Aim of the study We explored the relationships between the gingival fibroblasts and pro-B lymphoc... more Aim of the study We explored the relationships between the gingival fibroblasts and pro-B lymphocytes apoptosis in vitro because the current medical literature is lacking such data. Material and methods The aim of our study was represented by the effects of gingival fibroblasts on the apoptosis of Nur77 receptors-activated pro-lymphocytes in vitro, also in the presence of some retinoid derivatives. Gingival fibroblasts were obtained from male rats. Pro-B cells of murine type in cultures were used for actual experiments. The apoptosis of pro-B cells was induced using a combination of cytosporone B (50 µM) and cyclosporine A (1 µM). The apoptotic effects were revealed using the classical flow cytometry techniques. Results We clearly showed that pro-B lymphocyte apoptosis induced by the combination of cytosporone B (Nur77 agonist) and cyclosporine A in the presence of gingival fibroblasts could be slightly reduced by 13-cis retinoic acid. The inhibitory effects of 13-cis retinoic acid were powerful than those of tazarotenic acid. The effects of 13-cis retinoic acid and tazarotenic acid were evaluated toward the effects of the combination of cytosporone B and cyclosporine A. Conclusions The intrinsic pathways of retinoid derivatives remain to be established by further experiments

Background: The involvement of NR4A1 orphan receptors activation in pre-lymphocytes, pro-lymphocy... more Background: The involvement of NR4A1 orphan receptors activation in pre-lymphocytes, pro-lymphocytes and lymphocytes apoptosis is controversial, also when related to calcium fluxes. Aim: The goal of our study was represented by the characterization of NR4A1 activation-induced effects on pro-B cells apoptosis in the presence of ionomycin, a calcium overloading inducer. Methods: All cells batches were evaluated under contrast-phase microscopy, using an Eclipse TE-300 Nikon microscope with a 40x objective. The apoptotic cells number was expressed as a percentage of the total number of cells, taking into account their morphological appearance. Our studies aimed the effects of some endoplasmic reticulum stress inducers (thapsigargin, a Ca2+... more Our studies aimed the effects of some endoplasmic reticulum stress inducers (thapsigargin, a Ca2+-ATP-ase inhibitor; tunicamycin, a protein N-glycosylation inhibitor; brefeldin A, a protein transport inhibitor; paraquat, an enhancer of reactive oxygen species production; A23187, a Ca2+ ionophore), as well as some antioxidants (N-acetylcysteine; dithiothreitol, a disulfide bond formation inhibitor) on apoptosis of cultured rat mesenchymal stem cells. The analyze of obtained results evidenced that paraquat, a common and effective herbicide, induced the apoptosis of the isolated rat mesenchymal stem cells in a larger proportion as compared to other chemicals as follows: paraquat ] thapsigargin ] tunicamycin @A23187 ] brefeldin A. Dithiothreitol was effective as a reducer of mesenchymal stem cells apoptosis when was administered as co-treatment for paraquat for 24 h. In contrast, N-acetylcysteine, another potent antioxidant, had no protective effects against paraquat apoptotic effects.

This study complements the national screening program for cervical cancer, implemented in Romania... more This study complements the national screening program for cervical cancer, implemented in Romania. Inclusion criteria for this program of active detection of cervical cancer are women aged 25 to 64 years and exclusion criteria were other age groups and the diagnosis confirmed of cervical cancer. The testing was free and used the method of colored cervical smear Pap. In 2013, 23.680 women were tested, that means only 10.13% of the total female population eligible for screening. In 2014, the test group was 6337 (one smear required retesting), of which 5852 were negative results and 484 positive results. Squamous cell abnormalities were detected in 433 women (6.84% of total group who attended the screening). Cervical smears with atypical squamous lesions were present at the most women included in the screening, regardless of age group (67.43%). The screening detected 21 lesions HSIL (4.84%), 2 HSIL with suspicion of invasion (0.46%) and 5 lesions of squamous carcinoma (1.15%). The pres...
