In Brazil, about 36,000 new cases of CRC are estimated among men and women for 2018-2019. With advances in treatment, early CRC is being "reclassified" from a deadly disease to an illness that has great chance of cure. However, so far, the survivors who have completed treatment and are cancer-free, frequently suffer from late/longterm side effects, and some patients with metastasis have poor response to the therapies. In this context, the discovery of new therapeutic strategies is urgently needed. In this project we aimed to validate the hypothesis that 3-Bromopyruvate (3-BP), an alkylating agent, would be able to sensitize the human CRC cell lines to treatment with Vemurafenib, a BRAF inhibitor. Therefore, human CRC cell lines (HT-29 and HCT-116) were challenged with 3-BP followed by the treatment with Vemurafenib. Both cell lines were more responsive to Vemurafenib after sensitization with 3-BP (IC50 values were 7-and 1.4-fold lower than with Vemurafenib alone, for HT-29 and HCT-116 cells, respectively). The findings presented here, highlight the potential of 3-BP in increasing the response of metastatic cancer cells to Vemurafenib.

Research paper thumbnail of Use of sequential infiltration synthesis to improve the pattern transfer of PS-b-PLA DSA (Conference Presentation)

Advances in Patterning Materials and Processes XXXIV, 2017

Directed self-assembly of block copolymers is considered one of the candidates to fulfill the req... more Directed self-assembly of block copolymers is considered one of the candidates to fulfill the requirements of the next technological nodes [1,2]. Polymer domains are aligned by using a chemical and/or a topographical pre-pattern in which preferential surfaces to one of the two blocks or neutral wetting areas are created. In particular, polystyrene-block-polymetylmethacrylate (PS-b-PMMA) has been extensively studied during the last years showing strong capabilities to define periodic nanostructures. However, the relatively low Flory-Huggins parameter and, the resulting low segregation strength of PS-b-PMMA systems, limit their achievable resolution to around 11 nm [3]. The application of block copolymer on sub-10 nm technologies requires the development of the new block copolymer generation known as high- block copolymers. Additionally, an important requirement for their integration on the lithography roadmap is the capability of selectively remove one of the two blocks. The etch contrast between the two domains is typically low due to their organic chemistry. In this sense, selective sequential infiltration synthesis by ALD into one of the blocks can be used in order to incorporate an inorganic material. The formed organic/inorganic blend sustains better the plasma etching to remove the non-infiltrated organic block. In this contribution, we show the use of high- polystyrene-b-polylactide acid (PS-b-PLA) lamellar block copolymer for line/space applications. PS-b-PLA has a larger Flory-Huggins parameter (=0.218 at room temperature[4]) compared with PS-b-PMMA, allowing the size reduction of the self-assembled domains. The method to control the orientation of the polymer domains is similar to the one typically used for PS-b-PMMA. Chemical contrast and the subsequent alignment of the polymer domains are achieved by the definition of a chemical pre-pattern on a random copolymer PS-r-PMMA (48% of PS) (figure 1). The polymer brush is grafted on the substrate and then locally modified by the combination of e-beam lithography and soft oxygen plasma. Afterwards, the PS-b-PLA block copolymer is spin-coated and thermally annealed on the chemically pre-patterned substrate. A chemical contrast is observed between the modified and unmodified stripes. While, the lamellar domains are oriented perpendicular to the substrate on unmodified areas, PLA domains are strongly attracted to the O2 modified surfaces inducing a parallel orientation to the substrate. Additionally, the wetting behavior of the polymer domains is also studied through the difference of surface free energy between the substrate and each polymer block. The energy estimated by the Young´s equation [Δγ =γSA -γSB= γAB·cos(ØAB)], where γSA and γSB are the interface tensions between homo-polymers A and B with the substrate, and ØAB is the contact angle between A and B homo-polymers which is obtained in de-wetting experiments. Finally, sequential infiltration synthesis is used to selectively infiltrate alumina (Al3O2) on PLA domains (figure 2). A selective infiltration is achieved because the precursor molecules react with the carbonyl (C=O) groups that are only present in the PLA block. After five cycles of SIS, the SIS modified PLA domains become more resistant to O2 plasma etching than PS enabling the PS etching without using other kind of hard-masks. The research leading to these results received funding from the European Union’s Seventh Framework Program FP7/2007-2013, under the project CoLiSA and the European Nanoelectronics Initiative Advisory Council under the project PLACYD (ENIAC-2013-2). It is based on comprehensive approach in studies conducted in three matrix searches from family care experiences. We have taken three axes to organize our reflections: a) conformation of family care in chronic situation, highlighting the multiple costs incurred to the family, which can exhaust the potential of care and establish or increase its vulnerability if it is not backed by networks support and sustenance; b) family rearrangements for the care, giving visibility to care cores in which many loved family members share the care, dynamic, plural and changeable way; c) self care modeling family care, pointing to the range of possibilities of the person taking care of diseased conditions supported by people close to them. We learn that the family takes care of itself in everyday life and in the illness experience, creating network...

