The genome of Entamoeba histolytica encodes approximately 50 Cysteine Proteases (CPs) whose activ... more The genome of Entamoeba histolytica encodes approximately 50 Cysteine Proteases (CPs) whose activity is regulated by two Inhibitors of Cysteine Proteases (ICPs), EhICP1 and EhICP2. The main difference between both EhICPs is the acquisition of a 17 N-terminal targeting signal in EhICP2 and three exposed cysteine residues in EhICP1. The three exposed cysteines in EhICP1 potentiate the formation of cross-linking species that drive heterogeneity. Here we solved the NMR structure of EhICP1 using a mutant protein without accessible cysteines. Our structural data shows that EhICP1 adopts an immunoglobulin fold composed of seven β-strands, and three solvent exposed loops that resemble the structures of EhICP2 and chagasin. EhICP1 and EhICP2 are able to inhibit the archetypical cysteine protease papain by intercalating their BC loops into the protease active site independently of the character of the residue (serine or threonine) responsible to interact with the active site of papain. EhICP1 and EhICP2 present signals of functional divergence as they clustered in different clades. Two of the three exposed cysteines in EhICP1 are located at the DE loop that intercalates into the CP substrate-binding cleft. We propose that the solvent exposed cysteines of EhICP1 play a role in regulating its inhibitory activity and that in oxidative conditions, the cysteines of EhICP1 react to form intra and intermolecular disulfide bonds that render an inactive inhibitor. EhICP2 is not subject to redox regulation, as this inhibitor does not contain a single cysteine residue. This proposed redox regulation may be related to the differential cellular localization between EhICP1 and EhICP2.

Toxic epidermal necrolysis is related in 80% of cases to pharmacologic hypersensitivity and systemic consequences may be found; hepatic injury has been described, but the finding of liver abscesses has not been reported among common injuries. The case of a patient with a rapid development of multiple liver abscesses in the clinical setting of hypersensitivity due to lamotrigine and the discussion of probable etiologies and management is presented. En este articulo se presenta una investigacion de nivel licenciatura sobre la importancia de las ... more En este articulo se presenta una investigacion de nivel licenciatura sobre la importancia de las graficas de funciones dentro del curriculo escolar y para la explicacion dentro del concepto de funcion, asi como el desempeno de estudiantes de nivel superior en dos de nueve tareas de construccion e interpretacion de graficas de funciones de nivel secundaria, y modificadas por nosotros. Se evidencia las dificultades presentes en los estudiantes al momento de pasar de una representacion verbal a una grafica: la confusion de la trayectoria de una situacion descrita graficamente como la imagen de la situacion; la discretizacion de variables continuas y, preferencia por la linealidad al momento de construir una grafica que requiere de intervalos concavos. Consideramos que se requiere centrar la atencion en dichos resultados ya que estudiantes de nivel superior obtuvieron resultados similares a estudiantes de nivel secundaria, lo cual puede decirnos que hay un estancamiento en el aprendizaj...

In this paper, the impact of human migration on the dynamics of dengue epidemic has been discusse... more In this paper, the impact of human migration on the dynamics of dengue epidemic has been discussed. The vector-host model considers two patches with different dengue serotype in each patch. The model considers the constant rate of migration in susceptible and recovered class from one patch to other. Recovered migrants from prior infection are exposed to secondary infection in the patch where different serotype is present. The basic reproduction number is computed and analyzed in terms of migration parameters. The model is analyzed for the existence and local stability of various equilibrium states in terms of migration parameters. The numerical simulations for the choice of relevant data from literature have been performed to verify analytical results and to further explore the dynamics of the system. The sensitivity analysis of basic reproduction number with respect to migration parameters is carried out. It is found that immigration in a patch increases the basic reproduction in respective patch and vice-versa. The basic reproduction number has been estimated for the two states of Brazil which verifies the occurrence of severe epidemic in one of the states of Brazil.

Este estudio examina la contribución del bienestar, las expectativas y las actitudes de crianza m... more Este estudio examina la contribución del bienestar, las expectativas y las actitudes de crianza maternos (BEC) al logro escolar de los niños. Su objetivo es el de explorar si estas contribuciones pueden ayudar a explicar las confusas relaciones entre la participación de los padres (PP) y el logro escolar de los niños. Participaron 621 madres y sus niños de primero, segundo y tercer grado escolar, de los cuales 322 eran de sexo femenino y 299 masculino, y estaban distribuidos de manera semejante en las cinco ciudades mexicanas de procedencia. Se llevó a cabo un análisis de conglomerados que generó dos grupos de madres con califi caciones bajas o altas en las medidas de BEC. Los grupos nombrados BEC bajo o alto, mostraron diferencias en las distribuciones de las variables socioeconómicas y, por esto, se formaron seis nuevos grupos tomando en cuenta la infl uencia de estas variables. Los resultados indicaron que tres factores, ingresos familiares, escolaridad materna, y BEC, esta ban relacionados con el logro de los niños, pero se relacionaban pobremente con las medidas de PP. La única excepción importante fue la relación con la medida de promoción de la lectura en los niños. Los hallazgos sugieren que otros factores más que el concepto tradicional de PP tienen efectos diferenciales en el logro académico de los niños.

Dihydropyridomycins 2 and 3, which lack the characteristic enol ester moiety of the potent antimy... more Dihydropyridomycins 2 and 3, which lack the characteristic enol ester moiety of the potent antimycobacterial natural product pyridomycin (1), have been prepared from L-Thr, R-and S-hydroxy isovaleric acid, and 3pyridinecarboxaldehyde. The 2R isomer 2 shows only 4-fold lower anti-Mtb activity than 1, indicating that the enol ester moiety in the natural product is not critical for its biological activity. This finding establishes 2 as a potent and more practical lead for anti-TB drug discovery.

However, posttranslational modifications and the presence of intrinsic disulfide-bonds pose difficulties for their industrial use. The immunoglobulin fold is one of the most ubiquitous folds in nature and it is found in many proteins besides antibodies. An example of a protein family with an immunoglobulin-like fold is the Cysteine Protease Inhibitors (ICP) family I42 of the MEROPs database for protease and protease inhibitors. Members of this protein family are thermostable and do not present internal disulfide bonds. Crystal structures of several ICPs indicate that they resemble the Ig-like domain of the human T cell co-receptor CD8α As ICPs present 2 flexible recognition loops that vary accordingly to their targeted protease, we hypothesize that members of this protein family would be ideal to design peptide aptamers that mimic protein-protein interactions.
