The selection of appropriate locations for the reintroduction of endangered plant species is an i... more The selection of appropriate locations for the reintroduction of endangered plant species is an important process, because it usually influences the success of the conservation. The aim of this study was to select the optimal substitute habitats for Aldrovanda vesiculosa, taking into account the influence of physical–chemical factors (light intensity, temperature, pH, concentration of dissolved forms of nitrogen and cyanobacterial toxin microcystin-LR) on the efficiency of plant growth. Water analysis and field observations of the habitats of six lakes in Eastern Poland typified as potential substitute habitats for aldrovanda were carried out. The results of the experiments showed that both the concentration and the form in which nitrogen compounds are present in the environment were the factors limiting the growth rate and condition of plants. The cultivation system with cover crops as living mu lches is growing in importance in the worldw... more The cultivation system with cover crops as living mu lches is growing in importance in the worldwide agriculture as it combines an economical effect with pro -ec logical and soil protecting activities. The pla nt species tested in the research were not frost resistant : buckwheat, millet, white mustard and berseem clover . A control were the plots without cover crops. The inte rcrops were sown in the first decade of August, in autumn a grown biomass was a living mulch, and after be ing damaged by frost in spring, the mulch was covering ground surface. Winter garlic bulbs were plant ed in grown biomass of cover plants, in the first de cade of September. The main aim of the study was to investigate the viability of pollen of a boreal relict, Salix la... more The main aim of the study was to investigate the viability of pollen of a boreal relict, Salix lapponum, an endangered species in Poland as well as to evaluate the possibility to introduce the plant into a tissue culture. Two approaches were taken to estimate pollen viability: staining pollen with dyes and in vitro germination assay. The study results showed high pollen viability of the species, with regard to both fresh pollen and pollen stored for 12 months. The effectiveness of pollen tube growth was also found to be largely dependent on glucose content in a medium and thermal conditions. The study results have provided necessary information on the most optimal combinations for pollen germination of boreal willow in artificial conditions. The research on tissue culture initiation of S. lapponum was also undertaken. T... more Aldrovanda vesiculosa is a carnivorous water plant which is endangered by extinction worldwide. The number of natural stands and populations has decreased; therefore, there is a need for its active protection. The best method would be an in vitro culture. One of the main problems is disinfection of the explants. Therefore, it was decided that we should treat the explants with nano-silver particles. The explants were shoot fragments which were disinfected with sodium hypochlorite and then placed in a liquid 1/5 MS medium, supplemented with silver nanoparticles (AgNPs) at a concentration of 5 mg·dm−3. It was observed that AgNPs reduced the number of contaminations but also led to necrosis of the shoots. The shoots, which undertook regeneration in presence of AgNPs, were smaller and did not form traps; however, after being moved to fresh media twice, they started to develop normal leaves. The effect of auxins: IAA, IBA and NAA in concentrations of 1.0, 2.5 and 5.0 mg·dm-3 on rooting of Hebe buchananii and Hebe canterburiensis ‘Prostrata ’ in vitro was examined. Shoots used in the experiment were excised from aseptically grown shoots on 1/2 MS media. Auxins used in the experiment were showed to have a positive effect on rooting of shoots of Hebe buchananii and Hebe canterburiensis ‘Prostrata ’ in vitro. The biggest number of rooted microshoots and the longest roots of Hebe buchananii formed in presence of IBA. On the media supplemented with IAA in concentrations of 2.5 and 5.0 mg·dm-3, IBA in concentration of 5 mg·dm-3 and NAA in concentration of 1 mg·dm-3 100 % of rooted shoots of Hebe canterburiensis ‘Prostrata ’ were obtained. The aim of the study was to estimate the effect of activated charcoal (AC);1, 2 or 4 g/L) or ascorbic acid (AA; 10, 20 or 30 mg/L) added to the half strength MS medium on the growth and o-dihydroxyphenols content in Paphiopedilum insigne in vitro. A positive effect of AC on the shoot and root formation has been found. The highest multiplication rate (5.6 shoots/explant) and rooting frequency were obtained on medium containing 2 g/L of AC. However, AC reduced the leaf number as compared to the control. The lowest content of o-dihydroxyphenols was marked in Paphiopedilum insigne leaves when the shoots were grown on medium with 10 mg/L AA, followed by AC at 1 or 2 g/L.

