Pseudorabies virus (PRV) ICP22 (P-ICP22) is a homologue of HSV-1 ICP22 and is reported to be able to selectively modify the transcription of different kinetic classes of PRV genes, however, the subcellular localization, localization signal and molecular determinants for its transport to execute this function is less well understood. In this study, by utilizing live cells fluorescent microscopy, P-ICP22 fused to enhanced yellow fluorescent protein (EYFP) gene was transient expressed in live cells and shown to exhibit a predominantly nucleus localization in the absence of other viral proteins. By transfection of a series of P-ICP22 deletion mutants fused to EYFP, a bona fide nuclear localization signal (NLS) and its key amino acids (aa) of P-ICP22 was, for the first time, determined and mapped to aa 41-60 (PASTPTPPKRGRYVVEHPEY) and aa 49-50 (KR), respectively. Research paper thumbnail of Identification of molecular determinants for the nuclear import of pseudorabies virus UL31

Archives of biochemistry and biophysics, Jan 5, 2015

Herpes simplex virus 1 (HSV-1) UL31 is a multifunctional protein and important for HSV-1 infectio... more Herpes simplex virus 1 (HSV-1) UL31 is a multifunctional protein and important for HSV-1 infection. Pseudorabies virus (PRV) UL31 is a late protein homologous to HSV-1 UL31. Previous studies showed that PRV UL31 is predominantly localized to nucleus, however, the molecular determinants for its nuclear import were unclear to date. Here, by utilizing live cells fluorescent microscopy, UL31 fused with enhanced yellow fluorescent protein was transiently expressed in live cells and confirmed to exlusively target to the nucleus in the absence of other viral proteins. Furthermore, the nuclear import of UL31 was found to be dependent on the Ran-, importin α1-, α3-, α5-, α7-, β1-and transportin-1-mediated pathway. Therefore, these results would open up new avenues for depicting the biological functions of UL31 during PRV infection.

The viscosity of fluxes at low stress shows a good correlation with their wetting properties. The modulus of flux residue varies significantly between different fluxes and high modulus flux residue is difficult to remove. The correlation between underfill, substrate properties, and underfill flow voids will be discussed. An underfill flow study with quartz die packages was conducted. The effect of flux residue and substrate wetting properties were assessed by this underfill flow study. This study will help us in minimizing the flux residue effect on underfill flow and optimizing underfill flow process.

However, little is known concerning the subcellular localization signal of PRV UL31. Here, by transfection with a series of PRV UL31 deletion mutants fused to an enhanced yellow fluorescent protein (EYFP) gene, a bipartite nuclear localization signal (NLS) and a PY motif NLS of UL31 were identified and mapped to amino acids (aa) 4 to 20 (RRRLLRRKSSAARRKTL) and aa 21 to 34 (TRAARDRYAPYFAY), respectively. Additionally, the predicted nuclear export signal (NES) was shown to be nonfunctional. Taken together, this information opens up new avenues for investigating the biological functions of UL31 during PRV infection.

Several reports concerned with the clinicopathologic feature have been described, but this disease in puerperium has not been reported previously. Here, we report a 24-year-old woman diagnosed as IgG4-RLD in puerperium, who developed dry cough, low fever and exertional dyspnea following the delivery. The inflammatory markers and pulmonary lesions of the patient suggested pneumonia. However, there was no improvement after antibiotic treatment. The infiltration of IgG4-positive lymphoplasmacytes was found in lung biopsy by video-assisted thoracic surgery (VATS). And the serum IgG4 level was high. The patient was effectively treated with corticosteroids. This unique case highlights the occurrence of IgG4-RLD in puerperium and underscores it should be taken into consideration as a possible differential diagnosis when dense lymphoplasmacytic infiltration was found in pulmonary consolidation in complex puerperal respiratory...

(18)F-FDG PET/CT findings were reviewed in six RP patients. The initial scans were performed for all patients, follow-up scans were performed during steroid therapy for five patients. Changes in the abnormal lesions and the maximal standard uptake value (SUVmax) were analyzed. The initial PET/CT scans revealed intense FDG uptake in the cartilages for all six patients. The lesions of abnormal FDG uptake were tracheal/bronchial cartilage (n = 4), costicartilage (n = 4), nasal cartilage (n = 3), cricoid cartilage (n = 3), auricular cartilage (n = 3), arytenoid cartilage (n = 3), thyroid cartilage (n = 2), hyoid cartilage (n = 1) and mediastinum lymph node (n = 1). Research paper thumbnail of A novel order reduction method for nonlinear dynamical system under external periodic excitations

Using the modal transformation, a large nonlinear dynamical system is split into a ‘master’ subsystem, a ‘slave’ subsystem, and a ‘negligible’ subsystem. Accordingly, a novel order reduction method (Method I) is developed to construct a low order subsystem by neglecting the ‘negligible’ subsystem and slaving the ‘slave’ subsystem into the ‘master’ subsystem using the extended AIM. As a comparison, Method II accounting for the effects of both ‘slave’ subsystem and the ‘negligible’ subsystem is also applied to obtain the reduced order subsystem. Then, a typical 5-degree-of-freedom nonlinear dynamical system is given to compare the accuracy and efficiency of the traditional Galerkin truncation (ignoring the contributions of the slave and negligible subsystems), Method I and Method II. It is shown that Method I gives a considerable increase in accuracy for little computational cost in comparison with the standard Galerkin method, and produces almost the same accuracy as Method II. Finally, a 3-degree-of-freedom nonlinear dynamical system is analyzed by using the analytic method for showing predominance and convenience of Method I to obtain the analytically reduced order system.
