In a hospital-based study of 400 patients with multiple sclerosis (MS), 42 per cent of patients w... more In a hospital-based study of 400 patients with multiple sclerosis (MS), 42 per cent of patients who had had MS for 10 years or more had benign disease. Early age of onset and a long first remission were significantly associated with a good prognosis. There was a suggestion that initial presentation with paraesthesiae and possibly optic neuritis were associated with a benign prognosis, but the only significant finding was the association between limb weakness and a poor outcome (p less than 0.05). Fewer patients with benign disease had a progressive element to their disease than those in the more disabled group (p less than 0.001). The only laboratory test which was associated with a benign prognosis was the absence of CSF myelin basic protein in remission. Abnormalities of visual evoked response, CSF IgG and peripheral blood T lymphocytes appeared to have no value in assessing prognosis in the patients studied.

Adult mononuclear cells were stimulated with pokeweed mitogen (PWM) in the presence of supernatan... more Adult mononuclear cells were stimulated with pokeweed mitogen (PWM) in the presence of supernatants from cord T cells which had been previously stimulated with PWM for 24 h. Increased IgM, IgG and IgA production, as measured by ELISA, was observed with the addition of increasing concentrations of cord T-cell supernatants. The most significant increase in immunoglobulin production was observed with IgM (p < 0.001). Cord T lymphocytes appear to be capable of producing soluble helper factors which augment immunoglobulin production by adult lymphocytes.Copyright © 1985 S. Karger AG, Basel

9 We examined short-term effects of anesthesia and surgery on the lymphocyte response of neonates... more 9 We examined short-term effects of anesthesia and surgery on the lymphocyte response of neonates and older children to a range of mitogens using a whole blood mitogen stimulation assay. Following induction of anesthesia, both neonates and older children demonstrated significantly decreased lymphocyte transformation to phytohaemagglutinin (PHA; P < 0.005) and Con A (P < 0.025) compared with preanesthetic values. Immediately, postsurgery, older children recovered fully their responsiveness to both PHA and Con A, while there was significant depression in their responsiveness to PWM (P < 0.005) and SPA (P < 0.05). In contrast, neonatal responsiveness to all mitogens was significantly reduced (P < 0.005) immediately after surgery compared to preoperative values. At 24-hour postsurgery, neonatal responsiveness to PHA had returned to preoperative levels but Con A, PWM, and SPA responses continued to decline (P < 0.005). These results suggest that the newborn is particularly susceptible to the immunosuppressive effects of anesthesia and surgery.

The proliferative response of splenic lymphocytes in mice bearing the Landschfitz ascites tumour ... more The proliferative response of splenic lymphocytes in mice bearing the Landschfitz ascites tumour has been examined in vitro and correlated with the changes occurring in the growth characteristics of the tumour. Between 8 and 12 days after tumour transplantation the spleen increased markedly in size and this has been shown to be associated with an increased proliferative activity of the splenic lymphocytes. The activity rapidly diminished as the spleen atrophied. Maximal tumour cell DNA synthetic activity also occurred 12 days after transplantation corresponding to the attainment of maximum tumour cell numbers. Although the volume of the ascites fluid continued to increase qfter this time the absolute number of tumour cells fell in parallel with a reduction in D ~ ~ 1 .p, nthesis. The close temporal relationship between the changes in both spleen cell and turnout cell populations suggests that.J'aclor ~ ip!/hwncin/,, cell.~Jo~'/h at, ~ m4in2 .~im,//aneously on both cell popalations.

The growth characteristics of Landschütz ascites tumour in Schofield albino mice has been studied... more The growth characteristics of Landschütz ascites tumour in Schofield albino mice has been studied. Absolute numbers of tumour cells were determined. A pattern of growth similar to that reported for other ascites tumours was observed. No sex related difference in growth rate was found. The merits of the technique utilized are discussed and a correlation is noted between the size of the tumour cell inoculum and the survival time in this model.

A considerable reduction in tumour cell numbers following the transplantation of VCN-treated cells as compared to untreated cells has been shown which is readily reversible by in vitro incubation of the treated cells for a short period prior to transplantation. The possible significance of this is discussed.
