Reporter virus neutralization test (RVNT) has been used as an alternative to the more laborious a... more Reporter virus neutralization test (RVNT) has been used as an alternative to the more laborious and time-demanding conventional PRNT assay for both DENV and ZIKV. However, few studies have investigated how these techniques would perform in epidemic areas with the circulation of multiple flavivirus. Here, we evaluate the performance of ZIKV and DENV Rluc RVNT and ZIKV mCh RVNT assays in comparison to the conventional PRNT assay against patient sera collected before and during ZIKV outbreak in Brazil. These samples were categorized into groups based on (1) acute and convalescent samples according to the time of disease, and (2) laboratorial diagnostic results (DENV and ZIKV RT-PCR and IgM-capture ELISA). Our results showed that DENV Rluc assay presented 100% and 78.3% sensitivity and specificity, respectively, with 93.3% accuracy, a similar performance to the traditional PRNT. Brazil has experienced an unprecedented epidemic of Zika virus (ZIKV), with ~30,000 cases reporte... more Brazil has experienced an unprecedented epidemic of Zika virus (ZIKV), with ~30,000 cases reported to date. ZIKV was first detected in Brazil in May 2015 and cases of microcephaly potentially associated with ZIKV infection were identified in November 2015. Using next generation sequencing we generated seven Brazilian ZIKV genomes, sampled from four self-limited cases, one blood donor, one fatal adult case, and one newborn with microcephaly and congenital malformations. Phylogenetic and molecular clock analyses show a single introduction of ZIKV into the Americas, estimated to have occurred between May-Dec 2013, more than 12 months prior to the detection of ZIKV in Brazil. The estimated date of origin coincides with an increase in air passengers to Brazil from ZIKV endemic areas, and with reported outbreaks in Pacific Islands. ZIKV genomes from Brazil are phylogenetically interspersed with those from other South American and Caribbean countries. Mapping mutations onto existing structural models revealed the context of viral amino acid changes present in the outbreak lineage; however no shared amino acid changes were found among the three currently available virus genomes from microcephaly cases. Municipality-level incidence data indicate that reports of suspected microcephaly in Brazil best correlate with ZIKV incidence around week 17 of pregnancy, although this does not demonstrate causation. Our genetic description and analysis of ZIKV isolates in Brazil provide a baseline for future studies of the evolution and molecular epidemiology in the Americas of this emerging virus

Until now there are no reports on the isolation of Sporothrix sp. in cats in the state of Pará. The present work aimed to isolate Sporothrix spp. from a cat with possible zoonotic involvement in the city of Belém, Pará. Through the active search for suspected cases of sporotrichosis, carried out by members of the Center for the Control of Zoonoses of Belém (CCZ, Belém) the suspicion of the disease in a feline was reported, with probable transmission to its owner. Clinical evaluation was performed, and biological material was collected from the animal for identifying the agent. There are several infectious diseases that affect equines and can cause large economic losses in ... more There are several infectious diseases that affect equines and can cause large economic losses in this sector and damage to animal welfare. In this species, abscessing lesions of a purulent character preferentially in the withers are strongly suggestive of brucellosis. However, few studies have been carried out on the disease in this species, making its identification and control difficult due to the lack of standardization in the interpretation of results. Therefore, samples of whole blood, serum and purulent secretion were collected from three horses. The animals came from the state of Pará. The diagnostic methods selected were the Rose-Bengal plate test (RBPT) and Polymerase Chain Reaction (PCR), for the negative molecular results for brucellosis, microbiological analysis were chosen. Although part of the samples were positive for brucellosis in serological tests, all of them showed negative results for PCR. Currently, the Milwaukee protocol presents healing results in human beings affected by the rabies... more Currently, the Milwaukee protocol presents healing results in human beings affected by the rabies virus. However, there are many points to clarify on the action of drugs and the immune mechanism involved in the evolution of the disease. One of the drugs used is biopterin, which is an important cofactor for nitric oxide, important for preventing vasospasm. Thus, we describe the effect of biopterin on some inflammatory factors in a rabies virus infection developed in an animal model. The immunological mediators studied in animals infected with rabies virus submitted to doses of sapropterin were Anti-RABV, IL-6, IL-2, IL-17a, INF-gamma and Anti-iNOS. It is suggested that the medication in the context of a RABV infection already installed, had the effect of modulating the inflammatory mechanisms mainly linked to the permeability of the blood-brain barrier and the migration of cytotoxic cells.

