Introduction. Microbiological researches of the role of denture base materials in violation of th... more Introduction. Microbiological researches of the role of denture base materials in violation of the oral cavity micro ecology were conducted. Aim. Therefore, the purpose of research is to study the effect of different polymeric materials on denture bases, on the intensity of planktonic growth of resident and transient representatives of oral cavity microflora in a periodic culture on the basis of registration of its optical density. Material and methods. 7 types of denture base materials and their influence on the planktonic growth of microorganisms have been studied. Results. The planktonic growth of the S. oralis strain was suppressed by all base materials: SYNMA (by 35.5%), Bre.Flex (by 27.5%) and Protacryl-M (by 25.7%), in the smallest – Nylon (by 5.4%), Villacryl (by 9.5%). Conclusions. Through experimental research, we have proved the direct involvement of basic resin materials in changes in the oral cavity micro ecology and the process of occurrence of prosthetic stomatitis.

When using removable dentures, in 84.26±1.19 % of cases patients have various changes in the mucous membrane of the prosthetic bed with pronounced changes in microbial ecology which are exacerbated by poor oral hygiene and removable dentures. Ozone therapy is a new and promising area of treatment of microbial lesions of the oral mucosa. It has antiseptic, anti-inflammatory, analgesic, immunomodulatory effects and promotes activation of intracellular metabolism, improvement of rheological properties and microcirculation of blood; it also strengthens reparative ability of the injured tissues. In present work we report our studies of the interaction of 1-phenyl-1H-pyrrole-2,5-dione derivat... more In present work we report our studies of the interaction of 1-phenyl-1H-pyrrole-2,5-dione derivatives (N-arylmaleimides) and 3-bromodihydrofuran-2(3H)-one (α-bromo-γ-butyrolactone) as possible [C2]2+ synthons with 1,2,4-triazole-3(5)-thiole targeting on synthesis of novel 5-substituted thiazolo[3,2-b][1,2,4]triazole-6(5H)-ones. According to obtained results was establish that in above-mentioned interactions (conditions: convenient heating, range of solvents/reaction time) the thiol-ene click (for N-arylmaleimides) and SN (for α-bromo-γ-butyrolactone) processes take place, but not [2+3]-cyclocondensation reaction. The structure of compounds was studied using 1H, 13C NMR spectroscopy, LC-MS spectrometry, and X-ray analysis. The prescreening of antimicrobial activity for synthesized compounds was performed against Gram-positive and Gram-negative bacteria, as well as yeasts.

The aim of the work is to conduct a comparative analysis of the biofilm formation by representati... more The aim of the work is to conduct a comparative analysis of the biofilm formation by representatives of the oral microflora on the surfaces of basic materials. Materials and methods. The process of biofilm formation was examined on 7 types of basic plastic samples: Polyan, Breflex, Nylon, Protakryl, Vinakryl, Biocryl, which were used for the manufacture of removable prosthetic basis constructions, and SYNMA, which was used for comparison. Biofilm formation was analyzed by the method Y. Zhang (2017) with minor modifications. The test sample was placed in a test tube with 2.0 ml of nutrient broth Brain Heart Infusion to model the biofilm growth of microorganisms (HiMedia Laboratories Pvt. Ltd., India) supplemented with 1 % glucose, pre-inoculated with test strains at a final concentration of 1 × 104 CFU/ml. Фармакофорна модель для скринінгу протистафілококової активності серед тіазолідинон-споріднених с... more Фармакофорна модель для скринінгу протистафілококової активності серед тіазолідинон-споріднених структур Мета. Розробити фармакофорну модель, придатну для скринінгу протистафілококової активності серед похідних тіазолідинону, тіопірано[2,3-d]тіазолу, тіазоло[4,5-b]піридину. Результати та їх обговорення. Найкраща фармакофорна модель у серії розроблених має планарну структуру і складається з ароматичного кільця (або циклу з π-зв'язками), гідрофобної області, проєкції донора водневого зв'язку та двох проєкцій акцептора водневого зв'язку. Її точність класифікації становить 72,4 %. Лінія діаметра моделі утворена проєкцією донора та проєкцією акцептора водневого зв'язку, а її довжина дорівнює 8,05 Å. Під час аналізу узгоджених з фармакофорною моделлю конформацій активних сполук виявлено два різні способи просторового розміщення активних молекул, за яких повною мірою виконуються умови фармакофорної моделі. Експериментальна частина. Протистафілококову активність визначали методом дифузії в агар щодо резистентного до метициліну штаму Staphylococcus aureus (MRSA) й оцінювали шляхом вимірювання діаметра зони інгібування мікробного зростання. Планшети інкубували протягом 24 год за 37 °С. Сполуки з діаметром затримки розмноження мікроорганізмів понад 7,5 мм вважали активними. Комп'ютерне оброблення результатів мікробіологічного експерименту та моделювання ймовірного фармакофора виконували в програмному середовищі MOE версії 2007.09. Геометрію сполук оптимізували методом молекулярної механіки з використанням силового поля MMFF94x. Точність класифікації використовували як основний критерій оцінювання якості фармакофорної моделі. Висновки. Розроблену фармакофорну модель можна використовувати для віртуального скринінгу протистафілококової активності споріднених із навчальною вибіркою сполук. Під час застосування цієї моделі до бази даних протестованих нами сполук з'ясували, що фактор збагачення складає EF = 2,05.

Methods. Diffusion in agar; serial dilutions in agar. Clinical isolates of microorganisms: methicillin-sensitive strain of Staphylococcus aureus (MSSA), methicillin-resistant strain of Staphylococcus aureus (MRSA), methicillinresistant strain of Staphylococcus haemolyticus (MRSH), Escherichia coli; Pseudomonas aeruginosa, ESβL + Klebsiella pneumonia, Candida albicans, Candida tropicalis. Results. Screening of antimicrobial activity of 13 new2-thioxo-4-thiazolidinone derivatives was carried out. The methicillin-resistant strain of Staphylococcus aureus (MRSA) was the most sensitive to the tested compounds. A number of derivatives exhibit synergism in combination with amoxicillin against the ESβL+ Klebsiella pneumonia strain. The structure-antimicrobial activity relationshipis was analyzed in detail. An increase in the number of removable denture constructions being produced leads to an increase ... more An increase in the number of removable denture constructions being produced leads to an increase in the number of complications, which, in turn, reduces the timing of the effective use of orthopedic structures. At present, the problem of treating prosthetic stomatitis and their prevention has not been resolved. Considering the current socio-economic situation, orthopedic treatment of the patients with removable dentures will be relevant for some more time. Microbiological studies of the ability of basic materials to adhere different types of microorganisms onto their surfaces were performed. There were used 7 different types of resins manufactured according to different technologies. Having performed a series of microbiological studies on the adhesive ability of basic materials, the following results were obtained. The initial acid was obtained by an effective one-pot method using the reaction of thiourea with maleic anhydride in hydrochloric acid. The maximum anti-staphylococcal activity was exhibited by N-[5-(R-benzyl)thiazol-2-yl]-2-(2,4-dioxothiazolidin-5-yl)acetamides.