Identification of the aeroallergens involved in allergenic sensitisation is important for diagnosis, treatment and prevention. To verify the molecular pattern of sensitisation to aeroallergens in patients with allergic respiratory diseases using microarray technique for specific IgE antibody detection. Cross-sectional study of 101 children with allergic rhinitis was followed in an outpatient clinic. All patients had positive skin prick tests (SPT) to at least one of the following antigens: Dermatophagoides pteronyssinus, Blomia tropicalis, Blattella germanica, Lolium multiflorum, and dog and cat epithelium. Serum specific IgE antibodies (sIgE) to mites, animal epithelia, fungi, cockroach and pollens components were determined by ImmunoCAP ISAC. sIgE to group 1 and 2 mite allergens showed higher positive rates: Der p 1 (74.2%), Der p 2 (73.3%), Der f 1 (74.2%), Der f 2 (72.3%). sIgE to animal epithelia were less frequent, Can f 1, Can f 2, Can f 3 in 4.9%, 2.9%, 1.9% respectively and Fel d 1, Fel d 2, Fel d 4 in 16.8%, 0.9% and 1.9%. respectively. Sensitisation to fungi and cockroach were rare, except for Bla g 7, to which 16.8% were positive. There was no significant recognition for tree pollens group. For grass, sIgE were detected to Cyn d 1 in 16.8%, Phl p 1 and Phl p 4 in 14.8% and 12.9%, respectively. Knowing that the pattern of allergic sensitisation varies according to environment and population, our results reinforce the need for local studies, using molecular-based diagnosis.

Addressed as a situational study and grounded in Beth’s life story about getting sick and dying of cancer at age 34, the study’s data collection process employed interviews, observation and medical record analysis. The construction of the analytic-synthetic box based on the chronology of Beth’s clinical progression, treatment phases and temporal perception of occurrences enabled us to point out a linear medical therapy protocol time identified by the diagnosis and treatment sequencing process. On the other hand, Beth’s experienced time was marked by simultaneous and non-linear events that generated suffering resulting from the disease. Such comprehension highlights the need for healthcare professionals to take into account the time experienced by the patient, thus providing an indispensable cancer ther...

Methods: this was a cross-sectional study with 99 children and adolescents diagnosed with asthma and/or allergic rhinitis, who had skin reactivity to at least one of the six aeroallergens tested. Clinical data were evaluated: skin prick tests with Bombyx mori in-house extract, and total and specific IgE analysis using ImmunoCAP® were performed. Results: the frequency of Bombyx mori specific IgE was found to be 52.5% and 60% using the skin prick test and ImmunoCAP®, respectively. An association between a positive skin test for Bombyx mori and the presence of allergic rhinitis, atopic dermatitis, and urticaria was observed, but the same was not true for asthma or allergic conjunctivitis. There was no relation with the severity of asthma or rhinitis symptoms. Conclusions: a high frequency of sensitization to Bombyx mori was observed in a selected population of patients with respiratory allergic diseases in the city of Curitiba, state of Paraná, Brazil. The extract prepared from the wings of this moth species is effective in demonstrating this sensitivity.