Hortiterapia może odbywać się w ogrodach przy szpitalach, domach opieki społecznej, domach spokojnej starości, ośrodkach dla osób uzależnionych, a także w terapeutycznych gospodarstwach o profilu rolnym i ogrodniczym. However, due to a low propagation rate, it is difficult to keep up with the demand. Micropropagation would allow to produce lots of plants for the market. The aim of the study was to determine the effect of the growth regulators addition to the media and explants orientation on multiplication of Mandevilla sanderi, an exotic, ornamental pot plant. The shoot tips were placed vertically or horizontally on the Murashige and Skoog medium supplemented with benzyladenine (BA) or isopentenyladenine (2iP), at concentrations of 1, 2.5 or 5 mg·dm–3 singly or in combination with thidiazuron (TDZ) at concentrations of 0.01, 0.025 or 0.05 mg·dm–3. Maximum multiplication rate was noted on the media supplemented with 2.5 mg·dm–3 2iP + 0.025 mg·dm–3 TDZ or 5 mg·dm–3 2iP, when explants were placed horizontally. So far, there is no available information on micropropaga- tion of this beautiful species. Determination of the optimal sterilization methods as well as types and concentrations of plant growth regulators as medium constituents is one of the most important factors of successful micropropagation. With the aim of optimization of in vitro initiation and multiplication of C. radicans, the effect of different methods of disin- fection and terms of explants isolation on contamination rate of cultures as well as the in- fluence of cytokinins on growth and branching of shoots was studied. The cytokinins used in the experiments were: benzyladenine (BA), isopentenyl adenine (2-iP) and kinetin (KIN). Therefore, the producers seek for more effective methods of its propagation. In the years 2011-2014 in the Felin Experimental Farm of the University of Life Sciences in Lublin the research was undertaken to estimate the crowns efficiency of four Polish border dahlia cultivars. The tubers were taken out from storage in early spring, then planted in trays filled with peat and placed on the tables in a glasshouse. The soft stem cuttings snipped off from the sprouts arising from tubers were used for experiments which aim was to evaluate the quality of four types of rooted soft cuttings. It was stated that the number of sprouts appearing in spring on a mother plant depended on a cultivar and a season. Tubers of 'Krynica' and 'Halinka' cul- tivars formed the most sprouts. Shoot tips obtained from aseptical tissue cultures were grown for 6 weeks on Murashige and Skoog medium containing BA in concentration of 1 mg ∙ dm -3 . It was found that induction of axillary shoots was the best on double-phase medium (addition of liquid medium after 4 weeks of shoot cultures in vitro). No significant differences were found in regeneration potential and elongation of shoots depending of the media solidified by studied agars or Gelrite.

Simple Summary Salix lapponum, a downy willow, is a boreal relict species, threatened with extinc... more Simple Summary Salix lapponum, a downy willow, is a boreal relict species, threatened with extinction in Poland. In order to save populations in their refugia on the southern limit of the specie’s range, some activities were undertaken to rebuild their resources. The in vitro propagation was chosen to produce new plants, as it allows obtaining many individuals in a relatively short time with no harm to natural populations. The collected shoot pieces were multiplicated on a special growing media, containing all the necessary macro- and micronutrients, with addition of plant growth regulators to make them form shoots and roots. The obtained plants were then planted into soil and acclimated to natural habitat conditions. On the basis of the conducted genetic analysis and flow cytometry, it was stated that the new plants were genetically unchanged in comparison to the mother plants.
