ABSTRACT Bacteria are ubiquitous in the natural environment, even when nutrients are limiting and... more ABSTRACT Bacteria are ubiquitous in the natural environment, even when nutrients are limiting and they are in a physiological state of starvation-survival. How bacterial cells survive for extended periods of time when the nutrients and energy needed for growth and cell division are insufficient is examined. A method for determining carbon utilisation patterns in soil extracts is included. This determines the diverse range of carbon substrates metabolised by soil microorganisms when they are extracted from soil samples and incubated under conditions suitable for rapid metabolism. This practical also highlights the diversity of carbon substrate utilisation in environmental soil samples.

While this could be due to an increase in the number of female conceptuses surviving, several studies suggest that fewer males are conceived (8) or survive (9, 10) during maternal stress. Fukuda and coworkers (11) found that both maternal and paternal smoking decreased the sex ratio; they suggested that the stress produced by cigarette smoking may be responsible for the decreased sex ratio.

Immunization practices and knowledge of vaccine contraindications among pediatric residents were ... more Immunization practices and knowledge of vaccine contraindications among pediatric residents were assessed at five pediatric training facilities by surveying 227 pediatric residents. Residents administer vaccines even in the presence of an afebrile minor illness. They are less likely to vaccinate if a fever (<102F) is present. Only 57% of residents report administering vaccines at the 15-month well-child checks. Many residents had difficulty recognizing true and false contraindications, though third-year residents did better than first-year residents. Failure during residency to utilize 15-month well visits and ill visits in the presence of a fever and the lack of knowledge of true and false vaccine contraindications may be causes of missed opportunities to vaccinate among residents. Immunization practices resulting in missed opportunites to vaccinate seen during during residency may influence later immunization practices.

Using winter as the referent season, we observed differences in amplitude of seasonal change across the 2 sites with spring, summer, and fall values for MIN/ABQ parameter estimates of-0.32/-0.09,-0.31/-0.14,-0.18/-0.08 (p<.0001). In site-controlled models with terms for daylight and temperature, only temperature remained a significant predictor (p=.004); the stratified model for ABQ showed a retrograde estimate for light (0.02, p=0.31) which may be explained by ABQ's later peak A1c date of March 1, compared to December 31 for MIN. Discussion: The trends observed in this study reflect those published previously and indicate a strong seasonal element to A1c scores with latitudinal amplification. LOESS plots reveal two distinct "humps" starting at the solstice and ending early October where the slope of the line flattens for ABQ, and reverses for MIN from the equinoctial peak. ABQ's late A1c peak is also close to the spring equinox, suggesting that perhaps 12-hour days act as an independent signal above and beyond the greater trend of increasing or decreasing daylight.

The interactions between T cells and the innate immune system while combating sepsis are poorly understood. In this report, we observed that treatment with the potent, antiapoptotic cytokine interleukin-7 (IL-7) accelerated neutrophil recruitment and improved bacterial clearance. We first determined that T cells were necessary for the previously observed IL-7-mediated enhanced survival. Next, IL-7 increased Bcl-2 expression in T cells isolated from septic mice as early as 3 h following treatment. This treatment resulted in increased gamma interferon (IFN-γ) and IP-10 production within the septic peritoneum together with local and systemic increases of IL-17 in IL-7-treated mice. We further demonstrate that the increase in IL-17 was largely due to increased recruitment and production by γδ T cells, which express CXCR3. Consistent with increased IL-17 production, IL-7 treatment increased CXCL1/KC production, neutrophil recruitment, and bacterial clearance. Significantly, end-organ tissue injury was not significantly different between vehicle- and IL-7-treated mice. Collectively, these data illustrate that IL-7 can mediate the cross talk between Th1 and Th17 lymphocytes during sepsis such that neutrophil recruitment and bacterial clearance is improved while early tissue injury is not increased. All together, these observations may underlay novel potential therapeutic targets to improve the host immune response to sepsis.

There are numerous interacting biological, physical and chemical factors that affect survival of ... more There are numerous interacting biological, physical and chemical factors that affect survival of both indigenous and introduced bacteria in soil. Three important factors arc clay type, clay content and predation by soil protozoans. These factors are important when considering release of non-engineered and genetically engineered microorganisms (GEMS) into soil. In this review we examine the influence of clays and predatory activity of protozoans on bacteria1 survival. These aspects are related because protozoa may ingest clay particles as well as bacteria. In addition, clays may protect bacteria from predation by protozoans by increasing the number of protective microhabitats available to bacteria in soil.

Cytoplasmic proteins that regulate signal transduction or induce cellular transformation, includi... more Cytoplasmic proteins that regulate signal transduction or induce cellular transformation, including cytoplasmic protein-tyrosine kinases, p21w GTPase-activating protein (GAP), phospholipase Cy, and the v-crk oncoprotein, possess one or two copies of a conserved noncatalytic domain, Src homology region 2 (SH2). Here we provide direct evidence that SH2 domains can mediate the interactions of these diverse signaling proteins with a related set ofphosphotyrosine ligands, including the epidermal growth factor (EGF) receptor. In src-transformed cells GAP forms heteromeric complexes, notably with a highly tyrosine phosphorylated 62-kDa protein (p62). The stable association between GAP and p62 can be specifically reconstituted in vitro by using a bacterial polypeptide containing only the N-terminal GAP SH2 domain. The efficient phosphorylation of p62 by the v-Src or v-Fps tyrosine kinases depends, in turn, on their SH2 domains and correlates with their transforming activity. In lysates of EGF-stimulated cells, the N-terminal GAP SH2 domain binds to both the EGF receptor and p62. Fusion proteins containing GAP or v-Crk SH2 domains complex with similar phosphotyrosine proteins from src-transformed or EGF-stimulated cells but with different efficiencies. SH2 sequences, therefore, form autonomous domains that direct signaling proteins, such as GAP, to bind specific phosphotyrosine-containing polypeptides. By promoting the formation of these complexes, SH2 domains are ideally suited to regulate the activation of intracellular signaling pathways by growth factors.
