In previous studies on DNA repair during myogenesis, comparisons made of repair in post-replicati... more In previous studies on DNA repair during myogenesis, comparisons made of repair in post-replication myoblasts and in myotubes led to the conclusion that the capacity to repair damage in DNA decreased during myoblast differentiation. Using unscheduled DNA synthesis in response to UV-induced damage as an indicator of DNA repair in a myogenic line of rat skeletal muscle, it is demonstrated that nuclei in myotubes possess identical repair capacity as that in proliferating myoblasts. Furthermore, a brief increase in DNA repair capacity was observed to immediately follow the cessation of replicative DNA synthesis. This transient increase in repair capacity is consistent with the data of earlier reports and explains the previous but inappropriate conclusion that repair diminishes during myogenic differentiation. This transient increase in the capacity to repair DNA was not observed in a developmentally defective, non-differentiating line of similar myogenic origin.

Patients with a history of allergic reactions to local anesthetic drugs present a difficult probl... more Patients with a history of allergic reactions to local anesthetic drugs present a difficult problem during evaluation for cardiac catheterization procedures. In the worst cases the procedure may be deferred, or even performed without local anesthesia. In the vast majority of patients an acceptable agent for safe local anesthetic use can be found. Skin testing beginning with very dilute solutions of preservativefree local anesthetic agent may be administered easily. This report describes the practical aspects of skin testing and local anesthetic agent selection in patients with a history of allergic reaction to local anesthetic drugs.

To study the cardiac effects of progressive hypoxemia, we measured the left ventricular end-systo... more To study the cardiac effects of progressive hypoxemia, we measured the left ventricular end-systolic pressure-volume relation (ESPVR), myocardial oxygen consumption (MVOj), and myocardial oxygen delivery (MQOi) in eight thoracotomized dogs anesthetized with fentanyl and droperidol. We specifically looked for evidence of oxygen supply limitation of MVOj and depressed contractility (altered ESPVR) during stepwise decreases in inspired oxygen fraction. We hypothesized that the reported relation between MVOj and left ventricular pressure-volume area (PVA) may hold when inadequate MQOj determines MVOj, which then may limit PVA, manifested partly as a change in the ESPVR. Initially, as arterial oxygen saturation was decreased from 95 ±3 % to 64 ±14%, coronary blood Bow increased so that MQo? was maintained with no change hi myocardial extraction ratio (ER,, = MV(VMQOi).
