Research paper thumbnail of Inhibitory Susceptibility of Synthetic Selenium Nanoparticles and some Conjugate Nutritional Compounds in Inhibition of some Bacterial Isolates Causing Food Poisoning

Tikrit Journal for Agricultural Sciences, Dec 30, 2023

The results showed that the conjugations of zinc, vitamin D3 and cysteine with se-NPs were more effective in bacterial inhibition compared to inhibition of Se-NPs alone. The MIC of Se-NPs alone or in combination with Zn, D3 or Cysteine against both species of bacteria was appeared at 1% and above. The inhibitory activity increased against the bacterial species E.coli and S. aureus as the concentrations used in the treatments were increased, while the diameter of the inhibition zone was 25% against the two types of bacteria E.coli and Staph. aureus was the highest effect when treated with Se Nps + D3, which was at 30 mm, and 22 mm, respectively.

Research paper thumbnail of Molecular Evaluating the Therapeutic Application of Anticancer L-glutaminase Enzyme on Genetic Mutation Induced Rats

Mağallaẗ Tikrīt li-l-ʻulūm al-zirāʻaẗ̈, Dec 31, 2021

The Genomic DNA had been extracted from the liver tissue of the experimental rat groups following... more The Genomic DNA had been extracted from the liver tissue of the experimental rat groups following 15 weeks of treatment with partial purified L-glutaminase enzyme from E.coli in relation with ethylamine. The DNA samples of all the six treatment groups were amplified by PCR using three different primers specific for determining the presence of P53, bax and G3pdh genes and detect the effect of Ethylenimine and L-glutaminase on the animals at the molecular level. The agarose gel electrophoresis technique used to analyze the PCR amplification products. The result revealed the presence of P53 gene in all treatment groups except for diseased group (T2). This finding demonstrates the mutagenic effect of Ethylenimine that lead to mutation at P53 gene sequence, and therapeutic beneficial of L-glutaminase. Also, there is no PCR amplification product which represent Bax gene sequence for T2 and T5 groups which were administered doses of Ethylenimine, indicating that low doses of L-glutaminase failed to prevent the mutagenic effect of Ethylenimine. While, the G3pd genes is presented only in T2 and T5. Finally, analysis the DNA sequences of the PCR amplified products extracted from liver samples of T2 group treated with Ethylenimine was carried out, then the results were compared with NCBI. The expected mutations were found at thirteen locals and there were only three mutated sequences with the DNA of liver samples from T5 group, while the DNA of animals in group (T6) were showed no mutated regions. The results of this novel study make clear the therapeutic effect of L-glutaminase and how suppress the mutagenic and carcinogenic effect of ethylenimine on P53, Bax, and G3pd genes, and its effect was dose dependent..

Due to the widespread use of flavonoids in the treatment of many disease, The aim of this study was to see if Quercetin and Hesperidin could help reduce the effects of Acetamiprid on biochemical parameters which include Malondialdehyde, Aspartate aminotransferase, Alanine aminotransferase, Creatinine, and Urea, as well as antioxidants like Superoxide dismutase, Glutathion-S-transferase, Catalase and expression level of MAPK8 and NFKB genes. A total of 48 male rats were used in the investigation, with each group consisting of six animals that were dosed for 30 days. The groups were divided as follows: Control group (C) given drinking water only, (T1) given orally Acetamiprid in the dose (40mg/kg/B.W),(T2) given orally Quercetin in dose (20mg/kg/B.W),(T3) given orally Hesperidin in the dose (25mg/kg/B.W), (T4) given Quercetin in the dose(20mg/kg/B.W) then Hesperidin in the dose (25mg/kg/B.W)Concurrently,(T5) given Acetamiprid e in the dose (40mg/kg/B.W) then given Quercetin in the dose(20mg/kg/B.W) Concurrently, (T6) given Acetamiprid in the dose (40mg/kg/B.W) then given Hesperidin in the dose(25mg/kg/B.W) Concurrently, and(T7) given Acetamiprid in dose (40mg/kg/B.W) then given Quercetin in the dose(20mg/kg/B.W) then given Hesperidin in the dose(25mg/kg/B.W) Concurrently. The results clarified a significant increase (P<0.05) in Malondialdehyde, Urea, Creatinine, liver enzymes(AST,ALT) and gene expression of MAPK8 and NFKB genes, and a significant decrease in the level of enzymatic antioxidants (SOD, GST, CAT) in (T1) compared to (C) and other groups. On the other hand, results showed that giving animals Quercetin and Hesperidin separately or both simultaneously with the pesticide Acetamiprid in treatments (T5, T6, T7)caused of a significant improvement in the studied parameters compared to (T1) that was dosed with the pesticide alone. It was sometimes close to the control group, especially in treatment (T7), which gave the best results due to the synergistic effect of Quercetin and Hesperidin in reducing the oxidative effects caused by the pesticide. Concluded of this study that administration by Quercetin and Hesperidin alone or both together and simultaneously with the pesticide has a positive effectiveness and efficiency in reducing the oxidative damage resulting from the toxicity caused by the pesticide in the biochemical parameters, antioxidants and the level of gene expression in male albino rats.