This herd is located especially in the Marajo Island. The aim of this study was to compare three different types of tuberculin skin tests used for the diagnosis of tuberculosis in buffaloes (Bubalus bubalis) and compare the results with infected and uninfected animals by tuberculosis. The results of this study allows to the following conclusions: The single cervical test in buffaloes is not recommended due to the low specificity, producing a large margin of error (sacrifice of false positive animals); the double intradermal tuberculin test has a low specificity and the inconvenience of applying twice tuberculin, therefore is necessary three visits in the farm, raising the costs. The comparative cervical testis is the best method, because it has high specificity and sensitivity, and the reactions of tuberculin in buffaloes...

Free-living Columbidae are considered the second largest reservoir of Chlamydia psittaci, transmi... more Free-living Columbidae are considered the second largest reservoir of Chlamydia psittaci, transmitting the agent to humans and other animals. The present study aimed to identify the presence of Chlamydia spp. in samples of lung and stool collected from free-living domestic pigeons (Columba livia domestica) captured at the Mangal das Garças Naturalistic Park in Belém, Pará, Brazil, using semi-nested polymerase chain reaction, and also correlate the clinical and post-mortem findings of animals positive for Chlamydia spp. Among the 45 animals analyzed, 10 (22.2%) were positive for Chlamydia spp.; the positive findings originated from 5 (50%) lung samples and 5 (50%) stool samples, with no overlap between animals and samples. None of the animals evaluated in this study showed clinical signs of chlamydiosis; rather, these were only found during necropsy of positive animals, mainly through pulmonary, hepatic, splenic, and intestinal changes. In this study, we evaluated the prenatal conditions of pregnant female squirrel monkeys (Saimiri collinsi) infected during different gestational thirds (GTs) and assessed all clinical aspects, diagnostic imaging, viremia and the immune response. In our study, 75% of the infected animals in the 1st GT group had significant clinical manifestations, such as miscarriage and prolonged viremia associated with a late immune response. Consequently, their neonates showed fetal neuropathology, such as cerebral hemorrhage, lissencephaly or malformations of the brain grooves, ventriculomegaly, and craniofacial malformations. Com o objetivo de avaliar a epidemiologia da raiva, procedimentos complementares ao diagnostico -... more Com o objetivo de avaliar a epidemiologia da raiva, procedimentos complementares ao diagnostico - caracterizacao antigenica e genetica - foram incluidos neste estudo para investigar o perfil epidemiologico da raiva animal na Amazonia brasileira, entre janeiro de 2000 e julho de 2009. Foi realizada uma revisao cuidadosa das informacoes de amostras do sistema nervoso central (SNC) recebidas e analisadas no Laboratorio de Raiva do Instituto Evandro Chagas. The complete genome was sequenced using the next-generation sequencing strategy. The full-length genome of DrERV QR09 is 8,256 nucleotides in length and showed high similarity with other DrERVs.

Anticorpos IH foram detectados para diferentes arbovírus, com reações monotípicas para os VMAY e VORO, dois importantes arbovírus associados a epidemias na Amazônia. Anticorpos IgM para o VORO e VMAY foram detectados em soros humanos, sugerindo infecção recente por esses arbovírus. Duas cepas do VDEN-3 foram isoladas de pacientes febris residentes em Novo Progresso e identificadas como genótipo III. Em termos gerais, os dados obtidos sugerem uma área propícia para a circulação e manutenção de arbovírus e uma população pouco imunizada. O objetivo foi avaliar a prevalência de anticorpos para arbovírus na área de influência do Projet... more O objetivo foi avaliar a prevalência de anticorpos para arbovírus na área de influência do Projeto Juruti antes da exploração mineral local. Foram examinados 1.597 soros humanos e 85 de animais silvestres. A pesquisa de anticorpos foi realizada pelo teste de inibição da hemaglutinação contra antígenos dos arbovírus mais prevalentes na Amazônia brasileira e IgM-ELISA para dengue e febre amarela. Soros humanos inibidores da hemaglutinação positivos para Mayaro e Oropouche foram também testados por IgM-ELISA. Anticorpos inibidores da hemaglutinação para alfavírus, orthobunyavirus e/ou flavivírus foram detectados, sendo 28,7% reações monotípicas. Infecções recentes para Oropouche (n = 23), Mayaro (n = 5), e dengue (n = 20) foram confirmadas por IgM-ELISA.
