The aim of the study was to assess the degree of liver damage in a rabbit perfused with histidine-tryptophan-ketoglutarate (HTK [Custodiol]) solution with and without the presence of prolactin (PRL) based on biochemical studies in perfundate and ultrastructural analysis of hepatocytes. Materials and Methods. The experiment was carried out on rabbits. Liver ischemia was used in the study, based on Pringle's maneuver. About 70% of the rabbit liver lobes were perfused with HTK with or without the addition of PRL (2.5mg/g liver/h) under ischemic conditions for 2 hours. In the perfundate, the activity of enzymes alanine aminotransferase (ALT), aspartate aminotransferase (AST), lactate dehydrogenase (LDH), g-glutamyl transpeptidase (GGT), and lactate concentration were determined. Liver biopsies were collected for histopathologic evaluation under an electron microscope. Results. The addition of PRL to the HTK significantly reduced the leakage of enzymes from the liver to perfundate compared with the control group without PRL. The activity of ALT, AST, LDH, and GGT in the perfundates obtained after 2-hour perfusion with HTK-PRL solution was lower when compared with activity of the same parameters determined in perfundates with liver perfused with HTK without PRL. The area under the curve (AUC 0-2h) calculated for GGT, LDH, and lactates was significantly higher after perfusion with the HTK than with HTK with the addition of PRL. In the study group, bile was secreted throughout the whole experiment. The morphological confirmation of these results was obtained by means of transmission microscopy. Conclusion. PRL added to the preservation solution significantly inhibits the process of liver cell cytolysis, which may suggest its hepatoprotective effect.

Light microscopy and scanning electron microscopy (SEM) studies show the differences in structure... more Light microscopy and scanning electron microscopy (SEM) studies show the differences in structure, number and topography of terminal setae in the palpal organ in different stages of Hyalomma marginatum marginatum Koch, 1844. The palpal organ contains eight terminal setae in larvae and nymphs, and ten setae in adults. The length of these setae increases in the course of post-embryonic development. Other setae on palpal segments also undergo quantitative and morphological changes.

The study presents the external structure of Haller’s organ in all developmental stages of Dermac... more The study presents the external structure of Haller’s organ in all developmental stages of Dermacentor reticulatus (Fahr.). Our investigations, made by light and scanning electron microscopy, showed that the number of sensilla in the anterior pit varies in postembryonic stages of D. reticulatus. The larvae have five sensilla, whereas nymphs and adult ticks have six sensilla. The anterior pit sensilla show differences in topography, size and surface structure. The modifications of Haller’s organ are connected with life cycle, feeding, host specificity and habitat of D. reticulates.

Introdcution: Through interaction with receptors TNFR1 and TNFR2, TNF-α activates a signal path, ... more Introdcution: Through interaction with receptors TNFR1 and TNFR2, TNF-α activates a signal path, which exacerbates an inflammatory process, constituting an inseparable element of psoriasis. Aim: To evaluate changes in the expression of TNF-α, TNFR1, TNFR2 during the 4-year-long adalimumab therapy in psoriatic patients, searching for the correlation between molecular and clinical markers. In addition, the role of miRNAs was analysed. Material and methods: Whole blood and serum samples of psoriatic patients treated with adalimumab constituted material for the study. Changes in the expression of TNF-α and its receptors were evaluated with the use of the RTqPCR method and MALDI ToF mass spectroscopy, PASI, BSA, DAS28 indexes were used for the clinical analysis of the patients, while the role of miRNA molecules was determined basing on microrna.org database. Results: Different TNF-α expression patterns were determined in patients with observed resistance to the medicine. We found that there is a correlation between the molecular markers of an inflammatory process and the clinical indexes. The bioinformatic analysis indicates the potential role of miRNAs in the regulation of expression of the analysed genes. Changes in the profile of TNF-α during adalimumab therapy are significantly determined by the individual variability and susceptibility to the biological medicine or its loss. Conclusions: TNF-α seems to be a useful marker to evaluate the efficacy of therapy and occurring resistance to the medicine. A complex mechanism for the regulation of the analysed gene expression was underlined, which involved the potential role of miRNAs.

Natural origin substances with healing properties of potential use in the treatment of different ... more Natural origin substances with healing properties of potential use in the treatment of different diseases, particularly wound management, remain in the scientific and research interest for many years. The substance of natural origin with potentially wide use in medicine and pharmacy is snail mucus. The study aims to evaluate the effect of snail mucus extract on human fibroblasts and evaluate the biological and cytotoxic activity of this substance. In the present study, human NHDF fibroblast cells were exposed to snail slime at two different concentrations: 15 µL/mL and 70 µL/mL. Moreover, in this study, the assessment of the biological activity of cells was conducted using mitochondrial redox activity. At the same time, the analysis of the mucus sample was carried out throughout the infrared spectroscopy implementation. It has been shown that this substance contains compounds such as mucin, achacin, mitomycin, allantoin. The snail mucus affects the viability of fibroblasts-connective tissue cells found in the dermis, responsible for collagen and elastin production. The mechanism of positive action on human fibroblasts is not entirely known and requires further research.

The WHO forecasts an increase in the incidence of prostate cancer in the coming years. Patients may not respond to treatment, and may not tolerate the side effects of chemotherapy. Compounds of natural origin have long been used in the prevention and treatment of cancer. Flavonoids obtained from natural products, e.g., propolis, are compounds with proven antibacterial and antiviral efficacy which modulate the immune response and may be useful as adjuvants in chemotherapy. The main aim of the present study was to evaluate the cytotoxic and pro-apoptotic properties of selected flavonoids on prostate cancer cells of the LNCaP line. The compounds used in this study were CAPE, curcumin (CUR), and quercetin (QUE). Using NiTi alloys with shape memory for long-term medical implants requires modification of their... more Using NiTi alloys with shape memory for long-term medical implants requires modification of their surface due to the possible occurrence of corrosion. Hence, the surface of the staples used to join fractured bone within the craniofacial region was modified by applying a titanium oxy-nitrogen layer and a hydroxyapatite coating. Surface-modified clamps were tested in vivo using New Zealand white rabbits. After determining the mechanical characteristics of the bone and considering the initial state and surface modification, the diameter of the wire (used to make the clamps with the appropriate compression force) was selected. Implantation was performed on two groups of rabbits: experimental and control. In the experimental group, an intentionally induced bone fracture was treated in one tibia.
