To offer alternative management strategies to insecticides, several studies have identified the semiochemicals that are involved in the pepper weevil’s aggregation and mating behavior; however, there is no information on its perireceptor molecular mechanism, to date. In this study, bioinformatics tools were used to functionally annotate and characterize the A. eugenii head transcriptome and their probable coding proteins. We identified twenty-two transcripts belonging to families related to chemosensory processes, seventeen corresponding to odorant-binding proteins (OBP), and six to chemosensory proteins (CSP). All results matched with closely related Coleoptera: Curculionidae homologous proteins. Likewise, twelve OBP and three CSP transcripts were experimentally characterized by RT-PCR in different female and male tissues. The results by sex and tissue display the different expression pattern...

In a non-choice test, V. destructor adult females were introduced into a comb containing either queen or worker brood cells; 0.62 and 18.28% of the mites entered the queen and worker brood cells, respectively. Only 1 of the 11 mites that entered queen brood cells oviposited, laying a single egg. In another test, brood cells were combined in the same comb in a 1:25:3 queen:worker:drone ratio. The percentages of egg-laying mites in queen, worker, and drone brood cells were 16.66, 61.86, and 79.06%, respectively. When queen, worker, and drone brood cells were combined in equal proportions (33.3:33.3:33.3), percent infestation was significantly different among queen (3.25%), worker (49.12%), and drone (90.07%) brood. Multiple infestation was found in drone brood cells but not in others. Also, mites were inoculated into sealed q...

The evaluation was carried out with the release and recapture of Anthonomus eugenii adults at different distances from the four cardinal points in separate trials. The volatility of the synthetic mixture and aggregation pheromone was determined by gas chromatographic analysis of the volatiles captured by dynamic headspace. The traps with synthetic mixture and essential oil captured insects at 10 m, while the aggregation pheromone trapped up to 60 m. The combination of synthetic mixture or essential oil with the aggregation pheromone did not increase the number of recaptures compared to the single pheromone. The synthetic mixture together with geranic acid recaptured adults up to 15 m, although they were not significantly different from the control. The exclusion of geranic acid from the aggregation pheromo...

Spotted-wing drosophila, Drosophila suzukii (Matsumura), is regulated in Mexico because of economic damage it causes to crops. Demands for export products in national and international markets are increasing, and compliance with mitigation measures in the field is required to minimize use of insecticide. It is necessary to develop environmentally friendly alternatives, such as the sterile insect technique, that also might be part of an integrated pest management (IPM) program. Effects of irradiation with Cobalt-60 on quality parameters (emergence rate, longevity, and flight ability) and fertility of D. suzukii adults were evaluated. Adults emerged from 5-day-old irradiated pupae reared on artificial diet. Doses of radiation evaluated were 60, 70, 80, 90, 180, and 200 Gy. No significant statistical differences between irradiated and non-irradiated flies were observed in quality parameters, emergence rate of 53.667–74% (F 8,12 = 1.55, P = 0.2433), longevity of 51.53–40.22 hours (F8,12 = 0.24, P = 0.9544), and flight percentage 36–54.30% (F6,14 = 2.05, P = 0.1258). One hundred percent of sterility induction was determined at 180 and 200 Gy, and fertility of D. suzukii females pre-confined with irradiated males and post-confined with fertile males gradually recovered. Data suggested the use of the sterile insect technique is promising as a component of an IPM program to control D. suzukii.

The host volatile compounds could be used for this purpose. In this research the attraction of A. eugenii adults towards an individual compound and 12 mixtures of synthetic volatile compounds (identified previously in Capsicum spp.) was evaluated, the effect of three isomers [(Z)-b-ocimene, (E)b-ocimene, and D-limonene] was determined in the weevil response, and the best mixture of host volatiles was evaluated, as well as their combination with the A. eugenii aggregation pheromone and the pheromone alone. The insect's response was evaluated through pairwise choice experiments, carried out in a Y-type olfactometer, with two treatments. The proportions of insects respondig to each treatment were compared with the binomial test. Results showed that the mixture of (Z)-b-ocimene, D-limonene and 2-isobutyl-3methoxypyrazine attracted (p£0.01) A. eugenii adults of both sexes. The (Z)-3-hexenyl acetate and the terpinolene, added individually to the prior combination, increased the response of females (p£0.05) and males (p£0.01), respectively. The inclusion of (E)-b-ocimene as principal compound, in the same mixture, increased the response by males (p£0.05) and the exclusion of D-limonene did not decrease the effectiveness of the remaining combination. In the final experiments, males had a greater response to the mixture of host volatile compounds (p£0.05) and to its combination with the A. eugenii aggregation pheromone (p£0.001), than to the pheromone by itself; in contrast, females did not show a differentiation in their choice. The research allowed selecting pepper volatile compounds with potential to attract A. eugenii adults.

The avocado stem weevil Copturus aguacatae is an important pest in avocado plantations. Its presence hinders the production and marketing of avocado in Mexico, the largest avocado producer worldwide. Biological control through pheromone synthesis, a strategy favored over chemical control in crops, is currently limited by difficult field identification of this species. Materials and methods. Using DNA barcoding, we examine the patterns of genetic variation of C. aguacatae in avocado trees in Mexico to help facilitate its identification and biological control. Results. We show that there is one single species of avocado stem weevil throughout the sampled sites in Mexico. Overall, haplotype diversity is high, with Oaxaca forming one distinct group and all other sampled populations are admixed irrespective of geographic origin. Conclusion. The results suggest that high gene flow is maintained in this species and that a global strategy for biocontrol can be designed and implemented throughout the sampled range.

The validation tests were conduced with seven previously described compounds in Capsicum´s volatiles (3-hexanone, 1-bromohexane, 2-ethyl-1-hexanol, 6-methyl-2-hepten-4-ol, p-xylene, D-limonene y 2-ethyl hexyl éster). The method exhibited good selectivity and linearity remained within the 2-120 mg mL-1 range with correlation coefficients greater than 0.99 for each evaluated compound. The relative standard deviation of factor responses was less than 2%. The validation results show that the method is efficient for extracting volatile compounds with recoveries between 87-98% with R.S.D. less than 10% which is indicative of good accuracy. The standard uncertainty associated with each measurement ranged from 1.17 to1.36 µg: the lowest related to 3-hexanone and the highest to 1-bromohexane. www.relaquim.com