MATERIAL AND METHOD Pretreatment with apelin-13 (AP-13, 2 mg/kg, i.p.), sodium butyrate (BUT, 200... more MATERIAL AND METHOD Pretreatment with apelin-13 (AP-13, 2 mg/kg, i.p.), sodium butyrate (BUT, 200 mg/kg, s.c.) and N-acetyl-L-cysteine (NAC, 150 mg/kg, s.c.), all reduced the LPS-induced vascular leak measured as Evans blue extravasation, in rats lung tissue when compared to intranasal LPS (10 mg/100 mL) administered alone. RESULTS Although there is a significant difference either between AP-13 and BUT on one hand, and NAC and BUT on the other hand pretreatments, there is no significant difference between AP-13 and NAC pretreatments. Firstly, apelin-13 pretreatment might justify its effects through the modulation of endothelial layer functions. We recently demonstrated that AP-13 could diminish the endothelial dysfunction of pulmonary vein from both ovalbumin sensitized rats and rats with pulmonary hypertension. The inhibitors of AT1 receptor internalization we tested, that means nigericin, concanavalin A and monensin, did not significantly alter the contractions induced by Ang II in the rat aortic smooth muscle preparations. The same effects we obtained also when we used as agonists Ang I, Ang III and Ang IV. Thus, the contractile effects induced by angiotensin peptides administered in the rat aortic smooth muscle preparations are independent of their receptors internalization. Since -arrestins are mediating G-protein independent signaling via AT1 receptors their involvement in the contractile effects of angiotensin peptides must be further explored.

DESCRIPTIVE STUDY COUNTING CASES BETWEEN 2010 AND 2016 (Abstract): Infection with the parasite protozoa Toxoplasma gondii has a worldwide distribution. The mandatory intracellular parasite can infect humans, practically all warm-blooded animals, including mammals and birds. The study aim was to highlight descriptive epidemiology data on toxoplasmosis in patients admitted to a regional infectious disease hospital in North East Romania. Material and methods. We conducted a descriptive longitudinal study on a group of 160 patients admitted to the Iasi Clinical Hospital of Infectious Diseases with the diagnosis of toxoplasmosis, during 2010 and 2016. Results. Annual distribution of patients with toxoplasmosis had a maximum of frequency in 2010 (34 cases – 21.25%) and a minimum in 2016 (6 cases – 3.75%), with an increasing trend for the whole interval of study. Studies estimate that approximately 30% of all the CRC cases are a hereditary form of the disease with a potentially high impact on the quality of life (QoL), with a high risk of recurrence, and with bio–psycho–social functioning. The aim of the study was to assess the QoL of colorectal cancer patients with genetic risk by using the European Organisation for Research and Treatment of Cancer (EORTC) questionnaire version QLQ-CR29. Materials and methods: The cross-sectional, single-center study was performed on a group of 32 patients with genetic risk of colorectal cancer, who underwent preoperative chemoradiation and surgery. Trends in breast cancer incidence has been decreasing worldwide since several years, meanwhile sc... more Trends in breast cancer incidence has been decreasing worldwide since several years, meanwhile screening programs were properly implemented in countries with financial support for healthcare infrastructure and population awareness to choose prevention. The study aim was to evaluate the level of knowledge regarding risk factors and screening for breast cancer in general population from Romania. Material and methods: The study was conducted on a group of 470 individuals aged between 21 and 55, during 1st October and 31st November 2016. The study was based on fulfilling an online questionnaire of 20 items, sent by e-mail. Results: Participants chose “breastfeeding” as a protective factor (84.47%), “family history” as a risk factor (78.09%), lobular carcinoma in situ as precancerous lesion (20.00%), mammography as a screening method (94.04%), performed “once a year in 45-54 aged women” (90.21%). Drug delivery systems based on collagen represent a continuous challenge for chemistry and medici... more Drug delivery systems based on collagen represent a continuous challenge for chemistry and medicine. Our studies aimed the biochemical effects of biological collagen-coated matrices with rat mesenchymal stem cells on rat skin wound healing in vivo. When compared to control group, or to collagen matrices, the collagen matrices coated with mesenchymal stem cells induced a better repairing response, also from the point of view of macroscopic as well as microscopic examination. The obtained results proved evidence that the coating of collagen type I matrices with rat mesenchymal stem cells would accelerate the wound repairing processes at the level of experimentally damaged rat skin. The inflammatory processes are increased, the proliferative tissue is enhanced, and a large number of fibroblasts are active. A current area of interest is the determination of factors able to promote the transition from mu... more A current area of interest is the determination of factors able to promote the transition from muscle to adipose tissue. The current review has highlighted that treatment of myoblasts with fatty acids (especially oleic acid) and thiazolidindiones causes conversion to adipocytes. The molecular mechanisms mediating the adipogenic action of thiazolidinediones and fatty acids in myoblasts could involve peroxisome proliferators-activated receptor-gamma (PPARgamma and CCAAT-enhancer-binding protein C/EBP. The role of 1,25-D3 in adipogenesis is mediated at the molecular level through VDR-dependent inhibition of C/EBP and PPARgamma expression and a decrease in PPARgamma transactivation activity. Vitamin D supplementation increases muscle strength and ultimately reduces the incidence of falls. Additional research is needed to fully clarify the role of nutritional factors in adipogenesis.
