type of birth and location on the measured variables. Results shown that birth weight ranged between 1.50 to 3.50 kg, the overall means of the type of birth are: single, twins and triplets were 2.69±0.12, 2.24±0.11 and 1.96±0.20 respectively, male weight were 2.355±0.122, females were 2.220±0.126, also birth weight at Purwakarta is of 2.218±0.106 and Jatinangor is of 2.447±0.156. The aim of study was to explore the possibility of genetic diversity within Batur sheep to find i... more The aim of study was to explore the possibility of genetic diversity within Batur sheep to find indirect parameters for meat production that potentially could be used for selective purposes. The trial was carried out at Banjarnegara under the control of 13 owners with total 69 heads aged between 0.5 and 3 years, depending on age group: 6 months, year and adult. Body weight was recorded in the kilogram, whereas, body measures were taken by tape in the centimeter were Height at withers, Chest Girth and Body Length. Data were analyzed using simple descriptive analyses, GLM, and Correlation procedures by SPSS version 16. Body weight of three age groups were 27.28±1.76, 48.87±1.55 and 69.10a±2.80 kg respectively. Body weight measured monthly after weaning until six months of age. DNA Extraction used 200 ul of whole blood samples. To amplify the exon 3 region of MSTN gene a specific primer designed using the Primer3 software. Volume 25 μl contained 25 ng genomic DNA, with a 2x Reaction mix 12.5 μl for each primer. The PCR cycling protocol was 35 cycles with denaturation at 94 °C, 73.9 annealing for 45 sec, extended at 72°C, with a final extension at 72°C. Eleven polymorphic sites were observed in the third exon region transversions at c.*121 G instead of A, del-T at c.*129, one individual at c.*139 and one individual at c.*158 positions, however, one individual sequence disrupted reading frame in whole MSTN sequenced, MSTN gene was polymorphic in Batur sheep, however, frequency of allele A compare to allele B is low in diversity, this gene may not be a marker-assisted in the future selection program.

The present study aimed to investigate the association between myostatin (MSTN) genotype and two ... more The present study aimed to investigate the association between myostatin (MSTN) genotype and two traits, growth and carcass, in Batur sheep. Blood samples representing thirty head were collected and genomic DNA was extracted. A specific primer designed to amplify the MSTN gene, samples sequenced then, used the BioEdit program to identify any mutation. Calculation of genotypes, gene and allele frequencies, heterozygosities, and Chi-square test was performed. Only two alleles observed (G and C) resulted in three genotypes. 11 polymorphic sites were observed, transversion at c.*121G>C, and one individual G>A which disrupted reading frame of whole MSTN sequenced, genotypic and allelic frequencies were 0.552 GG, 0.379 GC, and 0.069 CC, where the allele frequency was 0.741 G and 0.259 C. Lambs that carrying genotype GC had slightly more pre-slaughter weight, hot carcass weight, cold carcass weight, carcass length, and total yield (lion, shoulder, leg yield, and thigh) compared to those carrying genotype GG. There was no significant effect of the MSTN genotype on carcass traits (p=0.05). Polymorphic site c.*121 G>C is present in Batur sheep for the first time about the association with the MSTN gene however, it has not effects weaning weight, 6-month weight, and studied carcass traits.
