Nodal peripheral T-cell lymphomas are a rare group of neoplasms derived from post-thymic and acti... more Nodal peripheral T-cell lymphomas are a rare group of neoplasms derived from post-thymic and activated T lymphocytes. A review of scientific articles listed in PubMed, Lilacs, and the Cochrane Library databases was performed using the term "peripheral T-cell lymphomas". According to the World Health Organization classification of hematopoietic tissue tumors, this group of neoplasms consists of peripheral T-cell lymphoma not otherwise specified (PTCL-NOS), angioimmunoblastic T-cell lymphoma (AITL), anaplastic large cell lymphoma-anaplastic lymphoma kinase positive (ALCL-ALK(+)), and a provisional entity called anaplastic large cell lymphoma-anaplastic lymphoma kinase negative (ALCL-ALK(-)). Because the treatment and prognoses of these neoplasms involve different principles, it is essential to distinguish each one by its clinical, immunophenotypic, genetic, and molecular features. Except for anaplastic large cell lymphoma-anaplastic lymphoma kinase positive, which has no adverse international prognostic index, the prognosis of nodal peripheral T-cell lymphomas is worse than that of aggressive B-cell lymphomas. Chemotherapy based on anthracyclines provides poor outcomes because these neoplasms frequently have multidrug-resistant phenotypes. Based on this, the current tendency is to use intensified cyclophosphamide, doxorubicin, vincristine, prednisolone (CHOP) regimens with the addition of new drugs, and autologous hematopoietic stem cell transplantation. This paper describes the clinical features and diagnostic methods, and proposes a therapeutic algorithm for nodal peripheral T-cell lymphoma patients.

A utilização de terapias curativas muitas vezes é impossibilitada em virtude das comorbidades apresentadas por esta população ao diagnóstico. Nós adotamos um protocolo alternativo de quimioterapia oral combinando um inibidor da topoisomerase II e dexametasona. Apresentamos os resultados parciais com este protocolo em três pacientes portadores de LDGCB com idade superior a 80 anos e comorbidades severas. Todos alcançaram remissão completa com baixa toxicidade. Esses resultados demonstram que protocolos curativos alternativos devem ser testados em pacientes idosos portadores LDGCB a despeito da presença de comorbidades severas.

Immunosuppressive drugs have been experimentally used to obtain a state of immunodeficiency in mice. This investigation aimed to quantify the circulating T and B cells of mice treated with the immunosuppressive agents dexamethasone (Dx), cyclosporine (CsA) and cyclophosphamide (CY), as well as to observe the behaviour of lymphocytic populations in the spleen of these animals. Blood samples were collected for counting the total peripheral blood leukocytes and T and B lymphocytes using flow cytometry. Total leukocytes of mice treated with the three drugs during all study showed a significant decrease when compared to the results of the control group. The proportion of B and T lymphocytes from the treated animals also decreased significantly. This study sought to evaluate the contamination level of absorbent paper points used routinely in... more This study sought to evaluate the contamination level of absorbent paper points used routinely in dental clinical practice. For this study, 60 absorbent paper points were collected and separated into 3 groups: 20 paper points from sealed commercial packages (Group 1), 20 paper points from open commercial packages in use for 30 days (Group 2), and 20 paper points from a sealed commercial package that were manipulated by the operator (Group 3). Evaluation criteria was the presence or absence of turbidity in the brain heart infusion (BHI) broth used as the culture medium. The results (Kruskal-Wallis test; significance level = 5%) demonstrated bacterial growth in most of the samples for all groups, with a statistically significant difference in Group 3 compared to Groups 1 and 2. Intracellular antigen investigations by means of flow cytometry are essential for immune phenotyp... more Intracellular antigen investigations by means of flow cytometry are essential for immune phenotypical studies of oncohematological diseases. The use of Saponin for cellular permeabilization has proved to be cost–effective. Due to its detergent properties, Saponin permeabilizes the cytoplasmatic membrane without harming it or altering membrane antigen expressions, thus allowing simultaneous detection of intracellular and surface antigens. The objective was to analyze the efficiency of the cell permeabilization technique using Saponin as a permeabilizing agent. Thirty–six samples of peripheral blood and bone marrow underwent an immune phenotypical study at the Immune Pathology Laboratory of the Hospital das Clínicas of the University of São Paulo. Direct immunofluorescence was accomplished using the CellQuest program (Becton Dickinson, San Jose, CA) of the FACSCalibur flow cytometer (Becton Dickinson, San Jose, CA) to acquire and analyze the data. Excessive cell loss was observed when using rinsing buffers stored for more than seven days at temperatures ranging from 2ºC to 8ºC. Minimal cell loss was found when rinsing buffers were used immediately after preparation or frozen at the time of preparation and thawed just before use. In order to eliminate excessive cell loss, a systematic analysis of the technique stages was carried out, allowing us to restructure the technique to our purposes and its use in our laboratory. In a country with scarce resources, a viable, faster, more cost–effective technique (35% reduction in cost) aimed at investigating intracellular antigens by flow cytometry has been standardized.