The aim of our study was represented by the leptin involvement as a mediator of lipopolysaccharid... more The aim of our study was represented by the leptin involvement as a mediator of lipopolysaccharide (LPS)-increased pulmonary permeability in rats. Pretreatment with aminoguanidine (20 mg/kg i.p), 2-aminopurine (20 mg/kg i.p.) and subjecting to food deprivation (48 h, with water ad libitum) reduced the LPS-induced vascular leak, measured as Evans blue extravasation in lung tissue, by 41.70%, 21.39% and, respectively, 52.92%, when compared to LPS (10μg/kg, 100μl aliquots) administered alone. In addition, leptin alone induced Evans blue leakage that represents 74.38% from that induced by LPS. Since the inhibitory effects of 2-aminopurine are with 59.34% higher than that induced by aminoguanidine on leptin, we were able to conclude that leptin is at least partially mediating the LPS-increased pulmonary permeability in rats. The study aim was to highlight descriptive epidemiology data on toxoplasmosis in patients admitted to a regional infectious disease hospital in North East Romania. Material and methods. We conducted a descriptive longitudinal study on a group of 160 patients admitted to the Iasi Clinical Hospital of Infectious Diseases with the diagnosis of toxoplasmosis, during 2010 and 2016. MATERIAL AND METHODS Inclusion criteria for this program of active detection of cervical cancer are women aged 25 to 64 years and exclusion criteria were other age groups and the diagnosis confirmed of cervical cancer. The testing was free and used the method of colored cervical smear Pap. RESULTS In 2013, 23.680 women were tested, that means only 10.13% of the total female population eligible for screening. In 2014, the test group was 6337 (one smear required retesting), of which 5852 were negative results and 484 positive results. Squamous cell abnormalities were detected in 433 women (6.84% of total group who attended the screening). Cervical smears with atypical squamous lesions were present at the most women included in the screening, regardless of age group (67.43%). The study aim was to evaluate the level of knowledge regarding risk factors and screening for breast cancer in general population from Romania. Material and methods: The study was conducted on a group of 470 individuals aged between 21 and 55, during 1st October and 31st November 2016. The study was based on fulfilling an online questionnaire of 20 items, sent by e-mail. Our studies aimed the biochemical effects of biological collagen-coated matrices with rat mesenchymal stem cells on rat skin wound healing in vivo. When compared to control group, or to collagen matrices, the collagen matrices coated with mesenchymal stem cells induced a better repairing response, also from the point of view of macroscopic as well as microscopic examination. The obtained results proved evidence that the coating of collagen type I matrices with rat mesenchymal stem cells would accelerate the wound repairing processes at the level of experimentally damaged rat skin. The inflammatory processes are increased, the proliferative tissue is enhanced, and a large number of fibroblasts are active. A current area of interest is the determination of factors able to promote the transition from mu... more A current area of interest is the determination of factors able to promote the transition from muscle to adipose tissue. The current review has highlighted that treatment of myoblasts with fatty acids (especially oleic acid) and thiazolidindiones causes conversion to adipocytes. The molecular mechanisms mediating the adipogenic action of thiazolidinediones and fatty acids in myoblasts could involve peroxisome proliferators-activated receptor-gamma (PPARgamma and CCAAT-enhancer-binding protein C/EBP. The role of 1,25-D3 in adipogenesis is mediated at the molecular level through VDR-dependent inhibition of C/EBP and PPARgamma expression and a decrease in PPARgamma transactivation activity. Vitamin D supplementation increases muscle strength and ultimately reduces the incidence of falls. Additional research is needed to fully clarify the role of nutritional factors in adipogenesis.
The aim of our study was represented by the leptin involvement as a mediator of lipopolysaccharid... more The aim of our study was represented by the leptin involvement as a mediator of lipopolysaccharide (LPS)-increased pulmonary permeability in rats. Pretreatment with aminoguanidine (20 mg/kg i.p), 2-aminopurine (20 mg/kg i.p.) and subjecting to food deprivation (48 h, with water ad libitum) reduced the LPS-induced vascular leak, measured as Evans blue extravasation in lung tissue, by 41.70%, 21.39% and, respectively, 52.92%, when compared to LPS (10μg/kg, 100μl aliquots) administered alone. In addition, leptin alone induced Evans blue leakage that represents 74.38% from that induced by LPS. Since the inhibitory effects of 2-aminopurine are with 59.34% higher than that induced by aminoguanidine on leptin, we were able to conclude that leptin is at least partially mediating the LPS-increased pulmonary permeability in rats.
